Genetic resistance to DEHP-induced transgenerational endocrine disruption

Di(2-ethylhexyl) phthalate (DEHP) interferes with sex hormones signaling pathways (SHP). C57BL/6J mice prenatally exposed to 300 mg/kg/day DEHP develop a testicular dysgenesis syndrome (TDS) at adulthood, but similarly-exposed FVB/N mice are not affected. Here we aim to understand the reasons behind this drastic difference that should depend on the genome of the strain. In both backgrounds, pregnant female mice received per os either DEHP or corn oil vehicle and the male filiations were examined. Computer-assisted sperm analysis showed a DEHP-induced decreased sperm count and velocities in C57BL/6J. Sperm RNA sequencing experiments resulted in the identification of the 62 most differentially expressed RNAs. These RNAs, mainly regulated by hormones, produced strain-specific transcriptional responses to prenatal exposure to DEHP; a pool of RNAs was increased in FVB, another pool of RNAs was decreased in C57BL/6J. In FVB/N, analysis of non-synonymous single nucleotide polymorphisms (SNP) impacting SHP identified rs387782768 and rs29315913 respectively associated with absence of the Forkhead Box A3 (Foxa3) RNA and increased expression of estrogen receptor 1 variant 4 (NM_ 001302533) RNA. Analysis of the role of SNPs modifying SHP binding sites in function of strain-specific responses to DEHP revealed a DEHP-resistance allele in FVB/N containing an additional FOXA1-3 binding site at rs30973633 and four DEHP-induced beta-defensins (Defb42, Defb30, Defb47 and Defb48). A DEHP-susceptibility allele in C57BL/6J contained five SNPs (rs28279710, rs32977910, rs46648903, rs46677594 and rs48287999) affecting SHP and six genes (Svs2, Svs3b, Svs4, Svs3a, Svs6 and Svs5) epigenetically silenced by DEHP. Finally, targeted experiments confirmed increased methylation in the Svs3ab promoter with decreased SEMG2 persisting across generations, providing a molecular explanation for the transgenerational sperm velocity decrease found in C57BL/6J after DEHP exposure. We conclude that the existence of SNP-dependent mechanisms in FVB/N inbred mice may confer resistance to transgenerational endocrine disruption.

Summary: In this study, a biocompatible hybrid nanosystem for drug delivery purposes was fabricated using the electrospraying technique, combining Ag2S nanoparticles and superparamagnetic iron oxide nanoparticles (SPION). The hybrid nanosystems showed good photoluminescent properties and relatively long decay times. Encapsulation and release capabilities were demonstrated using a model chemotherapeutic drug, and in vitro cytotoxicity assays showed promising results against tumor cell lines.

Predicting COVID-19 Severity Integrating RNA-Seq Data Using Machine Learning Techniques

Summary: A new intelligent pipeline was implemented to develop a specific gene signature that can detect the severity of patients suffering from COVID-19. The pipeline integrates RNA-Seq data from different databases using an artificial intelligence algorithm. CD93, RPS24, PSCA, and CD300E were identified as COVID-19 severity gene signatures, and a multi-class classifier was optimized to classify patients with an accuracy of 97.5%.

In Vitro Efficacy of Extracts and Isolated Bioactive Compounds from Ascomycota Fungi in the Treatment of Colorectal Cancer: A Systematic Review

Summary: Colorectal cancer (CRC) is a major cause of cancer-related deaths globally, and new treatment strategies are needed due to the side effects of current treatments. Fungi have gained attention as a potential source of antitumor compounds due to their antimicrobial and antioxidant properties. A systematic review of the literature found that extracts and compounds from Ascomycota fungi showed promising antitumor activity against CRC cells. The study identified the most studied order of fungi, the most commonly used extraction method, and highlighted specific compounds with high cytotoxic activity. The review also focused on the mechanisms through which fungi extracts and compounds inhibit CRC cell proliferation and migration, suggesting their potential for developing new therapies.

Single-cell transcriptomic profiling redefines the origin and specification of early adrenogonadal progenitors

Summary: This study provides a comprehensive single-cell transcriptomic atlas of early mouse adrenogonadal development, revealing the differentiation process and molecular mechanisms. It is found that the fate of adrenal and gonadal has diverged prior to Nr5a1 expression and involves differential expression of genes related to Wnt signaling and Hox patterning. This study provides important insights into the ontogenesis of adrenal and gonadal and will be a valuable resource for further research.

Gonadotropin axis and semen quality in young Swiss men after cannabis consumption: Effect of chronicity and modulation by cannabidiol

Summary: This study assessed the impact of cannabis consumption on semen parameters, reproductive hormones, and endocannabinoid levels in young Swiss men. The results showed higher levels of anandamide, androgens, estradiol, and sex hormone binding globulin in chronic, recent, and cannabidiol-positive cannabis users. Gonadotropin levels were not significantly different, while prolactin and albumin concentrations were lower in cannabis users. Moreover, cannabis users had a more basic semen pH and a higher percentage of spermatozoa with progressive motility, which may be influenced by sexual abstinence rather than cannabis use.

Vegetal-Derived Bioactive Compounds as Multidrug Resistance Modulators in Colorectal Cancer

Summary: Colorectal cancer is a major cause of illness and death, and drug resistance is a leading cause of treatment failure. Bioactive compounds derived from vegetables are being studied as a new approach to improve antitumor treatment and reduce drug resistance. In vitro and in vivo studies have shown that these compounds, such as polyphenols, flavonoids, and terpenes, can enhance therapeutic efficacy by reducing resistance. This review summarizes the current knowledge on the antitumor and chemo-sensitizing properties of vegetable-derived biomolecules and their potential in improving colorectal cancer treatment.

Cancer Stem Cells in Sarcomas: In Vitro Isolation and Role as Prognostic Markers: A Systematic Review

Summary: Sarcomas are a diverse group of neoplasms with a high incidence rate in childhood cancers. Cancer stem cells (CSCs) play a role in recurrence, metastasis, and drug resistance, making the search for diagnostic and prognostic biomarkers crucial. This systematic review analyzed the expression of CSC biomarkers in in vitro cell lines and patient tumor samples, revealing significant heterogeneity in markers and isolation techniques. ALDH was identified as a common marker in various types of sarcomas. The identification of CSC markers in sarcomas may aid in personalized medicine development and improve treatment outcomes.

Summary: Anemonia sulcata has potential as a source of marine natural products with antioxidant and antitumor activity. Further characterization and isolation of the bioactive compounds are required.

Combining Olaparib and Ascorbic Acid on Nanoparticles to Enhance the Drug Toxic Effects in Pancreatic Cancer

Summary: This study functionalized calcium phosphate-based nanoparticles with Olaparib and ascorbic acid to enhance their individual effects in treating pancreatic cancer. The resulting dual nanosystem showed a gradual release of Olaparib and ascorbic acid, demonstrating a better cytotoxic effect and apoptosis-mediated cytotoxic effect in human pancreatic ductal adenocarcinoma cell line. In vivo studies further confirmed the therapeutic benefits of the nanoparticle system in terms of tumor volume reduction and survival improvement.

Nanomedicine and Hyperthermia for the Treatment of Gastrointestinal Cancer: A Systematic Review

Summary: The incidence of gastrointestinal cancers has been increasing in recent years, leading to challenges in current treatments. A promising strategy is to use magnetic nanoparticles for hyperthermia therapy, generating heat in tumor regions using an external magnetic field. This systematic review analyzed 40 articles published in the last decade, showing that iron oxide is the preferred material for magnetism generation and colorectal cancer is the most studied gastrointestinal cancer. Combination therapies using nanoparticles loaded with chemotherapeutic drugs and magnetic hyperthermia demonstrated excellent antitumor effects. Hyperthermia treatments mediated by magnetic nanoparticles appear to be an effective approach for gastrointestinal cancers.

Evaluating Metabolite-Based Biomarkers for Early Diagnosis of Pancreatic Cancer: A Systematic Review

Summary: Pancreatic ductal adenocarcinoma (PDAC) is a deadly cancer with low survival rates. Surgical resection is the only curative option, but most patients are not eligible due to late diagnosis. Metabolomics has shown promise in identifying biomarkers for early PDAC detection. This review summarizes recent studies on metabolites for early PDAC diagnosis and highlights amino acid and lipid metabolic pathways as potential biomarkers.
