Evaluation and use of an anti-cynomolgus monkey CD79b surrogate antibody-drug conjugate to enable clinical development of polatuzumab vedotin

Background and Purpose Polatuzumab vedotin is an antibody-drug conjugate (ADC) being developed for non-Hodgkin's lymphoma. It contains a humanized anti-CD79b IgG1 monoclonal antibody linked to monomethyl auristatin E (MMAE), an anti-mitotic agent. Polatuzumab vedotin binds to human CD79b only. Therefore, a surrogate ADC that binds to cynomolgus monkey CD79b was used to determine CD79b-mediated pharmacological effects in the monkey and to enable first-in-human clinical trials. Experimental Approach Polatuzumab vedotin, the surrogate ADC, and the corresponding antibodies were evaluated in different assays in vitro and in animals. In vitro assessments included binding to peripheral blood mononuclear cells from different species, binding to a human and monkey CD79b-expressing cell line, binding to human Fc gamma receptors, and stability in plasma across species. In vivo, ADCs were assessed for anti-tumour activity in mice, pharmacokinetics/pharmacodynamics in monkeys, and toxicity in rats and monkeys. Key Results Polatuzumab vedotin and surrogate ADC bind with similar affinity to human and cynomolgus monkey B cells, respectively. Comparable in vitro plasma stability, in vivo anti-tumour activity, and mouse pharmacokinetics were also observed between the surrogate ADC and polatuzumab vedotin. In monkeys, only the surrogate ADC showed B-cell depletion and B-cell-mediated drug disposition, but both ADCs showed similar MMAE-driven myelotoxicity, as expected. Conclusions and Implications The suitability of the surrogate ADC for evaluation of CD79b-dependent pharmacology was demonstrated, and anti-tumour activity, pharmacokinetics/pharmacodynamics, and toxicity data with both ADCs supported the entry of polatuzumab vedotin into clinical trials.

Subsurface drainage influences the structure and assembly of soil bacterial and fungal communities in salinized cotton field

Summary: Exploring the impact of subsurface drainage on microbial community structures in saline-alkali soils is important for maintaining the sustainability of saline-alkali fields. This study found that subsurface drainage treatments significantly reduced soil salt, but had no effect on microbial richness and diversity. The depth and spacing of drainage had significant effects on bacterial and fungal composition structures, with soil microbial community assembly mainly driven by stochastic processes.

Study on the high-temperature damping properties of silicone rubber modified by boron-terminated polysiloxane

The reaction involves the formation of sugar-based alpha-substituted propanedinitriles followed by diverse oxidative transformations, offering a mild and compatible method for sugars with various functional/protecting groups. The method is demonstrated through the assembly of Campylobacter jejuni strain 81-176 capsular trisaccharide repeating unit derivatives.

Community attitudes towards Amur tigers (Panthera tigris altaica) and their prey species in Yanbian, Jilin province, a region of northeast China where tigers are returning

Summary: This study examines community attitudes and perceptions towards large carnivores, including the Amur tiger, Amur leopard, and bears, as well as their prey species in northeast China. The survey reveals neutral attitudes towards large carnivores but strongly negative attitudes towards wild boar, particularly when agricultural products are lost to crop raiding by wild boar. The findings suggest that targeting messaging around the tiger's role as a key predator of wild boar can encourage support for large carnivore recovery and conservation.

Quasi-solid polymer electrolytes with fast interfacial transport for lithium metal batteries

Summary: The quasi-solid electrolyte based on Poly(vinylidene fluoride-co-hexafluoropropylene) shows high ionic conductivity and lithium-ion transfer number through the synergistic effect with the NASICON-type lithium-ion conductor Li1.3Al0.3Ti1.7(PO4)3. This quasi-solid polymer electrolyte exhibits high conductivity and stable cycling performance at room temperature, and can be paired with different cathode materials for good electrochemical performance. In addition, the assembled pouch cell has good flexibility, making it suitable for various applications.

The (002) exposing facets of WO3 boosting photocatalytic degradation of nitrobenzene

Summary: This study presents a novel strategy for the construction of flexible fiber-shaped supercapacitors (FSSCs) using boron-doped diamond (BDD) film grown on a flexible tantalum (Ta) fiber substrate. The BDD fiber film exhibits significantly higher specific capacitance compared to planar BDD electrodes. The assembled symmetric supercapacitor device demonstrates desirable stability with high capacitance retention and satisfactory bendability under various bending deformations.

High performance of conversion and selectivity of methane to CO via Pt-Pd core-shell structural catalyst

Summary: This paper investigates the catalytic oxidation of methane to CO using a novel core-shell Pt-Pd catalyst. c-Jun as a one-way valve at the naive to primed interface

Summary: The study finds that c-Jun functions as a one-way valve in embryonic stem cells, preserving the primed state and preventing reversion to the naive state. It stabilizes the chromatin structure and inhibits the reverse transition. Loss or impairment of c-Jun accelerates the activation of naive-specific enhancers and enhances the efficiency of cell fate conversion.
