Summary: B7-H3 is highly expressed in various types of cancer and plays important roles in tumor immune evasion, carcinogenesis, metastasis, and angiogenesis. This study investigated the effects of cancer cell-derived exosomal B7-H3 on tumor angiogenesis and metastasis. The findings showed that exosomal B7-H3 from colorectal cancer cells was taken up by endothelial cells and activated the AKT1/mTOR/VEGFA signaling pathway, enhancing their migration, invasion, and tube formation abilities. Furthermore, exosomes with reinforced B7-H3 promoted pulmonary angiogenesis and metastasis in mice. High expression of B7-H3 was also observed in urinary exosomes from colorectal cancer patients.

Summary: MicroRNAs (miRNAs) play important roles in regulating posttranscriptional gene and protein expression in cancer. In this study, the researchers investigated the expression of VEGFA, a key protein involved in vascular endothelial growth, in hepatocellular carcinoma (HCC) cell lines after transfection with miR-612, miR-637, or miR-874. The results showed that miR-612 and miR-637 downregulated the gene and protein expression of VEGFA in both HCC cell lines. However, the modulation of VEGFA by miR-874 was observed only at the gene expression level and further investigation is needed.

Summary: The study reveals that WTAP is significantly upregulated in CRC cells and patients with CRC, and higher WTAP expression is consistent with the TCGA and CPATC databases in CRC tissues. WTAP over-expression exacerbates cell proliferation, migration, invasion, and angiogenesis, while WTAP knockdown inhibits the malignant biological behavior of CRC cells. Mechanistically, WTAP positively regulates VEGFA expression and YTHDC1 is identified as a downstream effector of the YTHDC1-VEGFA axis in CRC. Furthermore, increased WTAP expression activates the MAPK signaling pathway, which further enhances angiogenesis. The findings suggest that the WTAP/YTHDC1/VEGFA axis promotes CRC development, particularly angiogenesis, and may serve as a potential biomarker of CRC.
