Microcystin- (MC-) LR is the most frequent cyanotoxin produced by Microcystis aeruginosa cyanobacteria in the contaminated freshwater environment. MC represents a health hazard to humans and animals. Therefore, the present study was designed to evaluate the potential ameliorative effect of thymoquinone (TQ) and/or piperine (PP) against MC toxicity in mice. Fifty-six mice were randomly divided into seven experimental groups. Group I is the normal control that received distilled water for 21 days; Group II (TQ) was treated with TQ (10 mg/kg, i.p) for 21 days; Group III (PP) was treated with PP (25 mg/kg, i.p) for 21 days; Group IV (MC) was treated with MC (10 mu g/kg, i.p) for 14 days and served as the toxic control; and Groups V, VI, and VII received TQ and/or PP 7 days prior to MC and continued for 14 days with MC. The results revealed that MC elicited hepatotoxicity and neurotoxicity which was evident due to the significant elevation of serum AST, ALT, gamma GT, ALP, LDH, IL-1 beta, IL-6, and TNF-alpha levels. Furthermore, MC markedly increased MDA and NO contents along with reduction of GSH, SOD, CAT, and GSH-Px in liver and brain tissues. The electron transport chain may be a possible target for MC. TQ and/or PP ameliorated the MC-mediated oxidative damage in the liver and brain which might be attributed to their antioxidant properties. However, the concurrent treatment of TQ and PP showed the best regimen as a result of the PP-enhanced bioavailability of TQ.

Mechanistic Insights, Treatment Paradigms, and Clinical Progress in Neurological Disorders: Current and Future Prospects

Saad Alkahtani, Norah S. S. AL-Johani, Saud Alarifi

Summary: Neurodegenerative diseases (NDs) are a major cause of disability and are related to brain development. The neurological signs of brain lesions can vary from mild clinical shortfalls to more delicate and severe neurological/behavioral symptoms and learning disabilities, which are progressive. In this paper, we have tried to summarize a collective view of various NDs and their possible therapeutic outcomes.

Molecular Evolution of Interferon-Epsilon (IFNε) Pseudogene Modulates Innate and Specific Antiviral Immunity in Manis javanica

Hafiz Ishfaq Ahmad, Laraib Jameel, Quratulain Zahra, Jiabin Zhou, Linmiao Li, Xiujuan Zhang, Shakeel Ahmed, Daoud Ali, Gokhlesh Kumar, Aleena Safdar, Farhan Abbas, Jinping Chen

Summary: This study examines the molecular evolution of the IFN epsilon pseudogene in Manis javanica and its role in modulating the innate and specific antiviral immunity in this species. It finds evidence of positive selection in the human and primate IFN epsilon genes, suggesting their importance in the evolution of the immune system. Comparative genomic analysis reveals that the IFN epsilon pseudogene in pangolins originated from a gene duplication event and has lost its protein-coding function, but it may still play a regulatory role in antiviral immunity.

Health risk assessment of heavy metals in the seafood at Kalpakkam coast, Southeast Bay of Bengal

Kumar Pandion, Kantha Deivi Arunachalam, Rajinikanth Rajagopal, Daoud Ali, Saud Alarifi, Soon Woong Chang, Balasubramani Ravindran

Summary: The distribution of heavy metals in seafood intake was studied in the Kalpakkam coastal region. The average concentrations of heavy metals in 40 different fish species were determined. Zinc and copper were found to have higher bioaccumulation index and metal pollution index. Risk assessment calculations showed high values for different age groups. However, the cancer risk did not exceed recommended limits in the region.

Summary: An onco-informatics-based evaluation was conducted to assess the expression and predictive significance of the MDM2 gene in brain lower-grade glioma (LGG) cancer. The study analyzed the differential expression of MDM2 and searched for mutations and copy number variations. Several transcriptome servers and bioinformatics tools were used to determine the survival rate and protein interactions related to the MDM2 gene in LGG cancer patients.

In:SnO2/Yb2S3:Cu2S:ZnS: A Rare Earth Metal Sulfide-Conjugated Transition Metal Sulfide Photoactive Electrode

Mahwash Mahar Gul, Khuram Shahzad Ahmad, Andrew Guy Thomas, Daoud Ali

Summary: Spin coating with a single source precursor was used to produce a transparent conductive electrode for a photoelectrochemical cell. Yb2S3:Cu2S:ZnS composite with a crystallite size of 44 nm was discovered using X-ray diffraction analysis. The composite was found to have an optical bandgap of 2.49 eV and comprised Yb, Cu, Zn, and S. Electrical investigations in a photoelectrochemical cell showed that the photocurrent density of the electrode was higher under light and the specific capacitance increased at all scan rates under illumination. The study introduced a useful photoelectrode for renewable energy systems.
