Understanding mechanisms of oocyte development by follicular fluid lipidomics

PurposeThe present study aimed to provide a non-invasive approach to studying mechanisms responsible for oocyte development.MethodsTo this end, follicular fluid (FF) from 62 patients undergoing in vitro fertilization (IVF) cycles was split into two groups depending on the pregnancy outcome: pregnant (n=28) and non-pregnant (n=34) groups. Data were acquired by the MALDI-TOF mass spectrometry. Principal component analysis (PCA) and orthogonal partial least squares discriminant analysis (OPLS-DA) were applied to the data set. A ROC curve, to predict success rate, was constructed, and the lipids were attributed.ResultsSix ions were differentially represented in FF of pregnant and non-pregnant patients, with an area under the curve of 0.962. Phosphatidic acid, phosphatidylglycerol, and triacylglycerol were hyper-represented in the pregnant group, while glucosylceramide was hyper-represented in the non-pregnant group. Enriched functions related to these lipids are steroidogenesis, cellular response, signal transduction, cell cycle, and activation of protein kinase C for the pregnant group and apoptosis inhibition for the non-pregnant group.ConclusionHuman FF fingerprinting can both improve the understanding concerning mechanisms responsible for oocyte development and its effect on embryo implantation potential and assist in the management of IVF cycles.

Summary: In this study, adventitious roots of Pfaffia glomerata were cultured in vitro under dark conditions using roller bottle and orbital gyratory systems. The quantification of beta-ecdysone by LC-MS/MS revealed the highest accumulation after 8 weeks in the roller bottle system, demonstrating the potential of adventitious root culture as an alternative strategy for beta-ecdysone production. This study highlights the practical applications of bioreactors in the continuous production of high-quality compounds.

Previous infection with SARS-CoV-2 impacts embryo morphokinetics but not clinical outcomes in a time-lapse imaging system

Summary: The study aimed to investigate the impact of previous infection with SARS-CoV-2 on embryo morphokinetics and implantation. A historical cohort study was conducted in an in vitro fertilization center, including 1628 embryos from 88 patients. The results showed that embryos from patients with previous COVID-19 infection had longer duration in various developmental stages compared to the control group. However, there were no differences in clinical outcomes between the two groups.

Time-lapse monitoring: An adjunct tool to select embryos for preimplantation genetic testing

Summary: The present study aimed to investigate the effect of aneuploidy on embryo morphokinetics events using a time-lapse imaging (TLI) system incubator. A retrospective cohort study was conducted in a private university-affiliated in vitro fertilization center between March 2019 and December 2020. Our findings suggest that TLI monitoring may be an adjunct approach to select embryos for preimplantation genetic testing (PGT), but further investigation is still needed.

The effect of sperm DNA fragmentation on ICSI outcomes depending on oocyte quality

Summary: This study aimed to investigate the relationship between sperm DNA fragmentation and intracytoplasmic sperm injection (ICSI) outcomes, and whether this relationship is influenced by the presence of oocyte dimorphisms. The results showed that ICSI cycles with lower sperm DNA fragmentation had higher rates of fertilization, high-quality embryos, implantation, and pregnancy. The presence of oocyte dimorphisms significantly affected the outcomes. The lowest rates of fertilization and high-quality embryos were observed when high sperm DNA fragmentation was associated with certain oocyte defects, such as dark cytoplasm, vacuoles, resistant membrane, and non-resistant membrane.

Summary: Cystic fibrosis is a potentially fatal monogenic disease that has been significantly improved by the introduction of CFTR modulator drugs, particularly the triple combination therapy of elexacaftor, tezacaftor, and ivacaftor (ETI), which has shown safety and efficacy in reducing various disease symptoms. However, close monitoring by a healthcare team is necessary due to possible adverse effects associated with ETI therapy. This review aims to discuss the therapeutic benefits and adverse effects of ETI therapy for cystic fibrosis patients.

Biological Characterization and Antimicrobial Bioactives of Mycelium Extracts from Medicinal Mushrooms Phellinus linteus and Pleurotus albidus (Agaricomycetes)

Summary: This study aimed to evaluate the antimicrobial capacity and to separate and characterize bioactives from the mycelium of medicinal mushrooms Pleurotus albidus and Phellinus linteus. Results showed that P. albidus extracts had better antimicrobial activity against Bacillus strains, while Ph. linteus extracts were more effective against Staphylococcus aureus and Pseudomonas aeruginosa. Aqueous extraction was found to be the best method for obtaining bioactive compounds of P. albidus, while 30% hydroalcoholic extraction performed best for obtaining Ph. linteus. The identification of the main chemical compounds extracted from fungal biomasses was done through mass spectrometry analyses.
