A polymer-based anti-quorum catheter coating to challenge MDR Staphylococcus aureus: in vivo and in vitro approaches

Background: MDR Staphylococcus aureus is a major aetiological agent of catheter-associated infections. A quorum sensing targeted drug development approach proves to be an effective alternative strategy to combat such infections. Methods: Intravenous catheters were coated with polymethacrylate copolymers loaded with the antivirulent compound 2-[(methylamino) methyl] phenol (2MAMP). The in vitro drug release profile and kinetics were established. The anti-biofilm effect of the coated catheters was tested against clinical isolates of MDR S. aureus. The in vivo studies were carried out using adult male Wistar rats by implanting coated catheters in subcutaneous pockets. Histopathological analysis was done to understand the immunological reactions induced by 2MAMP. Results: A uniform catheter coating of thickness 0.1mm was achieved with linear sustained release of 2MAMP for 6 h. The coating formulation was cytocompatible. The in vitro and in vivo anti-adherence studies showed reduced bacterial accumulation in coated catheters after 48 h. The histopathological results confirmed that the coated catheter did not bring about any adverse inflammatory response. Conclusions: The developed anti-quorum-coated catheter that is non-toxic and biocompatible has the potential to be used in othermedical devices, thereby preventing catheter-associated infections.

The strain contains antibiotic synthesis genes and lipopeptides, confirmed through gene amplification and liquid chromatography. Formulations based on this strain may offer an effective solution for controlling early blight disease in tomato and other crops.

Design, dynamic docking, synthesis, and in vitro validation of a novel DNA gyrase B inhibitor

Summary: Methicillin-resistant Staphylococcus aureus (MRSA) and vancomycin-intermediate-resistant Staphylococcus aureus (VRSA) are high priority pathogens according to the WHO. This study focuses on the urgent need for new classes of anti-MRSA drugs and targets the GyrB subunit of bacterial DNA Gyrase. Through structure-based virtual screening and de novo ligand design, the most promising ligand qsl-304 was identified. In vitro experiments showed qsl-304's activity against the novobiocin resistant clinical isolate, Staphylococcus aureus sa-P2003, and its bacteriostatic nature with reduced resistance recurrence.

Evaluation of lectin nanoscaffold based in-situ gel against vulvovaginal candidiasis causing Candida biofilms using a novel ex-vivo model

Summary: Vulvovaginal Candidiasis (VVC) is a global issue for women of the reproductive age group. This study proposes a novel formulation that undergoes sol-to-gel transition and combines with standard antifungal drugs for VVC treatment. The formulated in-situ gel shows biofilm eradication against C. albicans and C. glabrata, and the researchers also explore the use of goat buccal mucosal membrane as an ex-vivo model for studying vaginal biofilms. The in vitro and ex vivo drug release kinetics demonstrate the effectiveness of the drug-loaded JCuSNPs formulation.

In-vivo pharmacokinetic studies of Dolutegravir loaded spray dried Chitosan nanoparticles as milk admixture for paediatrics infected with HIV

Summary: By using spray drying technology, chitosan nanoparticles were successfully developed to enhance the solubility and bioavailability of dolutegravir (DTG). The nanoparticles showed a significant increase in C-max and improved bioavailability compared to pure DTG. This study demonstrates the potential of chitosan-based nanoparticles as carriers for oral administration of DTG in pediatric HIV patients.

Hidden agenda of Enterococcus faecalis lifestyle transition: planktonic to sessile state

Development of an anti-infective urinary catheter composed of polyvinyl alcohol/sodium alginate/methylcellulose/polyethylene glycol by using a pressure-assisted 3D-printing technique

Polymer based dual drug delivery system for targeted treatment of fluoroquinolone resistant Staphylococcus aureus mediated infections

Summary: This study explores the use of a smart hydrogel delivery system for the treatment of S. aureus-induced soft skin infections. The system combines the antibiotic Ciprofloxacin (CIP) and an efflux pump inhibitor 5-Nitro-2-(3-phenylpropoxy) pyridine (5-NPPP) to re-sensitize the pathogen towards antibiotics and reduce antimicrobial resistance. The study involves the fabrication of CIP-loaded polyvinyl alcohol (PVA) hydrogels and the development of polymeric nanoparticles for the controlled release of 5-NPPP. In vivo studies using Balb/c mice models demonstrate the effectiveness of the dual drugs loaded hydrogels in reducing bacterial load and promoting re-epithelization.

Performance of ceramic cutting tool with groove and cross-chevron surface textures filled with semi-solid lubricants

Summary: This study integrates surface texturing and extended self-lubrication to achieve a hassle-free and toxic-free working environment for machining grey cast iron materials. The experimental results show that using semi-solid lubricant filled textured tools can reduce cutting forces and friction coefficients, particularly at higher cutting speeds. However, it was found that the texture surfaces and lubricant-filled gaps away from the cutting edge did not contribute to the cutting performance.
