Neurologic patients have an increased risk for bullous pemphigoid (BP), in which autoantibodies target BP180, a cutaneous basement membrane protein also expressed in the brain. Here we show that 53.6% of sera from patients with multiple sclerosis (MS) (n = 56) had IgG reactivity against full-length BP180 in immunoblotting, while in BP180 non-collagenous 16A ELISA (n = 143), only 7.7% of MS samples studied were positive. Epitope mapping with 13 fusion proteins covering the entire BP180 polypeptide revealed that in MS and Alzheimer's disease (AD) patients, IgG autoantibodies target regions located in the intracellular and mid-extracellular parts of BP180, but not the well-known BP epitopes located in the non-collagenous 16A domain and the distal part of extracellular domain. In indirect immunofluorescence analysis, 8.1% of MS sera recognized the cutaneous basement membrane and in full-length BP180 ELISA analysis, 7.5% MS and AD sera were positive, indicating that these autoantibodies rarely recognize BP180 in its native conformation. Thus, in MS and AD patients, BP180 autoantibodies have a different epitope profile than in patients with BP, and seldom bind to native BP180. This explains the inability of these autoantibodies to cause skin symptoms. Our results suggest that the autoantibodies against BP180 alone are not sufficient to induce BP in MS and AD patients.

Effectiveness of interventions used to prepare preschool children and their parents for day surgery: A systematic review and meta-analysis of randomised controlled trials

Heli Kerimaa, Heidi Ruotsalainen, Helvi Kyngas, Jouko Miettunen, Tarja Polkki

Summary: This systematic review found that certain preparatory interventions can reduce parental anxiety and stress, and alleviate children's fear and pain. However, more randomized controlled trials are needed to determine the most effective methods for preparing parents and children for day surgery.

Cross-cultural comparison of depressive symptoms on the Beck Depression Inventory-II, across six population samples

Marjo Seppanen, Tiina Lankila, Juha Auvinen, Jouko Miettunen, Raija Korpelainen, Markku Timonen

Summary: This study examined the cultural differences of Beck Depression Inventory (BDI-II) symptoms among different countries. The results showed significant differences in symptom scores between countries, highlighting the importance of considering cultural differences when interpreting BDI-II item scores.

Association of ADHD symptoms in adolescence and mortality in Northern Finland Birth Cohort 1986

Antti Mustonen, Anni-Emilia Alakokkare, James G. Scott, Anu-Helmi Halt, Miika Vuori, Tuula Hurtig, Alina Rodriguez, Jouko Miettunen, Solja Niemela

Summary: This study investigated the association between symptoms of attention deficit hyperactive disorder (ADHD) in adolescents and risk of mortality. The results showed that higher scores in inattentive, hyperactive, and combined symptoms were associated with increased risk of death. However, this association was partially attenuated by adolescent substance use.

Summary: There is a familial association between parental severe mental illness (SMI) and obesity in middle-aged offspring, especially in the offspring of mothers with SMI.

Absence of NC14A Domain of COLXVII/BP180 in Mice Results in IL-17-Associated Skin Inflammation

Outi Lindgren, Gwenaelle Le Menn, Jussi Tuusa, Zhi Jane Chen, Kaisa Tasanen, Nina Kokkonen

Summary: Deletion of exon 18 from Col17a1 in transgenic ANC14A mice leads to the absence of the NC14A domain, which corresponds to the immunodominant epitope in bullous pemphigoid. Before the age of 1 year, 84% of ANC14A mice develop severe itch and skin erosion. Further characterization reveals acanthosis, subepidermal blistering, and inflammatory cell infiltrates in the lesional skin, along with linear complement C3, IgG, and/or IgA deposition. Increased expression of IL-17-associated cytokines and changes in immune cell proportions are observed. Treatment with anti-IL-17A decreases the expression of certain cytokines in the nonlesional skin.

Dipeptidyl Peptidase 4 Inhibitor-Associated Bullous Pemphigoid Is Characterized by an Altered Expression of Cytokines in the Skin

Jussi Tuusa, Nina Kokkonen, Anja Mattila, Laura Huilaja, Outi Varpuluoma, Sirpa Rannikko, Virpi Glumoff, Jouko Miettunen, Kaisa Tasanen

Summary: Dipeptidyl peptidase 4 inhibitors (DPP4is), commonly used drugs for type 2 diabetes, increase the risk of bullous pemphigoid (BP). The mechanism leading to the loss of immunological tolerance and the differences between DPP4i-associated BP (DPP4i-BP) and regular BP are largely unknown. This study analyzed cytokine expression in lesional and nonlesional skin samples of patients with regular BP or DPP4i-BP, and found distinct molecular fingerprints between the two.

Employment trajectories until midlife in schizophrenia and other psychoses: the Northern Finland Birth Cohort 1966

Tuomas Majuri, Anni-Emilia Alakokkare, Marianne Haapea, Tanja Nordstrom, Jouko Miettunen, Erika Jaaskelainen, Leena Ala-Mursula

Summary: This study compared employment trajectories among individuals with schizophrenia, other psychoses, and without psychosis. The findings showed that most individuals with mental illnesses had unstable employment trajectories, indicating a higher risk of unemployment and part-time work. The study highlights the importance of improving labor market attachment during the early stages of mental illness.

Economic preferences and temperament traits among business leaders and paid employees

Mikko Vaaramo, Leena Ala-Mursula, Jouko Miettunen, Marko Korhonen

Summary: This study examines the differences in economic preferences and temperament traits between different categories of business leaders (i.e., managers, self-employed with employees, self-employed without employees) and paid employees. The results show that business leaders generally have different economic preferences and temperament traits compared to paid employees. The temperament trait of novelty-seeking seems relevant for becoming a manager, while the economic preference of risk-taking appears important for becoming self-employed.

Summary: This population-based birth cohort study examines the associations between self-reported temperament at age 31 and self-reported leisure-time physical activity (MVPA) levels and changes from age 31 to age 46. The results suggest that temperament may play a role in determining the level and sustainability of MVPA.

Summary: In the Open Dialogue (OD) based psychiatric services, adolescent patients receive less medication and are more likely to be treated as outpatients compared to standard services. This study aimed to evaluate the association between treatment under OD and the probability of suicide compared to standard psychiatric care. The results show no statistically significant differences in 5-year suicide hazard ratios between the OD group and the comparison group.
