Anti-diabetic activity evaluation of a polysaccharide extracted from Gynostemma pentaphyllum

In current study, a polysaccharide (GPP) was successfully extracted from Gynostemma pentaphyllum herb. Monosaccharide composition of GPP was rhamnose, arabinose, galactose, glucose, xylose, mannose, galacturonic acid and glucuronic acid in a molar ratio of 4.11: 734: 1331: 20.99:1.07: 0.91: 4.75: 036. Molecular weight and polydispersity (Mw/Mn) of GPP were 4.070 x 10(4) Da and 1.037, respectively. Primary structure features of GPP were determined to be a polysaccharide by FT-IR and NMR. Fasting blood sugar of diabetic mice decreased from 17.56 mmol/L to 7.42 mmol/L by orally administration of 0.5 mL GPP (1 mg/mL) for 30 days. GPP exhibited a dose-dependent inhibition effect on alpha-glucosidase activity. Moreover, GPP could inhibit the glucose absorption and affect the protein expression of GLUT2, but not the protein expression of SGLT1. These results indicated GPP could be used as an effective ingredient to prevent and cure diabetes. (C) 2018 Published by Elsevier B.V.

Summary: Biologically active gellan oligosaccharides (GOSs) are newly discovered plant elicitors and biostimulants produced from the hydrolysis of gellan gum. Traditional hydrolysis methods suffer from pollution and low functional oligosaccharide yield. In this study, irradiation was used to efficiently degrade gellan gum into low molecular weight gellan, which was further hydrolyzed to produce GOSs with antioxidant and prebiotic activities.

Exopolysaccharide synthesis repressor genes (exoR and exoX) related to curdlan biosynthesis by Agrobacterium sp

Summary: This study explored the roles of exoR and exoX genes in curdlan biosynthesis in Agrobacterium sp. ATCC 31749. The results showed that the curdlan yields and glucose utilization increased in delta exoR and delta exoX strains compared with the wild-type strain during batch fermentation. However, the curdlan yields and gel strength decreased in exoR and exoX overexpression strains.

The CD44FM nanozymes replaced unstable HRP and H2O2, and exhibited remarkable oxidase-like activities. By bioconjugating CD44 mAbs, the CD44FM nanozymes selectively entered MDA-MB-231 cells and catalyzed the oxidation of chromogenic substrate TMB for specific detection.

In vitro antioxidant analysis of flavonoids extracted from Artemisia argyi stem and their anti-inflammatory activity in lipopolysaccharide-stimulated RAW 264.7 macrophages

Summary: In this study, flavonoids were successfully extracted from Artemisia argyi stem, with a yield of 15.3 mg/g dry A. argyi stem. The flavonoid extract had a purity of 88.58% (w/w) and contained carbohydrates, proteins, and polyphenols. It demonstrated concentration-dependent scavenging effects on various radicals and exerted protective effects on stimulated macrophages. This research provides guidance for utilizing A. argyi as a plant-based diet and its flavonoid extract as antioxidant and anti-inflammatory ingredients to enhance the function and nutrition of foods.

Effect of ultrasonic degradation on the structural feature, physicochemical property and bioactivity of plant and microbial polysaccharides: A review

Summary: With their bioactivities including antioxidant, anti-bacterial, anti-inflammatory, immune regulation, antitumor and anti-coagulation properties, plant and microbial polysaccharides are widely used in the fields of food, medicine and cosmetics. However, the relationship between structure features and physicochemical properties as well as bioactivities of these polysaccharides remains unclear. Ultrasonic degradation, through mechanical bond breaking and cavitation effects, has the potential to modify the chemical or spatial structures of plant and microbial polysaccharides, thus affecting their physicochemical properties and bioactivities. This review provides an overview of the influence of ultrasonic degradation on the structure features, physicochemical properties and bioactivities of plant and microbial polysaccharides, and highlights the need for attention in the application of ultrasonication for polysaccharide degradation.

Antioxidant and antibacterial activities of a polysaccharide produced by Chaetomium globosum CGMCC 6882

Summary: In this study, a polysaccharide (GCP-TS) derived from tapioca starch by the endophytic fungus Chaetomium globosum CGMCC 6882 in Gynostemma pentaphyllum herb was analyzed. GCP-TS exhibited concentration-dependent scavenging ability against various radicals and enhanced antibacterial activity against Staphylococcus aureus and Escherichia coli with increasing concentrations.

A Rehmannia glutinosa caffeic acid O-methyltransferase functional identification: Reconstitution of the ferulic acid biosynthetic pathway in Saccharomyces cerevisiae using Rehmannia glutinosa enzymes

Summary: This study reconstructed the ferulic acid (FA) biosynthetic pathway from Rehmannia glutinosa in Saccharomyces cerevisiae and identified the functional activity of the RgCOMT enzyme involved in FA production. It was found that several other genes in R. glutinosa also participate in the FA biosynthetic pathway. By introducing these genes into yeast, the entire biosynthesis process from phenylalanine metabolism to FA was successfully achieved in vitro, resulting in an engineered strain with an FA titer of 148.34 mg/L.

Promoting substrates uptake and curdlan synthesis of Agrobacterium sp. by attenuating the exopolysaccharide encapsulation

Summary: During curdlan production, the encapsulation of Agrobacterium sp. by secreted exopolysaccharide (EPS) inhibits substrate uptake and curdlan synthesis. This study found that the addition of endo-ss-1,3-glucanase (BGN) effectively alleviated the EPS encapsulation effect and improved glucose consumption and curdlan yield. The upregulation of related genes and enhanced metabolic activity contributed to the increased production of curdlan.

Regulation strategy, bioactivity, and physical property of plant and microbial polysaccharides based on molecular weight

Summary: Structural features play a crucial role in the bioactivity, physical properties, and applications of plant and microbial polysaccharides. However, the complex structure-function relationship hinders the production, preparation, and utilization of these polysaccharides. Molecular weight, as a easily regulated structural feature, significantly impacts the bioactivity and physical properties of plant and microbial polysaccharides, and polysaccharides with specific molecular weights are essential for their optimal function. This review provides an overview of the regulation strategies for controlling molecular weight, including metabolic regulation, degradation by physical, chemical, and enzymatic methods, and explores the influence of molecular weight on the bioactivity and physical properties of plant and microbial polysaccharides. Furthermore, the study highlights the need for attention to potential problems and offers suggestions for the analysis of polysaccharide molecular weights. The findings of this work will contribute to the production, preparation, utilization, and investigation of the structure-function relationship of plant and microbial polysaccharides based on their molecular weights.

Antibacterial activity of a polysaccharide isolated from Artemisia argyi leaf against Staphylococcus aureus and mechanism investigation

Effects of aging and diabetes on the deformation mechanisms and molecular structural characteristics of collagen fibrils under daily activity

Summary: Crosslinking affects the mechanical behavior and tissue metabolism of collagen-based tissues. Aging and diabetes alter the crosslinking type and density, thus changing tissue properties. High-connectivity enzymatic crosslinking and advanced-glycation end-products result in uniform deformation under daily activity, while low-connectivity enzymatic crosslinking does not. High-connectivity model displays more sliding, while AGEs induce instability in structures near the binding sites.

An eco-friendly composite hydrogel based on covalently crosslinked cellulose/poly (glycerol citrate) for thallium (І) removal from aqueous solutions

Summary: In this study, a reusable biobased composite hydrogel was successfully synthesized and used for the removal of Tl(I) from aqueous solutions. The hydrogel exhibited high adsorption capacity and thermal stability, and the adsorption process was found to be thermodynamically spontaneous. The results demonstrated that the hydrogel can be reused multiple times without significant loss of its adsorption capacity.

The characteristics and optimal conditions of the adsorbent were assessed and analyzed.

Preparation and characterization of hemicellulose films reinforced with amino polyhedral oligomeric silsesquioxane for biodegradable packaging

Summary: In this study, CMH-based CPF films were prepared and their thermal and barrier properties were successfully regulated. The tensile strength and water vapor barrier property of CPF films were significantly improved, making them promising for food packaging applications.
