Chalcones exhibit a broad spectrum of biological activities, mainly due to alpha, beta-unsaturated ketone, and are precursors of the biosynthesis of flavonoids present in plants. These compounds have been shown to be useful in the biological approach, proven by the broad spectrum of biological activities reported, and also in the technological approach, considering their potential as nonlinear optic (NLO) material. In this context, this work aimed to examine the crystallization and characterization of fluorinated chalcone (E)-1-(4-fluoropheny1)-3-(4-isopropylphenyeprop-2-e-n-1-one (DFC). A comprehensive structural study of DFC was carried out to understand the process of stabilizing the crystalline lattice through X-ray diffraction, infrared spectroscopy, and molecular modeling studies. Finally, the electrical properties of DFC were calculated by using the supermolecule method (SM). DFC molecules are connected by means of C-H center dot center dot center dot O and C-H center dot center dot center dot F intermolecular contacts, forming dimers which play an important role in the stabilization of crystal packing. Molecular modeling studies indicated that this compound could act as an anti-tuberculosis ligand because of its high binding affinity with the M. tuberculosis enoyl-acyl carrier protein, InhA. On the other hand, theoretical calculations were performed to evaluate the NLO properties of DFC and indicated that it showed good potential. An Update on the Synthesis and Pharmacological Properties of Pyrazoles Obtained

Jaqueline E. Queiroz, Lucas D. Dias, Giuliana M. Vila Verde, Gilberto L. B. Aquino, Ademir J. Camargo

Summary: This review presented and discussed the synthesis and pharmacological properties of pyrazoles obtained from Chalcone in the last 5 years (2016-2020). The cyclization and acetylation method of alpha,beta-unsaturated chalcones and substituted hydrazine were selected and analyzed as synthetic approaches for pyrazoles. A total of 105 pyrazole derivatives were evaluated for their pharmacological activities, including antineoplastic, anti-inflammatory, antioxidant, antibacterial, antifungal, and other properties. It is important to research the synthetic methods and therapeutic properties of different pyrazole derivatives.

Jean M. F. Custodio, Fernanda S. Fernandes, Wesley F. Vaz, Allen G. Oliver, Clodoaldo Valverde, Francisco A. P. Osorio, Guilherme R. Oliveira, Tertius L. Fonseca

Summary: The theoretical analysis of electronic properties of a synthesized nitro chalcone with a neolignan moiety revealed the relationship between crystal structure and nonlinear application potential. The impact of environment polarization on absorption spectrum and linear polarizability was found to be moderate, while it was significant for the second hyperpolarizability of the molecule. The results indicate that the NOPP crystal exhibits promising nonlinear optical properties.

Interaction mechanism of nordentatin with human α-1 acid glycoprotein and human colorectal cancer HCT-116 cells

Bo Yi, Li Zhang, Haiyang Zhou

Summary: Plasma protein binding plays a crucial role in the pharmacokinetics and pharmacodynamics of therapeutic bioactive materials. In this study, the interaction of nordentatin, a member of coumarins, with human alpha-1 acid glycoprotein (AGP) was investigated, and the anticancer potency of nordentatin against human colorectal cancer cells was evaluated. The results showed that nordentatin binds to AGP through a static quenching mechanism and exhibits inhibitory effects on the growth of HCT-116 cells by increasing intracellular ROS and [Ca2+] levels and triggering apoptosis-related gene overexpression. Therefore, nordentatin could be a promising plant-based therapy for colorectal cancer.
