TRPV2 Calcium Channel Gene Expression and Outcomes in Gastric Cancer Patients: A Clinically Relevant Association

Gastric cancer (GC) is characterized by poor efficacy and the modest clinical impact of current therapies. Apoptosis evasion represents a causative factor for treatment failure in GC as in other cancers. Since intracellular calcium homeostasis regulation has been found to be associated with apoptosis resistance, the aberrant expression of intracellular calcium regulator genes (CaRGs) could have a prognostic value in GC patients. We analyzed the association of the expression levels of 98 CaRGs with prognosis by the log-rank test in a collection of 1524 GC samples from four gene expression profiling datasets. We also evaluated differential gene expression in comparison with normal stomach tissue, and then we crossed results with tissue microarrays from the Human Protein Atlas. Among the investigated CaRGs, patients with high levels of TRPV2 expression were characterized by a shorter overall survival. TRPV2 expression was found to increase according to tumor stage. Both mRNA and protein levels were significantly higher in tumor than normal stomach samples. TRPV2 was also associated with poor prognosis in the Lauren's intestinal type GC and in patients treated with adjuvant therapy. Overall, we highlighted the relevance of TRPV2 not only as a prognostic biomarker but also as a potential therapeutic target to improve GC treatment efficacy.

Summary: Recent advances in molecular subtyping of gastric cancer have identified new clinically relevant biomarkers and potential therapeutic targets. Checkpoint inhibitors have also expanded treatment options, showing promising results in certain subgroups of gastric cancer patients. However, cytotoxic chemotherapy remains the only choice for patients who do not have these biomarkers. This study aims to investigate the prevalence, clinicopathologic features, and treatment outcomes of molecular subtypes in a new-diagnosed cohort of Western gastric cancer patients.

Can legionellosis be considered an occupational risk in the healthcare sector? A systematic review and meta-analysis

Summary: This study systematically reviewed the literature on healthcare workers' (HCWs) exposure to legionella and assessed whether there is an occupational risk. The findings suggest a higher risk of legionella exposure for HCWs, but clinical evidence is currently lacking. Further research with appropriate study design is needed to determine whether legionella infection is an occupational risk for HCWs.

Analysis of Knowledge, Attitudes and Behaviours of Health Care Workers towards Vaccine-Preventable Diseases and Recommended Vaccinations: An Observational Study in a Teaching Hospital

Summary: This study aimed to evaluate the knowledge, attitudes, and behaviors of healthcare workers (HCWs) in relation to national vaccination recommendations. The study found that half of the HCWs were aware of the vaccination recommendations, with occupation and age being predictive factors. Although a majority of the sample perceived the diseases as a risk to their patients' health, this perception was lower in the over-40 age group. There were statistically significant differences between the care units. Approximately three-quarters of the sample agreed that vaccination should be mandatory, and willingness to undergo a future booster vaccination was correlated with age and care unit. The protection of health in the workplace can be achieved through strategic actions such as educational strategies, monitoring of immunocompetence, and improvement of vaccination.

Relationship between formaldehyde exposure, respiratory irritant effects and cancers: a review of reviews

Summary: The study aimed to investigate the correlation between formaldehyde exposure and irritant diseases (mainly asthma) and neoplastic diseases. The results showed a strong association between formaldehyde exposure and asthma, but a weak association with cancer. Although the associations were not strong, the World Health Organisation recommends limiting formaldehyde levels to 0.1 mg/m3 (0.08 ppm).

Summary: This study aims to analyze the role of vaccination status in modulating the pattern of symptomatology associated with COVID-19 infection. The results showed that females were at a higher risk of developing symptoms. Four SARS-CoV-2 waves were identified, with pharyngitis and rhinitis being more frequent during the fourth wave among vaccinated subjects, while cough, fever, flu syndrome, headache, anosmia, ageusia, arthralgia/arthritis, and myalgia were more frequent during the first three waves among unvaccinated subjects. The study concluded that vaccination status and viruses' mutations had a synergic effect in mitigating the symptomatology caused by SARS-CoV-2.

Role of IL-6/STAT3 Axis in Resistance to Cisplatin in Gastric Cancers

Summary: Gastric cancer, a leading cause of death globally, is difficult to treat with chemotherapy due to multidrug resistance. This study investigated the role of STAT3 in gastric cancer drug efficiency through gene expression analysis. The study found that gastric cancer cells become resistant to chemotherapy by activating alternative signaling pathways. However, inhibiting IL-6/STAT3 in combination with cisplatin treatment prevented STAT3 activation and increased lethality compared to single-agent treatment. This suggests that IL-6/STAT3 axis blockage may be a strategy to overcome resistance.

Summary: The methylation profile of HSPCs following AHSCT was analyzed and found to undergo changes that may be useful in transplant diagnostics. The methylation signature was also found to be related to patient outcome. This study suggests that the methylation profile of HSPCs can provide prognostic information for engraftment success and graft failure prediction in AHSCT.

Summary: The impact of pregnancy and breastfeeding on the development and outcomes of Multiple sclerosis (MS) is uncertain and debated. Two cases of late-onset MS in parous women who breastfed their children and one of them received hormone replacement therapy were reported. Studies suggest that the beneficial effects of pregnancy and breastfeeding on MS outcome may only occur with multiple pregnancies. However, further research is needed to evaluate the protective role of multiparity in larger prospective studies.
