Fucoidans are sulfated polysaccharides from brown algae, known to have immunomodulatory activity. Their effects on the response of airway epithelial cells to Toll-like receptor 3 (TLR3) stimulation have not been characterized. Our objective was to evaluate the effects of a marine-sourced fucoidan solution (MFS) on the TLR3-induced expression and/or production of cytokines and prostaglandin by human primary bronchial epithelial cells as a model of the airway epithelium. The cells were incubated with MFS in the presence or absence of Poly(I:C) (a TLR3 agonist that mimics viral RNA). Cytokine expression and production were assessed using RT-qPCR and ELISA The expression of cyclooxygenase-2 and the production of prostaglandin E2 were also measured. Relative to control, exposure to MFS was associated with lower Poly(I:C)-induced mRNA expression of various cytokines and chemokines, and lower COX-2 production. The MFS inhibited the production of some cytokines (IL-1 alpha, IL-1 beta, TNF alpha, and IL-6), chemokines (CCL5, CCL22, CXCL1, CXCL5 and CXCL8) and prostaglandin E2 but did not alter the production of IL-12/25, CCL2 and CCL20. At clinically relevant concentrations, the MFS inhibited the TLR3-mediated production of inflammatory mediators by human primary bronchial epithelial cells - suggesting that locally applied MFS might help to reduce airway inflammation in viral infections. In Vitro Chemopreventive Potential of Phlorotannins-Rich Extract from Brown Algae by Inhibition of Benzo[a]pyrene-Induced P2X7 Activation and Toxic Effects

Melody Dutot, Elodie Olivier, Sophie Fouyet, Romain Magny, Karim Hammad, Emmanuel Roulland, Patrice Rat, Roxane Fagon

Summary: The study evaluated the effects of phlorotannins on the response of human lung cells to benzo[a]pyrene (B[a]P), showing that the brown algae extract had a protective effect against B[a]P-induced cytotoxicity. It was observed that the brown algae extract inhibited CYP1 activity, reduced P2X7 receptor activation, and lowered reactive oxygen species production during exposure to B[a]P.

Pregnant Women and Endocrine Disruptors: Role of P2X7 Receptor and Mitochondrial Alterations in Placental Cell Disorders

Sophie Fouyet, Elodie Olivier, Pascale Leproux, Melody Dutot, Patrice Rat

Summary: This study investigates the toxicity mechanisms of endocrine-disrupting chemicals (EDCs) in pregnant women by focusing on the activation of the P2X7 receptor in different cell types. The results show that EDCs can activate the P2X7 receptor in skin and placenta cells, but not in lung non-endocrine cells. The activation of the P2X7 receptor and induction of apoptosis are shared pathways among all tested EDCs in placental cells.

Cocktail Effect of Endocrine Disrupting Chemicals: Application to Chlorpyrifos in Lavender Essential Oils

Sophie Fouyet, Elodie Olivier, Pascale Leproux, Sabrina Boutefnouchet, Melody Dutot, Patrice Rat

Summary: Chlorpyrifos, a banned pesticide, can still be found in soil, posing a risk to pregnant women through drinking water and herbal products. This study evaluated the endocrine disrupting effects of chlorpyrifos in lavender essential oil (EO) compared to its free form. The results showed that free chlorpyrifos disrupted placental hormones and activated the P2X7 receptor, while chlorpyrifos in lavender EO only disrupted placental hormones. This study confirms that chlorpyrifos is an endocrine disrupting chemical (EDC) for pregnant women and highlights the potential protective effects of lavender EO.

Evaluation of Placental Toxicity of Five Essential Oils and Their Potential Endocrine-Disrupting Effects

Summary: This study aimed to explore and compare the placental toxicity and endocrine disrupting effects of various essential oils (EOs) and their key compounds. The results showed that the EOs and their key components disrupted at least one of the studied hormones in placental cells, but did not activate the cell death receptor. Therefore, these tested EOs appear to act as hormonal modulators rather than endocrine disrupting compounds in human placental cells.

Effects of aging and diabetes on the deformation mechanisms and molecular structural characteristics of collagen fibrils under daily activity

Wei-Han Hui, Yen-Lin Chen, Shu-Wei Chang

Summary: Crosslinking affects the mechanical behavior and tissue metabolism of collagen-based tissues. Aging and diabetes alter the crosslinking type and density, thus changing tissue properties. High-connectivity enzymatic crosslinking and advanced-glycation end-products result in uniform deformation under daily activity, while low-connectivity enzymatic crosslinking does not. High-connectivity model displays more sliding, while AGEs induce instability in structures near the binding sites.
