Summary: Antibody-drug conjugates (ADCs) are innovative biopharmaceuticals that combine potent cytotoxic drugs with monoclonal antibodies to target cancer cells. This study investigated the stability and degradation of two ADCs, POLA and SGN-35, under various stress conditions. Different analytical techniques, including SE-HPLC, DLS, HIC-HPLC, RP-HPLC, and ELISA, were used to quantify degradation products, monitor aggregation and fragmentation, determine drug antibody ratio, and test binding activity. The results demonstrated the importance of using multiple techniques to evaluate the quality and stability of ADCs.

Summary: The antibody-drug conjugate polatuzumab vedotin (pola) in combination with bendamustine and rituximab (pola-BR) has shown efficacy for patients with refractory or relapsed large B-cell lymphoma in a real-world setting. The treatment achieved encouraging overall response rates in salvage therapy and successfully bridged a significant number of patients to CAR T-cell therapy. These findings suggest that pola may serve as an effective salvage and bridging treatment option for patients with refractory or relapsed LBCL.

Summary: The CD30 antibody-drug conjugate Brentuximab Vedotin (BV) shows strong anti-cancer efficacy not only in CD30(+) hematological malignancies but also in diffuse large B cell lymphoma (DLBCL) with poor CD30 expression, possibly through the involvement of extracellular vesicle (EV)-associated CD30 in tumor cells. EVs not only communicate within the tumor microenvironment but also influence cancer treatment, indicating a potential mechanism for BV binding and uptake in CD30(-) DLBCL cells. Further studies in animal models are warranted to confirm these findings.

Summary: CD30 antibody-drug conjugate Brentuximab Vedotin (BV) is effective in treating CD30(+) hematological malignancies and shows strong anti-cancer efficacy in diffuse large B cell lymphoma (DLBCL) even with poor CD30 expression. Research suggests that CD30 released on extracellular vesicles (EVs) may influence BV binding in tumor cells.

Summary: This retrospective analysis examined the efficacy and progression of PV combination therapy in LBCL patients who relapsed or progressed after CAR-T cell therapy. The treatment achieved a favorable response in some patients, but the majority still experienced disease progression. Bone marrow involvement and elevated lactate dehydrogenase levels were associated with shorter progression-free survival.

Summary: The study focuses on the impact of CD3 binding affinity on the pharmacokinetics of T cell-dependent bispecific antibodies. The results demonstrate that CD3 binding affinity affects the clearance rate of the antibody and may be related to the internalization rate of CD79b and the binding ability of CD3.

Summary: Enfortumab vedotin (EV), an antibody-drug conjugate, showed significant survival benefits compared to standard chemotherapy in patients with urothelial carcinoma who had previously received platinum-based chemotherapy and a PD-1 or PD-L1 inhibitor. Three cases of patients with active brain metastases receiving EV treatment were reported, with two cases showing significant reduction in metastases and one case achieving partial remission and disappearance of neurological symptoms. These reports provide the first evidence of the efficacy of EV in urothelial carcinoma patients with brain metastases.

Summary: Exploring the impact of subsurface drainage on microbial community structures in saline-alkali soils is important for maintaining the sustainability of saline-alkali fields. This study found that subsurface drainage treatments significantly reduced soil salt, but had no effect on microbial richness and diversity. The depth and spacing of drainage had significant effects on bacterial and fungal composition structures, with soil microbial community assembly mainly driven by stochastic processes.

Summary: An efficient approach to construct 6-deoxy-heptose compounds has been developed using the sugar chain of hexoses. The reaction involves the formation of sugar-based alpha-substituted propanedinitriles followed by diverse oxidative transformations, offering a mild and compatible method for sugars with various functional/protecting groups. The method is demonstrated through the assembly of Campylobacter jejuni strain 81-176 capsular trisaccharide repeating unit derivatives.

Summary: This study examines community attitudes and perceptions towards large carnivores, including the Amur tiger, Amur leopard, and bears, as well as their prey species in northeast China. The survey reveals neutral attitudes towards large carnivores but strongly negative attitudes towards wild boar, particularly when agricultural products are lost to crop raiding by wild boar. The findings suggest that targeting messaging around the tiger's role as a key predator of wild boar can encourage support for large carnivore recovery and conservation.
