In type 1 diabetes, a renewable source of human pancreatic beta cells, in particular from human induced pluripotent stem cell (hiPSC) origin, would greatly benefit cell therapy. Earlier work showed that pancreatic progenitors differentiated from human embryonic stem cells in vitro can further mature to become glucose responsive following macroencapsulation and transplantation in mice. Here we took a similar approach optimizing the generation of pancreatic progenitors from hiPSCs. This work demonstrates that hiPSCs differentiated to pancreatic endoderm in vitro can be efficiently and robustly generated under large-scale conditions. The hiPSC-derived pancreatic endoderm cells (HiPECs) can further differentiate into glucose-responsive islet-like cells following macroencapsulation and in vivo implantation. The HiPECs can protect mice from streptozotocin-induced hyperglycemia and maintain normal glucose homeostasis and equilibrated plasma glucose concentrations at levels similar to the human set point. These results further validate the potential use of hiPSC-derived islet cells for application in clinical settings.

Mitochondrial ion channels in pancreatic β-cells: Novel pharmacological targets for the treatment of Type 2 diabetes

Umberto De Marchi, Silvia Fernandez-Martinez, Sergio de la Fuente, Andreas Wiederkehr, Jaime Santo-Domingo

Summary: Pancreatic beta-cells play a central role in regulating glucose homeostasis, with mitochondria playing a crucial role in linking nutrient metabolism to insulin secretion. Mitochondria, as double-membrane organelles with various channels, regulate energy production, signaling, and cell death within beta-cells.

The mitochondrial calcium uniporter (MCU) activates mitochondrial respiration and enhances mobility by regulating mitochondrial redox state

Anna Weiser, Aurelle Hermant, Flavien Bermont, Federico Sizzano, Sonia Karaz, Pilar Alvarez-Illera, Jaime Santo-Domingo, Vincenzo Sorrentino, Jerome N. Feige, Umberto De Marchi

Summary: Regulation of mitochondrial redox balance is crucial for cell signaling, but the connection between the redox state and cell conditions is not well understood. The study found that activation of the MCU protein modulates mitochondrial redox state, leading to mitochondrial reduction and improved respiratory capacity and mobility. By bypassing MCU, similar benefits can be achieved by directly reducing mitochondrial proteins. This research demonstrates the importance of MCU in regulating mitochondrial redox balance and its effects on respiratory and mobility functions.
