Two pairs of new flavans, moracinflavan A and B (1a/1b and 2a/2b), together with another five new flavan derivatives moracinflavan C-G (3-7), and three known flavans (8-10) were isolated from the leaves of Morus alba. Their structures were elucidated by detailed analyses based on spectroscopic data. Separation of the stereoisomeric mixtures of 1a and 1b, 2a and 2b was achieved using chiral HPLC, and the absolute configurations of the respective stereoisomers were determined based on comparison of experimental and calculated electronic circular dichroism (ECD) spectra. Using an ARE-dependent luciferase reporter assay, we identified that these flavans were capable of increasing ARE-luciferase activities. This indicated that these flavans are potential Nrf2 activators.

Matrix-Assisted DOSY for Analysis of Indole Alkaloid Mixtures

Yu-Lin You, Fei-Fei Li, Ning Wang, Shu-Qi Wang

Summary: DOSY is a powerful tool for investigating mixtures, but similar diffusion coefficients of components can limit resolution. This study used matrix-assisted DOSY to explore diffusion resolution in complex mixtures and found that using SDS micelles as the matrix can improve resolution. Increasing SDS concentration can also enhance resolution of DOSY spectrum.

Reconstitution of Caveolin-1 into Artificial Lipid Membrane: Characterization by Transmission Electron Microscopy and Solid-State Nuclear Magnetic Resonance

Yanli Zhang, Xinyan Zhang, Wenru Kong, Shuqi Wang

Summary: Researchers developed a highly reliable and reproducible method for the expression and purification of CAV1, demonstrating a caveolae-like membrane curvature in artificial lipid membranes. Solid-state nuclear magnetic resonance (ssNMR) structural characterization revealed that CAV1 is primarily alpha-helical in structure, with its scaffolding domain adopting an alpha-helical structure.

Biochemometric approach combined with 1D CSSF-TOCSY for the identification of sensitization agents in Curcuma longa L. and prediction of their action mechanisms

Fei-Fei Li, Yan-Li Zhang, Dong-Xiao Guo, Chang-Jun Zhao, Yun-Feng Yao, Yong-Qiang Lin, Shu-Qi Wang

Summary: This study demonstrated the sensitization effects of Curcuma longa L. on 5-FU resistant colorectal cancer cells and established a method to identify potential sensitization agents and predict their action mechanisms before obtaining the agents. The results indicated that curcumin, demethoxycurcumin, and bisdemethoxycurcumin were responsible for the effects, and two possible mechanisms, reversal of drug resistance and disruption in lactate excretion, were suggested. The compounds significantly enhanced the sensitivity of the cancer cells to 5-FU treatment, which was supported by the downregulation of drug resistance-related transporters and lactate membrane transporter.

Epimedokoreanin C, a prenylated flavonoid isolated from Epimedium koreanum, induces non-apoptotic cell death with the characteristics of methuosis in lung cancer cells

Xiaoqing Liu, Shuqi Wang, Hao Zheng, Qingying Liu, Tao Shen, Xiaoning Wang, Dongmei Ren

Summary: Methuosis is a novel type of non-apoptotic cell death characterized by cytoplasmic vacuolization. This study demonstrated that EKC, a prenylated flavonoid from Epimedium koreanum, induces methuosis-like cell death in lung cancer cells and synergizes with conventional chemotherapy drugs. The regulation of Rac1 and Arf6 appears to be at least partially involved in EKC-induced methuosis.
