Initial findings of striatum tripartite model in OCD brain samples based on transcriptome analysis

Obsessive-compulsive disorder (OCD) is a psychiatric disorder characterized by obsessions and/or compulsions. Different striatal subregions belonging to the cortico-striato-thalamic circuitry (CSTC) play an important role in the pathophysiology of OCD. The transcriptomes of 3 separate striatal areas (putamen (PT), caudate nucleus (CN) and accumbens nucleus (NAC)) from postmortem brain tissue were compared between 6 OCD and 8 control cases. In addition to network connectivity deregulation, different biological processes are specific to each striatum region according to the tripartite model of the striatum and contribute in various ways to OCD pathophysiology. Specifically, regulation of neurotransmitter levels and presynaptic processes involved in chemical synaptic transmission were shared between NAC and PT. The Gene Ontology terms cellular response to chemical stimulus, response to external stimulus, response to organic substance, regulation of synaptic plasticity, and modulation of synaptic transmission were shared between CN and PT. Most genes harboring common and/or rare variants previously associated with OCD that were differentially expressed or part of a least preserved coexpression module in our study also suggest striatum subregion specificity. At the transcriptional level, our study supports differences in the 3 circuit CSTC model associated with OCD.

Nutrition and bipolar disorder: a systematic review

Summary: This review aims to summarize the relationship between nutrition and bipolar disorder (BD). Findings suggest that the intake of unsaturated fatty acids, seafood, folic acid, and zinc is associated with improved BD symptoms. The impacts of creatine, carnitine, vitamin D, inositol, or NAC supplementation on BD are variable and mainly non-significant. Promising results are associated with Coenzyme Q10 (Coq10) and probiotics. Overall, dietetic approaches might be included as part of BD treatment, and individuals with BD should choose healthy dietary lifestyles.

Summary: Maternal prenatal psychosocial stress is associated with altered hypothalamic-pituitary-adrenal axis (HPAA) function among infants. This study found that altered DNA methylation, specifically in the oxytocin receptor gene, may contribute to the regulation of HPAA during infancy. Maternal distress and anxiety symptoms were negatively associated with DNA methylation and cortisol levels in infants.

Adaptive Significance Levels in Tests for Linear Regression Models: The e-Value and P-Value Cases

Summary: The full Bayesian significance test (FBST) is a Bayesian alternative to traditional significance tests based on p-values, characterized by the e-value as evidence in favor of the null hypothesis (H). This work presents a method to determine a cutoff value for the e-value in the FBST by minimizing the linear combination of averaged type-I and type-II error probabilities. The methodology is compared with the test with adaptive significance level, using linear regression models under the Bayesian approach.

The longitudinal trajectory of emotional cognition in subgroups of recently diagnosed patients with bipolar disorder

Summary: In this study, the trajectories of emotional cognition subgroups in recently diagnosed bipolar disorder (BD) patients were assessed over a 16-month follow-up period. Two distinct emotional cognition subgroups were identified, with one subgroup showing heightened negative emotional reactivity and faster recognition of emotional facial expressions, while the other subgroup showed blunted reactivity in positive social scenarios, impaired emotion regulation, and poorer recognition of facial expressions. Longitudinal analysis revealed that the first subgroup exhibited improvement in emotional cognition abnormalities, while the second subgroup showed persistent impairments and poorer course of illness, indicating a need for earlier and more targeted therapeutic interventions.

Summary: Network Medicine is a useful platform for studying the molecular complexity of complex diseases and identifying disease modules and pathways. It can provide insights into how environmental chemical exposures affect human cells and help monitor and prevent exposure-related diseases. In this study, benzene and malathion-exposed differentially expressed genes were used to construct interaction networks and identify important hub genes associated with these chemicals.

Summary: The results of this systematic review and meta-analysis show that the risk of premature death in patients with bipolar disorder is not only related to suicide and unnatural causes, but also to physical comorbidities. Therefore, in addition to suicide prevention, promoting physical health and preventing physical conditions in individuals with bipolar disorder may reduce premature mortality risk.

Self-regulation in youth with bipolar disorder

Summary: This study examines the components of self-regulation in pediatric bipolar disorder (PBD) by studying the relationship between executive functions, emotion processing, and family environmental factors. The results show that adolescents with PBD demonstrated significant deficits in all three areas compared to the control group. Emotion processing was negatively correlated with inhibition and attention, and positively correlated with mental flexibility/working memory. Family environmental factors were negatively correlated with mental flexibility/working memory and emotion processing, and positively correlated with attention and inhibition.

Summary: This study aimed to cross-culturally adapt and assess the content validity evidence of the Cognitive Symptom Checklist-Work-21 in the Brazilian context. The final version, named "Lista de verificacao de sintomas cognitivos relacionados ao trabalho -22 itens", showed strong evidence of content validity and good linguistic equivalence in the Brazilian context.
