Summary: The aim of this study was to identify an efficacious, novel therapeutic biomarker for the successful diagnosis of lung cancer at the early stage of the disease and the molecular insights involved. The expression levels of HDAC2 and YY1 were found to be upregulated in lung cancer cell lines and tumor samples. Inhibition of HDAC2 reduced YY1 expression, and knockdown of YY1 inhibited proliferation, migration, invasion, and cell cycle progression in lung cancer cell lines by suppressing c-Myc. These findings suggest that HDAC2's anticancer role may be attributed to YY1 regulation and it could serve as a potential novel candidate biomarker for lung cancer diagnosis.

Mechanistically, miR-669a-5p targets G3BPs to inhibit HDAC6 expression, which further upregulates AKAP12 expression, ultimately leading to greater cilium length.
