BackgroundRING finger protein 38 (RNF38), a member of the RNF protein family, has just emerged as a vital driver of cancer progression. However, the oncogenic mechanisms of RNF38 remain unexplored.MethodsUsing frozen tumor tissue and tissue microarray from hepatocellular carcinoma (HCC) patients, we tried to probe the expression of RNF38 in HCC and its clinical value. Then the biological functions of RNF38 were analyzed in vivo and vitro. Stable isotope labeling with amino acids (SILAC) in cell culture and co-immunoprecipitation proteomic analyses were combined to reveal the potential mechanism of RNF38 in HCC progression.ResultsWe report that RNF38 expression was markedly higher in HCC tissues than in peritumor tissues. Correspondingly, RNF38 overexpression promoted the HCC cell migration and invasion and inhibited apoptosis both in vitro and in vivo. And elevated RNF38 expression induced HCC cell epithelial-mesenchymal transition by facilitating transforming growth factor- (TGF-) signaling via ubiquitinating and degrading neuroblast differentiation-associated protein (AHNAK), a well-established inhibitor of TGF- signaling. Furthermore, AHNAK interference restored the HCC cell invasion and metastasis deprived by RNF38 downregulation. Clinically, elevated RNF38 and transforming growth factor beta receptor 1 (TGFBR1) expression was related to short overall survival (OS) and high cumulative recurrence rates in HCC patients.ConclusionsHigh levels of RNF38 promote HCC by facilitating TGF- signaling and are a novel marker for predicting the prognosis of HCC patients and a potential therapeutic target in HCC.

Genetic diversity and structure of a core collection of Huangqi (Astragalus ssp.) developed using genomic simple sequence repeat markers

Fanshu Gong, Yaping Geng, Pengfei Zhang, Feng Zhang, Xinfeng Fan, Yaling Liu

Summary: In this study, a core collection of Astragalus germplasm resources was established using 10 SSR markers. Among different methods, the CoreFinder-based M strategy was found to be the most suitable approach. The genetic diversity and population structure of the core collection were similar to the initial collection. The construction of a core collection is beneficial for the understanding and utilization of Astragalus, and serves as a valuable reference for constructing core collections of other plants and fungi.

Decoupling effects of driving factors on sediment yield in the Chinese Loess Plateau

Xiaojing Tian, Guangju Zhao, Xingmin Mu, Pengfei Zhang, Peng Gao, Wenyi Sun, Xiaoyan Lu, Peng Tian

Summary: This study comprehensively investigated the spatiotemporal variations of sediment yield in the Loess Plateau and analyzed its responses to changes of precipitation, land use/cover, and vegetation cover. The results showed a significant reduction in sediment yield during 1961-2016, with precipitation, land-use change, vegetation dynamics, and runoff reduction explaining 77.9% of this variation. The study found that runoff had the most direct effect on sediment yield, followed by vegetation cover and land-use change, while the correlation between sediment yield and precipitation indices was relatively low. The findings provide valuable insights for soil and water conservation planning and sediment load management in the Yellow River Basin.

A Generalized Deep Learning Clustering Algorithm Based on Non-Negative Matrix Factorization

Dexian Wang, Tianrui Li, Ping Deng, Fan Zhang, Wei Huang, Pengfei Zhang, Jia Liu

Summary: In this article, a Generalized Deep Learning Clustering (GDLC) algorithm based on Non-Negative Matrix Factorization (NMF) is proposed to address the slow convergence and low clustering accuracy in the update process of NMF. A nonlinear constrained NMF (NNMF) algorithm is constructed to achieve sequential updates of the matrix elements guided by the learning rate, and the GDLC algorithm is constructed by transforming gradient values into generalized weights and biases through a nonlinear activation function. The experimental results on eight datasets demonstrate the efficient performance of the GDLC algorithm.

Genetic gain and inbreeding from simulation of different genomic mating schemes for pig improvement

Fuping Zhao, Pengfei Zhang, Xiaoqing Wang, Deniz Akdemir, Dorian Garrick, Jun He, Lixian Wang

Summary: Genomic mating (GM) is an effective approach to optimize pairing combinations after genomic selection, achieving sustainable genetic progress and controlling rates of inbreeding accumulation in a population. Our study investigated the impact of various factors on the efficiency of GM using stochastic simulation. Results showed that GM with runs of homozygosity (ROH)-based genealogical relatedness achieved faster genetic gains and lower rates of inbreeding compared to traditional mating schemes.

A nitric oxide and hydrogen sulfide dual-donating nanosystem for highly synergistic gas-radiotherapy against hepatocellular carcinoma

Junjie Fu, Yong Mao, Jing Han, Pengfei Zhang, Yunying Tan, Jing Hu, Peter H. Seeberger, Jian Yin

Summary: A drug delivery system based on gold-capped mesoporous silica nanoparticles is developed for the concurrent delivery of nitric oxide and hydrogen sulfide into hepatocellular carcinoma cells. The combined gasradiotherapy and X-ray irradiation exhibit highly synergistic anti-cancer activity against both normoxic and hypoxic cancer cells. Further studies show that the combined treatment not only retains the individual anticancer mechanisms of NO, H2S, and X-ray, but also alleviates cancer hypoxia through unique pathways involving NO and H2S. In mice, the treatment significantly inhibits the growth of subcutaneous and orthotopic tumors and shows high biocompatibility. This research promotes the clinical application of combined gasradiotherapy as an effective cancer treatment.

High-Accuracy Quantitative Functional Imaging Technique for Retinal Opto-Physiology with Optical Coherence Tomography

Yanhong Ma, Pengfei Zhang

Summary: Retinal opto-physiology is the physiological response of the retina to a visible light stimulus, reflecting the function of the retina to a certain extent. Optoretinography (ORG) is a newly developed functional imaging technique that uses optical coherence tomography to precisely quantify the opto-physiological response of the retina. By combining adaptive optics and phase analysis techniques, ORG can achieve high resolution and be used to measure retinal opto-physiological functions at the cellular level. Once standards are established, ORG may have broad applications in ophthalmic research and clinical practice.
