Papillary thyroid carcinoma with tall cell features is as aggressive as tall cell variant: a meta-analysis

There are still ongoing debates as to which cut-off percentage of tall cell (TC) should be used to define tall cell variant (TCV) papillary thyroid carcinoma (PTC). In this meta-analysis, we aimed to investigate the clinicopathological significance of PTC with tall cell features (PTC-TCF, PTC with 10-50% of TCs) in comparison with classical PTC and TCVPTC (PTC with more than 50% of TCs) to clarify the controversial issue. Four electronic databases including PubMed, Web of Science, Scopus and Virtual Health Library were accessed to search for relevant articles. We extracted data from published studies and pooled into odds ratio (OR) and its corresponding 95% confidence intervals (CIs) using random-effect modeling. Nine studies comprising 403 TCVPTCs, 325 PTC-TCFs and 3552 classical PTCs were included for meta-analyses. Overall, the clinicopathological profiles of PTC-TCF including multifocality, extrathyroidal extension, lymph node metastasis, distant metastasis and patient mortality were not statistically different from those of TCVPTC. Additionally, PTC-TCF and TCVPTC were both associated with an increased risk for aggressive clinical courses as compared to classical PTC. The prevalence of BRAF mutation in PTC-TCF and TCVPTC was comparable and both were significantly higher than that in classical PTC. The present meta-analysis demonstrated that even a PTC comprising only 10% of TCs might be associated with a poor clinical outcome. Therefore, the proportions of PTC in PTC should be carefully estimated and reported even when the TC component is as little as 10%.

Prognostic Implication of Patient Age in H3K27M-Mutant Midline Gliomas

Summary: This study investigated the plasma metabolic profiles of tuberculosis (TB) patients and TB patients with type 2 diabetes mellitus (DM) using a multimodal metabolomics and lipidomics approach. Significant differences in biological processes and biomarkers were identified between TB and TB-DM, providing insights into the pathophysiology of TB-DM and potential improvements in TB treatment. The integration of multimodal metabolomics and lipidomics provides a comprehensive understanding of the metabolic changes associated with TB-DM, enhancing our knowledge of DM comorbidity in TB infection and contributing to the development of more effective treatment.

De novo Versus Secondary Dedifferentiated Chordomas: A Population-Based Analysis and Integrated Individual Participant Data Meta-Analysis

Summary: This integrated study aimed to investigate the similarities and differences in clinicopathological manifestations, prognoses, and molecular profiles of de novo versus secondary dedifferentiated chordoma (DC). The results showed that de novo and secondary DCs were statistically comparable in terms of patient demographics, clinical manifestations, and prognoses. Gross total excision and radiotherapy were optimal treatments associated with better outcomes of DC patients.

Summary: Mitochondria-associated ER membranes (MAMs), formed by the endoplasmic reticulum (ER) and mitochondria, play an important role in maintaining calcium homeostasis and neurotransmitter release in the neuronal system. The kinase CK2A1 and its interaction with PACS2 regulate the composition and calcium transport of MAMs. Mutations in PACS2 associated with developmental and epileptic encephalopathy disrupt MAM integrity, leading to impaired mitochondrial calcium uptake and increased cytosolic calcium levels, ultimately causing abnormal neurotransmitter release.

Summary: This study investigated the expression of lipid-related genes during anti-TB treatment and their potential use as biomarkers of treatment response. The expression levels of 10 genes were altered during standard anti-TB treatment. The 10-lipid-gene signature could distinguish patients with TB from those with latent tuberculosis infection and non-TB controls, and it could also be useful for monitoring response to anti-TB treatment.

The clinicopathological features and prognosis of silent corticotroph tumors: an updated systematic review and meta-analysis

Summary: This study compared silent corticotroph tumors (SCT) with other non-functioning pituitary neuroendocrine tumors (NFT) and functioning corticotroph tumors (FCT) and found that SCTs have demographic, clinicopathological and prognostic differences. These differences indicate that SCTs are high-risk tumors, and appropriate treatment decisions and stricter follow-up plans should be tailored to improve patient outcomes.
