Purpurolide A (1), an unprecedent sesquiterpene lactone with a rarely encountered 5/5/5 spirocyclic skeleton, along with two new 6/4/5/5 tetracyclic sesquiterpene lactones (2 and 3), were isolated from the cultures of the endophytic fungus Penicillium purpurogenum IMM003. The structures and absolute configurations of 1-3 were established by spectroscopic analysis, single-crystal X-ray diffraction, and calculations of the C-13 NMR and ECD data. Compounds 1-3 showed significant inhibitory activity against pancreatic lipase.

Summary: A new family of circularly polarized luminescence-active small organic molecules (CPL-SOMs) with high fluorescence efficiency and fascinating CPL properties was discovered and characterized. The usefulness of CPL-calculation as a unique method for stereochemical assignment was demonstrated. This study may open up a new perspective for the future development of stereochemical studies and CPL materials.

Penicipurate A, a new polyketide derivative from the endophytic fungus Penicillium purpurogenum

Summary: A new polyketide derivative penicipurate A (1) containing a 3-hydroxydecanoic acid ester moiety was isolated from the solid cultures of the endophytic fungal strain Penicillium purpurogenum in the leaves of Edgeworthia chrysantha. The structure of 1 was determined using various spectroscopic methods and chemical degradation. Compound 1 exhibited moderate inhibitory activity against pancreatic lipase.

Summary: This study analyzed the serum metabolites in hamsters with ZSP-induced liver damage using metabolomics methods and identified 12 metabolites associated with ZSP-induced hepatotoxicity. Amino acid metabolism and the urea cycle were significantly altered, and the role of three amino acid metabolic enzymes was validated. The major hepatotoxic constituents in ZSP were identified as furanoids from two herbs, and a preparation method with low hepatotoxicity was proposed.

Deciphering the species differences in CES1A-mediated hydrolytic metabolism by using a bioluminescence substrate

Summary: This study investigated the species differences in CES1A-mediated hydrolytic metabolism using a bioluminescence sensor and liver microsomes from six mammalian species. The results showed significant variation in hydrolytic rate among different species. The findings suggest the importance of selecting appropriate animal models for evaluating the in vivo effects of CES1A inhibitors.

OL showed excellent specificity, high detection sensitivity, and high signal-to-noise ratio for human Notum. This study provides a reliable and easy-to-use tool for sensing Notum activities in biological systems, which will strongly facilitate fundamental studies, disease diagnosis, and drug discovery related to Notum.
