Prevalence of Leishmania tropica in school boys of khyber agency, FATA near Pak-Afghan border

In Pakistan leishmaniasis occurs periodically throughout the year and various out breaks are reported frequently. In continuation of our research on this neglected disease, the aim of present study is to explore: (1) the prevalence of cutaneous leishmaniasis in school boys; (2) Leishmania species identification in order to epidemiology and dynamics of the disease; (3) Identification of risk factors for Leishmaniasis especially for CL. The data was collected in August 2014 The experimental strategy involved a questionnaire for data collection and along with clinical diagnosis of 134 out of 9368 students for incidence of Leishmania spp. in 7 square kilometres area in the schoolboys at Tehsil Landi Kotal, District Khyber Agency, FATA Pakistan. The parasitological and molecular diagnosis of clinically suspected cutaneous leishmaniasis cases were performed using microscopical examination of Giemsa-stained smears of lesion exudates and minicircle kDNA semi nested PCR, respectively. Microscopy (x = 1000) positive cases were 84/134 (62.6%) and 50/134 (37.4%) slides did not showed any presence of amastigotes of Leishmania spp. The samples were amplified using kDNA semi nested PCR and confirmed the presence of L Tropica (Ac.no KT 985473). PCR positive cases were 97/134 (72.4%) and 37(27.6%) were negative. The prevalence of L. tropica in school boys was 1.4% (134/9368) in the total population studied (n = 9368). The parasite prevalence might be greater as only male students were considered in the study due to ethical and social issues and limitations. (C) 2016 Elsevier B.V. All rights reserved.

Summary: By using density functional theory (DFT) calculations, we investigated the reactivity of synthesized quinazolinone towards bromination and nitration. The results showed a good agreement with experimental data, with a correlation above 90%. The optimized molecular geometries were compared with X-ray structures, and the nucleophilic site was determined through computational analysis.

New Succinimide-Thiazolidinedione Hybrids as Multitarget Antidiabetic Agents: Design, Synthesis, Bioevaluation, and Molecular Modelling Studies

Summary: Diabetes mellitus (DM) is a metabolic disorder resulting from pancreatic dysfunction, characterized by reduced insulin production or insulin resistance. In this study, we designed and synthesized new succinimide-thiazolidinedione hybrids for the treatment of DM. We successfully synthesized five new derivatives (10a-e) with different substitution patterns on the N-atom of the succinimide ring. Compound 10d showed the most potential as an antidiabetic agent in in vitro assays. Further investigations in experimental animals confirmed its promising antidiabetic activity. Molecular docking studies also demonstrated the interaction of compound 10d with target enzymes involved in diabetes. Overall, we have identified a new class of compounds with potential as antidiabetic agents.

An SUI-based approach to explore visual search results cluster-graphs

Summary: Nowadays, with the exponential growth of online production and extensive perceptual power of visual contents, users' information needs have become complicated. Research has shown that users have a keen interest in accessing image objects to satisfy their visual information needs. However, existing search engines hinder image exploration due to linear lists or grid layouts that sort image results by relevancy. This research proposes a Search User Interface (SUI) approach to enable non-linear reachability of image results by offering interactive exploration and visualization options, resulting in high satisfaction (76.83%) and usability (83.73%) scores according to usability tests.

Summary: In this study, a newly synthesized compound (Succ-5) was analyzed through spectral methods and molecular docking. It was pre-clinically evaluated for therapeutic effects and safety profile. The results showed that Succ-5 had significant cardioprotective, hepatoprotective, and lipolytic effects in 5-FU-induced toxicity.

Synthesis, Characterization, and Pharmacokinetic Studies of Thiazolidine-2,4-Dione Derivatives

Summary: Various derivatives of thiazolidine-2,4-dione (C1-C5) were synthesized through chemical reaction and modification, and their chemical structures were determined by NMR spectroscopy. In silico ADME studies revealed the pharmacokinetic properties of the compounds, including rotatable bonds, lipophilicity, water solubility, GI absorption, blood-brain barrier permeability, and drug metabolism. The study also evaluated the drug likeness properties of the derivatives. The results suggest that these compounds have potential for improving pharmacokinetic profiles and reducing toxicity.

Effect of Gabapentin-Fluoxetine Derivative GBP1F in a Murine Model of Depression, Anxiety and Cognition

Summary: This study evaluated the effects of GBP1F, a complex of fluoxetine and gabapentin, on locomotor activity, anxiety, depression-like behavior, and cognitive behavior in mice. The results showed that GBP1F mildly suppressed locomotor activity, improved anxiety and depression-like behavior, had no effect on cognitive behavior, and increased serotonin and 5-HIAA concentrations in the hippocampus and striatum, as well as dopamine and vitamin C levels in the striatum. Thus, GBP1F has anxiolytic and antidepressant-like effects, but further studies are needed to investigate its precise mechanism of action.

Current research aims to optimize this lead compound for diabetes treatment by manipulating multiple pathways simultaneously.

Multi-step attack detection in industrial networks using a hybrid deep learning architecture

To address these threats, security systems have been implemented, but they only respond after the attacks have occurred and do not actively prevent them. Identifying malicious attacks is crucial for industrial networks as they can cause system malfunctions, network disruptions, data corruption, and sensitive information theft. This study proposes a hybrid model that combines convolutional neural networks (CNN) and deep belief networks (DBN) for intrusion detection in industrial networks, utilizing the MSCAD dataset and various evaluation metrics to test its effectiveness.

Summary: A series of novel tetrahydro-2H-1,3,5-thiadiazine-2-thiones compounds were synthesized and characterized. The compounds showed promising anticancer activity against HeLa cell line, especially compounds 3, 5, 6, and 9 with IC50 values <4 μM. The compounds also exhibited potent anti-inflammatory potential by inhibiting nitric oxide (NO). Docking studies revealed strong interactions between the compounds and inducible NO synthase enzyme, supporting the in vitro results. These compounds could serve as potential candidates for the development of anticancer drugs.

Rapid, sensitive, and convenient detection of Plasmodium falciparum infection based on CRISPR and its application in detection of asymptomatic infection

Summary: Rapid and convenient detection of Plasmodium falciparum, the deadliest malaria parasite in Africa, is crucial for malaria eradication efforts in medical resource-limited settings. A CRISPR-based diagnostic method that can detect P. falciparum without nucleic acid extraction was developed and showed high sensitivity and consistency with qPCR test. It can also detect artemisinin-resistant strains and other laboratory-cultured strains. This method has great potential for clinical testing and large-scale community screening in Africa and other regions.

Summary: Porcine circovirus 3 (PCV-3) has been reported globally since its first identification in 2015. Limited data on PCV-3 in Africa led to a study in Namibia, which found a high prevalence of PCV-3 in backyard piggeries, but not in commercial farms with higher biosecurity measures. The source of viral introduction and factors such as semen importation, feeding backyard pigs with human food waste, pig exchanges, and wildlife movements may be involved in the dispersal of PCV-3 within Namibia. Further studies are needed to understand the epidemiology and impact of PCV-3 on pig productivity and wildlife health.

Spatial analysis of human and livestock anthrax in Lai Chau province, Vietnam (2004-2021)

Summary: This study examined the epidemiology of anthrax in Lai Chau province, Vietnam, using historical data. Spatial clusters of human and livestock anthrax were identified, and the characteristics of the disease were described. Additionally, the decrease in livestock anthrax vaccine coverage was associated with an increase in disease incidence.
