Summary: Radiotherapy can cause DNA damage and destroy cancer cells, but it can also induce cancer stem cell properties, leading to metastasis and invasion. Researchers have developed a ROS-responsive liposome containing the BMI1 inhibitor PTC-209 to target radioresistant HCC tissue. The study found that the released PTC-209 can restore radiosensitivity in HCC tumors by differentiating cancer stem cells.

Summary: The upregulation of SNRPB in HCC is associated with tumor malignancy and patient survival. Functional studies indicate that SNRPB promotes HCC cell proliferation and stemness maintenance, while also inducing metabolic reprogramming in HCC cells. In addition, SNRPB facilitates the formation of specific splice variants that activate pathways contributing to HCC progression.

Summary: This study reveals the role of HNRNPM in regulating the positive association between cancer stemness and immune evasion in hepatocellular carcinoma. By controlling the alternative splicing of MBD2, HNRNPM affects the activation of the WNT/beta-catenin pathway and further regulates the expression of OCT4 and SOX2. Inhibition of HNRNPM promotes the activation of CD8+ T cells and enhances the effectiveness of anti-PD-1 immunotherapy. These findings provide new insights for the development of tailored immunotherapeutic approaches for hepatocellular carcinoma.

Summary: This study reveals the crucial role of alanine-glyoxylate aminotransferase (AGXT) in supporting liver cancer stem cells (LCSCs) and maintaining their stemness. AGXT may sustain the self-renewal potential of LCSCs by upregulating the expression of SRY-box transcription factor 2 (SOX2) and octamer-binding transcription factor 4 (OCT4).

Summary: Therapeutic induction of senescence may increase cancer stem cell phenotype, leading to increased tumor aggressiveness and disease recurrence. In liver cancer cells, doxorubicin can induce cellular senescence, increase stem cell gene expression, and cause a shift in stem cell characteristics.

Summary: LGR5 is a newly defined stem cell marker expressed in endoderm-derived organs such as the small intestine, colon, and pancreas. It plays a significant role in liver regeneration and stem cell maintenance, with high expression observed in various cancers including hepatocellular carcinoma. Activation of the canonical Wnt pathway increases LGR5 expression in a subset of HCC cell lines with a "hepatoblast-like" appearance, resulting in enhanced tumor-promoting abilities.

Summary: This study revealed a novel molecular pathway that facilitates HCC cell proliferation, migration, and stemness, shedding new light on the treatment of HCC. Up-regulated SEC61A1 was found to promote these processes in HCC cells, with MiR-491-5p negatively regulating SEC61A1 and inhibiting cell proliferation and migration. VPS9D1-AS1 was identified as an upstream regulator of SEC61A1 through miR-491-5p, and EGR1 was shown to be responsible for the transcriptional activation of both SEC61A1 and VPS9D1-AS1.

Summary: Hepatocellular carcinoma (HCC) is mainly driven by liver cancer stem cells (LCSCs), which contribute to tumor progression, metastasis, and treatment resistance. However, the regulatory mechanisms of LCSCs in HCC are still unclear. Understanding the signaling pathways responsible for LCSC maintenance and survival may improve patient outcomes.

Summary: Intra-tumoral immune cells promote the stemness of cancer stem cells (CSCs) in the tumor microenvironment, which can play a role in tumor progression, relapse, and resistance to immunotherapy. Stem-like T cells and stem cell-derived NK cells show promising potential in improving the therapeutic efficacy of immunotherapy.
