Summary: Analysis of miRNA expression in head and neck squamous cell carcinoma (HNSCC) revealed downregulation of the miR-30 family, especially miR-30e-5p and miR-30c-1-3p, which are associated with shorter patient survival. Further investigation showed that miR-30e-5p suppresses HNSCC cell migration and controls multiple oncogenic genes, including FOXD1. Knockdown of FOXD1 inhibited malignant phenotypes of HNSCC cells. These findings provide potential prognostic markers and therapeutic targets for HNSCC.

Summary: This study found that miR-199a-3p/5p is down-regulated in non-small cell lung cancer (NSCLC), and overexpression of miR-199a-3p/5p can inhibit cell proliferation, migration, and promote apoptosis. Rheb is identified as a common target of miR-199a-3p and miR-199a-5p, participating in the regulation of the mTOR signaling pathway. Additionally, miR-199a significantly inhibits tumor growth and metastasis in NSCLC and enhances the sensitivity of gefitinib in EGFR-T790M mutation. MiR-199a-3p/5p has the potential to serve as an early diagnostic marker or therapeutic target for NSCLC.

Low expression of circPTTG1IP is associated with poor prognosis in HCC patients. Mechanistically, circPTTG1IP functions as a competing endogenous RNA (ceRNA) of RNF125 by binding miR-16-5p, thereby increasing the level of E3 ubiquitin ligase RNF125 and promoting the degradation of JAK1 protein. Filgotinib, a JAK1 inhibitor, can restrict HCC progression induced by low circPTTG1IP expression.

It was found that miR-181a-5p, miR-181b-5p, and miR-383-5p were differentially expressed, with miR-181a-5p and miR-181b-5p downregulated and miR-383-5p upregulated in the uncontrolled group. ROC curve analysis showed that miR-383-5p had an NPV of 84.3% and a PPV of 84.5%. Thus, these miRNAs can serve as biomarkers and tools to guide clinical decisions in response to fg-SRLs treatment.

Summary: Chronic allograft dysfunction (CAD) is a major condition affecting long-term kidney graft survival. In this study, the correlation between plasma miR-21 and the severity of CAD was investigated. RNA sequencing was performed and differentially expressed miRNAs in CAD were identified. MiR-21-5p, miR-20a-5p, and miR-101-3p were found to be associated with the TGF-beta pathway and CAD. These findings may lead to the development of early prediction miRNA biomarkers and therapeutic tools for CAD in the field of kidney transplantation.
