Fluorinated Bambusurils as Highly Effective and Selective Transmembrane Cl-/HCO3- Antiporters

The exchange of chloride and bicarbonate across lipid bilayers is an important biological process. Synthetic molecules can act as mobile carriers for these anions, although most show little selectivity. Here we report on three bambus[6] uril macrocycles functionalized with fluorinated benzyl groups, which are able to exchange Cl- and HCO3- efficiently. Remarkably, rates for Cl-/NO3- exchange are two orders of magnitude lower. The higher rates of a Cl-/HCO3- transport can be explained by the ability of the bambusurils to complex Cl- and HCO3- simultaneously, facilitating their exchange at the bilayer interface. Furthermore, the exceptionally high affinity and selectivity of these systems for NO3- appear to contribute to the poor Cl-/NO3- exchange. This work not only demonstrates the importance of anion binding characteristics on anion transport but also the potential relevance of bambusurils for anion transport applications considering the high rate observed for Cl-/HCO3- exchange.

Bambusuril Macrocycles as Mediators of Supramolecular Interactions: Application to the Europium Cage Helicate

Summary: Bambusurils can be used for tuning and characterizing supramolecular systems by acting as anion scavengers in metal-mediated self-assemblies. They can manipulate the subtle equilibria in the system, trigger the self-assembly process, and enhance specific interactions between components. Bambusuril has the ability to selectively remove coordinating anions from mixtures, enabling regenaration of the supramolecular structure.

Transmembrane transport of fluoride studied by time-resolved emission spectroscopy

Summary: Here we introduce a novel approach using a europium(iii) complex to monitor the transmembrane transport of fluoride into liposomes. By utilizing the long emission lifetime of this probe, we are able to accurately measure the transport activity of fluorescent transporters. The high sensitivity, selectivity, and versatility of this assay enable us to investigate various types of fluoride transporters and unravel their mechanisms of action.

Synthesis of Glycoluril Dimers with the Ability to Form Polymeric Self-Associates in Water

Summary: Supramolecular self-assembly in water, leading to polymeric structures, has potential applications in biology and medicine. In this study, we report the self-association of host molecules based on glycoluril dimers into linear oligomers in water. The polymerization degree of the resulting supramolecular aggregates was calculated using the isodesmic model and Carothers equation. A model compound was prepared for a better understanding of the forces responsible for the self-association of glycoluril dimer-based monomers in water.

Dissecting transmembrane bicarbonate transport by 1,8-di(thio)amidocarbazoles

Summary: This study investigated the transport of bicarbonate and chloride ions by carbazoles with two amido/thioamido groups using a bicarbonate-sensitive probe. The researchers found a significant concentration dependence of the transporters. Based on insights into anion transport mechanisms, the compounds were tested for antimicrobial activity and showed a strong correlation with their ion transport properties.

Dynamic covalent chemistry with azines

Summary: Dynamic covalent chemistry has been widely used for applications requiring stability of covalent bonds and the ability to exchange building blocks. In this study, azines are introduced as a dynamic covalent functional group that combines the best characteristics of imines and acylhydrazones. The results demonstrate that azines are stable in the presence of water and that dynamic combinatorial libraries of azines and aldehydes equilibrate in less than an hour.

Calix[6]arenes with halogen bond donor groups as selective and efficient anion transporters

Summary: This study demonstrates the anion binding and anion transport properties of a series of calix[6]arenes decorated with halogen bond or hydrogen bond donating groups on their small rim. The results show that the halogen bond donating iodotriazole groups enable highly selective transport of chloride and nitrate anions, without the transport of protons or hydroxide.
