Causal effect of alcohol consumption on hyperuricemia using a Mendelian randomization design

Aim We used a Mendelian randomization analysis to assess the causal effect of alcohol consumption on hyperuricemia in Koreans. Methods The Korean Cancer Prevention Study-II (KCPS-II) Biobank cohort consisted of 156 701 healthy Korean aged 20 years or older. Clinical data including serum uric acid, alcohol consumption, and other related confounding variables were collected at baseline. The 27 single nucleotide polymorphisms (SNP) including rs671 in aldehyde dehydrogenase 2 (ALDH2) were obtained from a genome-wide association study of alcohol consumption in the KCPS-II Biobank among 11 678 men and women in 2017. Both unweighted and weighted genetic risk score (wGRS) were calculated using 10 SNPs selected based on linkage disequilibrium. Results As strong instrumental variables, both rs671 and wGRS were associated with an increased amount of alcohol drinking in men and women. Alcohol consumption was also positively associated with hyperuricemia risk in men (P < .001) and women (P = .014). Both rs671 major G allele and wGRS were not associated with hyperuricemia. In Mendelian randomization analysis, the causal relationship between any alcohol consumption and hyperuricemia was found only in men, albeit non-significant after correction for multiple testing. The associations did not change after excluding heavy drinkers or the elderly. Conclusions These results provide evidence that alcohol consumption is causally associated with risk of hyperuricemia in Korean men and support its role as a risk determinant.

Summary: This study aimed to examine the association between blood concentration of dioxin-like polychlorinated biphenyls (DL-PCBs) and polychlorinated dibenzo-p-dioxins and dibenzofurans (PCDD/DFs) and the risk of type 2 diabetes mellitus (T2DM) and thyroid cancer. The results showed a significant positive association between PCDD/DFs concentrations and both T2DM and thyroid cancer.

Summary: The age at death of people with diabetes has increased and reached a comparable level to those without diabetes. The increase in death age is higher in the subgroup of diabetes patients with dyslipidemia and/or hypertension. In contrast, deaths from vascular disease and diabetes have decreased, while deaths from cancer and pneumonia have increased. The findings suggest a narrowing gap in life expectancy between people with and without diabetes.

Transcriptome-metabolome-wide association study (TMWAS) in rats revealed a potential carcinogenic effect of DEHP in thyroid associated with eicosanoids

Summary: In this study, the mechanism of di2-ethylhexyl phthalate (DEHP) induced thyroid hyperplasia was explored through a transcriptome-metabolome-wide association study. It was found that metabolic shifts were significantly altered at doses above 30 mg DEHP/kg bw/day. RNA-seq analysis revealed several cancer-related genes that were upregulated or downregulated. Integration of these genes and the whole metabolome data uncovered thyroid cancer-related metabolic pathways, particularly those related to eicosanoids. These findings provide further insight into the mechanism of DEHP-associated thyroid cancer.

Summary: This study identified 89 independent loci associated with stroke and its subtypes through cross-ancestry GWAS analyses, with high replication rates for both primary and secondary stroke risk loci. It also revealed potential causal genes, variants, and drug targets, and demonstrated the predictive power of genetic risk scores for ischemic stroke across different ancestries.

Prognostic value of resting heart rate in predicting undiagnosed diabetes in adults: Korean National Health and Nutrition Examination Survey 2008-2018

Summary: This study aimed to investigate the association between resting heart rate (RHR) and the prevalence of undiagnosed diabetes. The results showed that participants with higher RHR had a significantly higher prevalence of undiagnosed diabetes, and this association was stronger among younger and leaner individuals. Therefore, RHR can be used as an indicator to identify and prevent undiagnosed diabetes.

Summary: This study identified 205 independent risk associations for colorectal cancer through a genome-wide association study meta-analysis. Transcriptome and methylome analysis further revealed 53 risk associations. These findings provide insights into the genetic and molecular mechanisms of colorectal oncogenesis and suggest potential targets for new treatment and prevention strategies.

Comparisons of the prediction models for undiagnosed diabetes between machine learning versus traditional statistical methods

Summary: This study compares the prediction performance of machine learning-based undiagnosed diabetes prediction models and traditional statistics-based prediction models. The results show that the machine learning-based models have higher AUC values when using features such as sex, age, and waist circumference. Similarly, when additional features including family history of diabetes, hypertension, alcohol consumption, and smoking status are included, the machine learning-based models still outperform the traditional statistics-based models. The study concludes that machine learning-based prediction models using anthropometric and lifestyle measurements may be more accurate in predicting undiagnosed diabetes.

Summary: Monitoring metabolic biomarkers could be used as an effective tool for the early detection of gastric cancer risk. This study discovered that changes in serum levels of l-carnitine and citric acid are associated with gastric cancer occurrence, and may serve as predictive serum biomarkers for gastric cancer.
