Summary: Piezo1 is upregulated in hepatocellular carcinoma (HCC) and is closely associated with clinicopathological features and poor prognosis. Knockdown of Piezo1 inhibits proliferation, migration, invasion, and epithelial-mesenchymal transition (EMT) of HCC cells, as well as tumor growth and metastasis. Piezo1 promotes HCC progression through activating the TGF-beta signaling pathway.

Summary: The study revealed that CYP1A2 is significantly down-regulated in HCC tumor tissues and its high expression is associated with better prognosis. CYP1A2 functions as a tumor suppressor in HCC by inhibiting HGF/MET signaling and MMPs expression, thus reducing cell growth and invasion capabilities.

Summary: NOX4 is necessary for apoptosis induced by TGF-I3 in hepatocytes and HCC cells, particularly in the SMADs signaling pathway. It also plays a role in non-canonical signals and other parallel actions induced by TGF-I3. In addition, NOX4 is required for TGF-I3-induced migratory effects and regulates cytoskeleton dynamics and focal adhesions. However, high expression of TGF-I31-related genes combined with high expression of NOX4 is associated with a worse prognosis in HCC patients.

Summary: The study demonstrated that eIF6 expression was significantly increased in HCC and correlated with its pathological progression. eIF6 served as a new diagnostic biomarker and an independent risk factor for OS in HCC patients. Functional studies showed that deletion of eIF6 exhibited tumor-suppressor activity in HCC cells.

Summary: In this study, it was found that regorafenib treatment activated Wnt/beta-catenin signaling only in hepatoblast-like HCC cell lines and induced enrichment of markers associated with hepatic stem/progenitor cells. On the other hand, regorafenib resistant cells demonstrated diminished Wnt/beta-catenin signaling activity and enhanced TGF-beta signaling activity. The inhibition of TGF-beta pathway in combination with regorafenib administration might increase regorafenib-induced cell death in HCC tumors with aberrant activation in the Wnt/beta-catenin pathway.

Summary: Deregulated Wnt/β-catenin signaling is a major genetic alteration in hepatocellular carcinoma (HCC), and its activation plays an oncogenic role in hepatocarcinogenesis. The activated pathway is associated with specific gene expression patterns and pathological features, and it synergizes with other signaling cascades to promote HCC formation. Therefore, understanding and targeting this pathway has important implications for the diagnosis, classification, and personalized treatment of HCC.

Summary: This study evaluated the antiproliferative and antiangiogenic effects of atorvastatin (AT) in the development of hepatocellular carcinoma (HCC). The results showed that AT inhibits the growth of liver tumors and cell proliferation, as well as the formation of blood vessels. These antitumor effects may be mediated by disrupting multiple hepatocarcinogenic pathways.

Summary: Patients with primary biliary cholangitis (PBC) are at higher risk for developing hepatocellular carcinoma (HCC), especially in the presence of comorbidities like excessive alcohol consumption. This study found that the balance of Smad signals may serve as a biomarker to predict the likelihood of HCC occurrence in PBC patients. Shifting favorably Smad phospho-isoforms through therapies could potentially reduce the risk of PBC-related HCC development.

Summary: The study reveals that DDR1 promotes liver cancer cell proliferation and tumor growth, while downregulation of DDR1 has the opposite effect. DDR1 interacts with SLC1A5 to regulate the mTORC1 signaling pathway. DDR1 serves as an independent prognostic marker, and its expression is positively correlated with SLC1A5 in clinical samples, providing new insights into HCC development and potential targets for treatment.

Summary: In this study, a core collection of Astragalus germplasm resources was established using 10 SSR markers. Among different methods, the CoreFinder-based M strategy was found to be the most suitable approach. The genetic diversity and population structure of the core collection were similar to the initial collection. The construction of a core collection is beneficial for the understanding and utilization of Astragalus, and serves as a valuable reference for constructing core collections of other plants and fungi.

Summary: This study comprehensively investigated the spatiotemporal variations of sediment yield in the Loess Plateau and analyzed its responses to changes of precipitation, land use/cover, and vegetation cover. The results showed a significant reduction in sediment yield during 1961-2016, with precipitation, land-use change, vegetation dynamics, and runoff reduction explaining 77.9% of this variation. The study found that runoff had the most direct effect on sediment yield, followed by vegetation cover and land-use change, while the correlation between sediment yield and precipitation indices was relatively low. The findings provide valuable insights for soil and water conservation planning and sediment load management in the Yellow River Basin.

Summary: In this article, a Generalized Deep Learning Clustering (GDLC) algorithm based on Non-Negative Matrix Factorization (NMF) is proposed to address the slow convergence and low clustering accuracy in the update process of NMF. A nonlinear constrained NMF (NNMF) algorithm is constructed to achieve sequential updates of the matrix elements guided by the learning rate, and the GDLC algorithm is constructed by transforming gradient values into generalized weights and biases through a nonlinear activation function. The experimental results on eight datasets demonstrate the efficient performance of the GDLC algorithm.

Summary: Genomic mating (GM) is an effective approach to optimize pairing combinations after genomic selection, achieving sustainable genetic progress and controlling rates of inbreeding accumulation in a population. Our study investigated the impact of various factors on the efficiency of GM using stochastic simulation. Results showed that GM with runs of homozygosity (ROH)-based genealogical relatedness achieved faster genetic gains and lower rates of inbreeding compared to traditional mating schemes.

Summary: A drug delivery system based on gold-capped mesoporous silica nanoparticles is developed for the concurrent delivery of nitric oxide and hydrogen sulfide into hepatocellular carcinoma cells. The combined gasradiotherapy and X-ray irradiation exhibit highly synergistic anti-cancer activity against both normoxic and hypoxic cancer cells. Further studies show that the combined treatment not only retains the individual anticancer mechanisms of NO, H2S, and X-ray, but also alleviates cancer hypoxia through unique pathways involving NO and H2S. In mice, the treatment significantly inhibits the growth of subcutaneous and orthotopic tumors and shows high biocompatibility. This research promotes the clinical application of combined gasradiotherapy as an effective cancer treatment.

Summary: Retinal opto-physiology is the physiological response of the retina to a visible light stimulus, reflecting the function of the retina to a certain extent. Optoretinography (ORG) is a newly developed functional imaging technique that uses optical coherence tomography to precisely quantify the opto-physiological response of the retina. By combining adaptive optics and phase analysis techniques, ORG can achieve high resolution and be used to measure retinal opto-physiological functions at the cellular level. Once standards are established, ORG may have broad applications in ophthalmic research and clinical practice.
