Dietary advice is the cornerstone in first-line treatment of metabolic diseases. Nutritional interventions directed at these clinical conditions mainly aim to (a) improve insulin resistance by reducing energy-dense macronutrient intake to obtain weight loss and (b) reduce fluctuations in insulin secretion through avoidance of rapidly absorbable carbohydrates. However, even in the majority of motivated patients selected for clinical trials, massive efforts using this approach have failed to achieve lasting efficacy. Less attention has been given to the role of micronutrients in metabolic diseases. Here, we review the evidence that highlights (a) the importance of iron in pancreatic beta-cell function and dysfunction in diabetes and (b) the integrative pathophysiological effects of tissue iron levels in the interactions among the beta cell, gut microbiome, hypothalamus, innate and adaptive immune systems, and insulin-sensitive tissues. We propose that clinical trials are warranted to clarify the impact of dietary or pharmacological iron reduction on the development of metabolic disorders.

Treatment of Hyperthyroidism Reduces Systemic Oxidative Stress, as Measured by Markers of RNA and DNA Damage

Summary: The study found that urinary excretion of oxidative stress markers in hyperthyroid patients was positively correlated with plasma thyroxine levels, with higher levels in GD compared to TNG patients. After treatment, both GD and TNG patients showed a decrease in the excretion of oxidative stress markers, indicating a reduction in systemic oxidative stress load with restoration of euthyroidism.

Thyroid Function and Mood Disorders: A Mendelian Randomization Study

Allostatic load as predictor of mortality: a cohort study from Lolland-Falster, Denmark

A novel integrated biomarker index for the assessment of hematological responses in MPNs during treatment with hydroxyurea and interferon-alpha2

Summary: This study describes a novel index for assessing hematological and molecular responses in MPN patients during treatment with IFN or HU. The results demonstrate the superiority of IFN over HU in normalizing cell counts. Further validation and studies on the safety and efficacy of combination therapy are warranted.

Kidney function and risk of dementia: Observational study, meta-analysis, and two-sample mendelian randomization study

Summary: This study investigated the association between impaired kidney function and the risk of developing dementia. The results showed a modestly increased risk of dementia in individuals with impaired kidney function, but genetic analysis did not support a causal relationship. However, further research is needed to re-evaluate the causal hypothesis.

Risk of asthma in offspring of asthmatic fathers versus mothers: A population-based study of 21,000 individuals in Denmark

Summary: Parental asthma or allergy is associated with increased risk of asthma in children, although the extent of this relationship varies in different study populations. The impact of maternal and paternal asthma history is debated, with some countries/populations showing a stronger maternal effect, others showing equivalent parental effects, and a few studies indicating a dominant paternal effect. In the Danish GESUS study, we aimed to determine the effect of parental asthma and allergy on asthma risk in the population. Our findings suggest that the contribution of maternal and paternal asthma effects is approximately equal, except for a slightly greater effect of paternal allergy on ever asthma. The presence of gestational tobacco smoking did not affect the maternal results. Overall, our study highlights the strong association between parental asthma/allergy and increased asthma risk in offspring, without a prominent maternal or paternal effect.

Long-term stability of thyroid peroxidase antibody (anti-TPO) in serum in the Danish General Suburban Population Study

Summary: The long-term stability of thyroid peroxidase antibody (anti-TPO) was evaluated in a Danish population study. It was found that there was minimal change in anti-TPO levels after 12 years of storage at -80 degrees Celsius.

Allometric body shape indices, type 2 diabetes and kidney function: A two-sample Mendelian randomization study

Summary: This study used two-sample Mendelian randomization analysis to examine the association between BMI-independent allometric body shape indices and kidney function. The results showed that genetically high ABSI and WHI levels were associated with higher urinary albumin-to-creatinine ratio (UACR) in women, while genetically high HI levels were associated with lower UACR in both men and women. The risk of type 2 diabetes (T2D) was also affected by these body shape indices. A score of 8 out of 10 is given for the importance of this study.

Electrocardiography in euthyroid individuals: a Danish general population study

Summary: This study investigated the association between thyroid hormone concentrations within the euthyroid range and various electrocardiographic markers in people free of thyroid disease. The results showed that concentrations of T3 and T4 were associated with an increase in heart rate, while TSH showed no relation to heart rate. Additionally, higher concentrations of T3 and T4 were linked to changes in other electrocardiogram parameters.
