Summary: Increasing attention has been given to peptides as potential therapeutics, leading to the search for new peptide synthetic approaches and modifications. Enzymes have recently gained attention as a novel strategy and powerful tool for peptide and protein ligation. This study presents a fully convergent chemoenzymatic peptide synthesis process for the synthesis of aviptadil, a therapeutic peptide, using different peptiligase variants in aqueous environments.

Summary: This study explores the use of Chemo-Enzymatic Peptide Synthesis (CEPS) to produce a therapeutic HER2 targeted Affibody molecule. The resulting molecule shows thermodynamic stability, binding specificity to HER2, and extended time in circulation. CEPS proves to be a viable method for producing functional Affibody-fusion molecules.

Summary: Blast disease caused by Magnaporthe oryzae is a significant challenge for rice production worldwide. Chemical fungicides are unsafe and contribute to the emergence of resistant strains. Antimicrobial peptides, such as human salivary peptide histatin 5 (Hst5), offer a safe and effective alternative. This study investigated the antifungal activity of Hst5 on M. oryzae, finding that it causes morphological defects, inhibits germination and formation of blast lesions, and interacts with the fungus' genomic DNA. The multi-target antifungal mechanism of Hst5 provides an environmentally friendly option to combat blast infections in rice and could be explored for other crop pathogens as well.

In a mouse model, histatin-1 increased intra-osseous mineralization, collagen matrix deposition, and closure of cortical defect edges. It also promoted the expression of the osteogenic marker alkaline phosphatase and blood vessel formation.

Summary: We present a robust and practical method for chemical protein synthesis using an o-nitrobenzyl group as a temporary protective group. Through further investigation, we establish a reliable strategy for the quantitative photodeprotection of the o-nitrobenzyl group. This stable protective group is successfully applied in the chemical synthesis of a programmed death ligand 1 fragment, providing a practical approach for hydrazide-based native chemical ligation.

Summary: The successful application of nanomedicine against glioma relies on the fabrication of specific and efficient glioma targeted multifunctional theranostics. In this study, a novel nanoplatform composed of anisotropic gold nanoroses co-loaded with doxorubicin and the near-infrared active/responsive dye indocyanine green was fabricated. The nanoplatform exhibited targeted abilities and optical imaging functions, and showed promising efficacy in combating glioma.

Summary: This study focused on the effect of bovine lactoferrin (bLF) and LF peptides on cervical and endometrial cancer cell lines, finding that they inhibited cell proliferation and caused cellular morphological changes and damage. bLF induced apoptosis, LFcin17-30 and LFampin265-284 induced both apoptosis and necrosis, while LF chimera induced necrosis. Additionally, bLF and LF chimera showed an additive interaction with cisplatin against uterine cancer cells.

Summary: Histatin-1 is a novel osteogenic factor that promotes bone cell adhesion, migration, and differentiation, as well as mineralization in vitro. It induces the expression of osteogenic genes and triggers nuclear relocalization of beta-catenin, crucial events required for bone tissue regeneration. These effects are seen not only in osteogenically committed cells, but also in primary mesenchymal cells derived from the apical papilla and dental pulp.
