Furanosyl Oxocarbenium Ion Conformational Energy Landscape Maps as a Tool to Study the Glycosylation Stereoselectivity of 2-Azidofuranoses, 2-Fluorofuranoses and Methyl Furanosyl Uronates

The 3D shape of glycosyl oxocarbenium ions determines their stability and reactivity and the stereochemical course of S(N)1 reactions taking place on these reactive intermediates is dictated by the conformation of these species. The nature and configuration of functional groups on the carbohydrate ring affect the stability of glycosyl oxocarbenium ions and control the overall shape of the cations. We herein map the stereoelectronic substituent effects of the C2-azide, C2-fluoride and C4-carboxylic acid ester on the stability and reactivity of the complete suite of diastereoisomeric furanoses by using a combined computational and experimental approach. Surprisingly, all furanosyl donors studied react in a highly stereoselective manner to provide the 1,2-cis products, except for the reactions in the xylose series. The 1,2-cis selectivity for the ribo-, arabino- and lyxo-configured furanosides can be traced back to the lowest-energy E-3 or E-3 conformers of the intermediate oxocarbenium ions. The lack of selectivity for the xylosyl donors is related to the occurrence of oxocarbenium ions adopting other conformations.

Synthetic Zwitterionic Streptococcus pneumoniae Type 1 Oligosaccharides Carrying Labile O-Acetyl Esters

Summary: We report the first total synthesis of the Streptococcus pneumoniae serotype 1 (Sp1) oligosaccharide, which is a zwitterionic capsular polysaccharide carrying unstable O-acetyl esters. The synthesis was achieved by assembling trisaccharide building blocks in a highly stereoselective manner, up to the 9-mer level. The structure investigation revealed that the oligomers adopt a right-handed helical structure, with the acetyl esters exposed on the periphery and in close proximity of the neighboring AAT residues.

Origin of Stereoselectivity in SE2′ Reactions of Six-membered Ring Oxocarbenium Ions

Mapping the effect of configuration and protecting group pattern on glycosyl acceptor reactivity

Summary: By systematically surveying 67 acceptor alcohols in glycosylation reactions, this study reveals how the reactivity of a carbohydrate acceptor depends on its configuration and substitution pattern, which greatly influences the yield and stereoselectivity of the reaction. The study demonstrates the influence of functional groups flanking the acceptor alcohol on its reactivity, highlighting the importance of both the nature and relative orientation. The empirical acceptor reactivity guidelines obtained will assist in the rational optimization of glycosylation reactions and serve as an important tool in oligosaccharide assembly.

Stereoselective Synthesis of the O-antigen of A. baumannii ATCC 17961 Using Long-Range Levulinoyl Group Participation

Summary: In this study, we successfully synthesized the pentasaccharide and decasaccharide of A. baumannii ATCC 17961 O-antigen for the development of a synthetic carbohydrate-based vaccine against A. baumannii infection. We employed an organocatalytic glycosylation method to efficiently synthesize the rare sugar 2,3-diacetamido-glucuronate. We also discovered the improved beta-selectivity in glycosylations by the long-range levulinoyl group participation via a hydrogen bond, which solved the stereoselectivity problem of highly branched galactose acceptors. The proposed mechanism was supported by control experiments and DFT computations.

S(N)2 versus E2 Competition of Cyclic Ethers

Summary: The influence of ring strain on the competition between the S(N)2 and E2 pathways has been studied using quantum chemical methods. It was found that increasing ring strain decreases the activation energy of the S(N)2 pathway but generally increases the activation energy of the E2 pathway. This results in a mechanistic switch in the preferred reaction pathway for strong Lewis bases, favoring S(N)2 for small cyclic substrates and E2 for large cyclic substrates.

In this study, different chemistries were explored to construct cyclitol aziridines with the aziridine electrophile attached in an exocyclic fashion, closer to an inverting glycosidase active site nucleophile. These newly synthesized cyclitol aziridines can be evaluated as inhibitors or inactivators of both alpha- and beta-glucosidases.

Summary: Here, a unique organo-catalysis relay glycosylation method is described for the regio- and stereoselective synthesis of 2-amino-2-deoxy-dithioglycosides. This method exhibits excellent site- and stereoselectivity, good to excellent yields, mild reaction conditions, and broad substrate scope. The dithiolated O-antigen of E. coli serogroup 64 was successfully synthesized using this newly developed method.

Re-Exploring the Anthracycline Chemical Space for Better Anti-Cancer Compounds

Summary: Anthracycline anti-cancer drugs are widely used in the clinic to treat various cancers. They induce DNA double strand breaks and chromatin damage, which contribute to their reported side effects. Despite advancements in understanding the structure-activity relationship of anthracyclines, there are still many unexplored structural variations. This study prepared a diverse set of anthracyclines with variations in the sugar moiety, amine alkylation pattern, saccharide chain, and aglycone, and evaluated their cytotoxicity, DNA- and chromatin damage-inducing capacity in vitro. This work provides guidelines for anthracycline design and identifies novel and highly potent anthracyclines that may have improved patient tolerance.
