Summary: In this study, the feasibility and mechanism of cordycepin combined with doxorubicin for liver cancer treatment were investigated. Both in vitro and in vivo studies demonstrated that the combination effectively inhibited cell proliferation and induced apoptosis in hepatocellular carcinoma. Network pharmacology analysis revealed the potential targets and pathways involved in the anticancer mechanism. Additionally, the combination treatment regulated the expression of genes related to cell migration, thus inhibiting cell migration.

Summary: Platycodon grandiflorum is a medicinal plant commonly used in East Asia, and its triterpene saponins have shown anti-tumor effects. In this study, the inhibitory effect of polygalacin D (PGD) on hepatocellular carcinoma cells was investigated. PGD was found to induce apoptosis and autophagy in these cells through the mitochondrial apoptosis and mitophagy pathways. In addition, in vivo experiments confirmed the anti-tumor activity of PGD. These findings suggest that PGD can be developed as a novel anti-tumor agent by targeting the apoptosis and autophagy pathways.

Summary: In this study, caspase-8 was eliminated in MC3T3-E1 cells using CRISPR/cas9 technology, and the consequences of caspase-8 deficiency on non-apoptotic pathways in osteoblastic cells were investigated. The results showed that caspase-8 deficiency inhibited osteogenic differentiation and affected the expression of osteogenic genes.

Summary: In this study, the inhibitory effects of Fangchinoline (FCN) on c-Met, a protein involved in cancer growth and metastasis, were analyzed. FCN was found to inhibit the activation of c-Met and its downstream signals and induce apoptotic cell death. Additionally, FCN suppressed the expression of various oncogenic proteins. In a mouse model, FCN significantly attenuated tumor volume and weight. These findings suggest that FCN could potentially be a potent anticancer agent against hepatocellular carcinoma.

Summary: The tumor-suppressive lncRNA CASC2 is downregulated in hepatocellular carcinoma (HCC) and its low expression is associated with shorter overall survival. CASC2 overexpression inhibits HCC cell proliferation and induces cell apoptosis. CASC2 regulates TRAIL-sensitive HCC cells through the miR-24/caspase-8 and miR-221/caspase-3 axes and caspase cascades, while in TRAIL-resistant HCC cells, it regulates cell growth through the miR-18a/RIPK1 axis and the NF-kappa B signaling.

Summary: In this study, it was found that dimethyl itaconate (DI) can effectively inhibit the development of hepatocellular carcinoma (HCC) in vivo by regulating multiple signaling pathways and molecular mechanisms. Through the modulation of mitochondrial apoptosis, the NF-kappa B pathway, and metabolomic profiles, DI shows promising anti-HCC activity in DEN-induced HCC rats.
