Summary: The combination of cisplatin-based induction or adjuvant chemotherapy with concurrent chemoradiotherapy has been found to improve overall survival in patients with locoregionally advanced nasopharyngeal carcinoma. The optimal platinum doses for the concurrent and induction/adjuvant phases were determined to be 160 mg/m(2) and 260 mg/m(2) respectively.

Summary: The study found that the combination of nimotuzumab with chemoradiotherapy is effective for the treatment of nasopharyngeal carcinoma, especially when used in combination with induction chemotherapy and concurrent chemoradiotherapy. The addition of nimotuzumab did not result in a significant increase in toxicities.

Summary: The study revealed that in locoregionally advanced nasopharyngeal carcinoma patients, the use of IMRT appears to increase the risk of acute radiation-induced injuries compared to 3D-CRT, particularly in dermatitis. Patients in the IMRT group showed a lower risk of bone marrow toxicity, but a higher risk of leukopenia and gastrointestinal injury.

Summary: The study found that a higher cumulative cisplatin dose during concurrent chemoradiotherapy is associated with improved disease-free survival and distant metastasis-free survival in children and adolescents with locoregionally advanced nasopharyngeal carcinoma receiving neoadjuvant chemotherapy. Further matched analysis confirmed the favorable prognostic influence of high cisplatin dose on survival outcomes.

Summary: The study analyzed the outcomes and toxicities of ICT followed by CCRT plus ACT in LA-NPC patients, and found that this treatment approach did not significantly improve DFS and OS compared to adding ICT to CCRT, with more acute adverse events. Longer-term clinical studies are needed to evaluate treatment outcomes and late toxicities.

Summary: Concurrent chemoradiotherapy plus nimotuzumab after neoadjuvant chemotherapy for locally advanced nasopharyngeal carcinoma achieved higher complete remission rate and significantly improved distant metastasis-free survival compared with concurrent chemoradiotherapy alone. Additionally, an increasing trend was observed in progression-free survival, and the incidence of side effects was similar in both groups.

Summary: The purpose of this study was to evaluate the long-term survival, late toxicity profile, and quality of life of patients with advanced nasopharyngeal carcinoma who received combined induction chemotherapy and concurrent chemoradiotherapy. The results showed that reducing the target volumes of intensity modulated radiation therapy after induction chemotherapy did not increase the risk of locoregional relapse and may improve quality of life and reduce late toxicity.

Summary: Different induction chemotherapeutic strategies have varying effects and risks on the survival outcomes of patients with locoregionally advanced nasopharyngeal carcinoma. Gemcitabine plus platinum and docetaxel combined with 5-fluorouracil plus platinum strategies were associated with the highest rates of overall survival. The objective response and disease control rates were high after the completion of induction chemotherapy.

Summary: A deep-learning model based on MRI features and clinical variables was developed to predict distant metastasis-free survival in locoregionally advanced nasopharyngeal carcinoma patients. The model showed good performance in training, validation, and testing cohorts. Patients in low-risk groups treated with concurrent chemoradiotherapy alone had better outcomes compared to those who received additional chemotherapy.

Summary: This study investigated the long-term survival rates and toxicities of early-stage nasopharyngeal carcinoma (NPC) in an endemic area. The results showed that intensity-modulated radiotherapy (IMRT) can achieve superior tumor control and satisfactory long-term outcomes with mild late toxicities in early-stage NPC patients. Chemotherapy should be carefully performed in stage II patients due to increased toxicities.

Summary: For patients with high PD-L1 expression, PD-1 inhibitors combined with platinum-doublet therapy have significant benefits in progression-free survival. For patients with PD-L1 <50%, anti-PD-1/PD-L1 therapy is comparable to bevacizumab. In the liver metastatic population, anti-PD-1 combined with chemotherapy may offer benefits in progression-free survival.

Summary: This study investigated the radiobiological response of ultrahigh dose-rate irradiation (FLASH-IR) using 6 MV X-ray and compared it with conventional dose-rate irradiation (CONV-IR). The results showed that FLASH-IR had a radioprotective effect, causing less acute intestinal damage compared to CONV-IR. Inflammatory responses and redox status also differed between the two groups.

Summary: This study found that ionizing radiation can promote PD-L1 expression in macrophages, and ATM kinase inhibitor further enhances this effect. ATM inhibition leads to DNA damage, which activates the STING/IRF3 signaling pathway, upregulates type I interferon expression in macrophages, and ultimately leads to PD-L1 upregulation. ATM inhibition also increases CD8(+) T cell infiltration induced by IR and promotes anti-tumor efficacy in vivo.

Summary: In this study, a modified fully convolutional neural network called OrganNet was used for auto-segmentation of 24 organs at risk (OARs) in the head and neck. The model achieved good segmentation results, with an average Dice similarity coefficient of 83.75%. Specifically, it performed well in both large-volume organs and small-volume organs. Compared to manual-delineation, OrganNet showed comparable or better performance in organ segmentation based on the STAPLE contours.

Summary: This study compared the effectiveness of afatinib and osimertinib in patients with non-small cell lung cancer (NSCLC) with uncommon EGFR mutations. The results showed that afatinib had a better PFS benefit compared to osimertinib, but there was no significant difference in ORR between the two. For patients with non-ex20ins single uncommon EGFR mutations, the treatment outcomes were similar for afatinib and osimertinib.

Summary: In this study, the researchers found that IGF2BP3 acts as a key m6A reader in nasopharyngeal carcinoma (NPC) and maintains the stability of NOTCH3 mRNA through an m6A-dependent mechanism, thereby promoting tumor metastasis and enhancing the stemness phenotype. IGF2BP3 is associated with poor prognosis and may serve as a novel prognostic biomarker and therapeutic target in NPC metastasis.
