Impact of primary colorectal Cancer location on the KRAS status and its prognostic value

BackgroundColorectal cancer (CRC) originating from the right-sided or left-sided colon is distinct clinicopathological entity. The KRAS status and its prognostic value in CRC remain controversial. This study aimed to investigate the association of KRAS status with clinicopathological features and prognostic value in CRC.Methods178 colon cancer and 145 rectal cancer patients were enrolled. KRAS mutation test was performed on paraffin-embedded tumor samples using PCR methods. The colon cancer was divided into right-sided colon cancer (RCC) and left-sided colon cancer (LCC). Studies that reported the association of KRAS mutation with CRC clinical features and prognosis in databases were searched prior to 2018. The data of the present study was combined with the data of published studies using meta-analysis methods.ResultsNo significant difference between colon cancer and rectal cancer regarding the KRAS status. The KRAS mutation was much frequent in RCC than in LCC (p=0.010). 17 studies with 11,385 colon cancer patients were selected, the pooled results of our data and previous published data showed that KRAS mutation was more frequent in RCC compared with in LCC (p<0.01); KRAS mutation was not associated with the prognosis in RCC patient; however, KRAS mutation indicated a poor prognosis in LCC patients compared with KRAS wild type (p<0.01).ConclusionKRAS status has no difference between colon cancer and rectal cancer. KRAS mutation was more frequent in RCC than in LCC, and associated with a poor prognosis in LCC patients, but not in RCC patients.

Percentage of Natural Killer (NK) Cells in Peripheral Blood Is Associated with Prognosis in Patients with Gastric Cancer: A Retrospective Study from a Single Center

Summary: In this study involving 180 gastric cancer patients, it was found that patients with a higher percentage of NK cells had longer survival times and could serve as an independent prognostic indicator for gastric cancer. NK cell percentage was positively correlated with lymphocyte count and albumin, while negatively correlated with CA125 and neutrophil-lymphocyte ratio. NK cells showed predictive value for the 5-year survival rate of gastric cancer, especially when combined with neutrophil-lymphocyte ratio. Patients at early T, N, and clinical stages tended to have a higher percentage of NK cells compared to those at advanced stages.

This study identified location-specific lncRNAs as ceRNAs in colon cancer and established a DEL-based prognostic signature. Tumor-infiltrating immune cells based TMEscore and related gene signature is associated with the survival of CRC patients and response to fluoropyrimidine-based chemotherapy

Summary: This study explored the relationship between tumor-infiltrating immune cells (TIICs) and the prognosis of patients with colorectal cancer (CRC) who underwent chemotherapy. The results showed that TIIC-based tumor microenvironment score (TMEscore) was associated with chemotherapy response and survival rate. Key TMEscore-related genes were identified using machine learning methods, and a gene signature was constructed to predict chemotherapy response. The study found that the expression of TMEscore-related genes increased in tumor tissues. These findings provide a new approach to understanding chemotherapy response and prognosis in CRC patients.

Insights on Ferroptosis and Colorectal Cancer: Progress and Updates

Summary: Patients with advanced-stage or treatment-resistant colorectal cancer (CRC) may benefit from new therapeutic strategies such as inducing ferroptosis, a type of regulated cell death characterized by lipid reactive oxygen species (ROS) accumulation. Recent studies have identified mechanisms, pathways, and molecules that trigger or inhibit ferroptosis in CRC cells, including coding and non-coding RNA, small-molecule compounds, and nanoparticles. Additionally, gene signatures and clusters with predictive value on CRC have been summarized.

SPINK4 promotes colorectal cancer cell proliferation and inhibits ferroptosis

Summary: The role of serine peptidase inhibitor Kazal type 4 (SPINK4) in colorectal cancer (CRC) and ferroptosis was investigated in this study. It was found that SPINK4 expression was decreased in CRC tissues, and overexpression of SPINK4 promoted cell proliferation and metastasis while inhibiting cell ferroptosis.
