Combination therapy of sorafenib with mesenchymal stem cells as a novel cancer treatment regimen in xenograft models of hepatocellular carcinoma

Aim Hepatocellular carcinoma (HCC) is the most common liver malignancy and the second leading cause of cancer-related deaths in the world. Sorafenib is the first-line treatment of HCC. Although sorafenib has positive effects on the survival of patients, novel therapeutic strategies are needed to extend survival and improve the efficacy of sorafenib. This study combines sorafenib with mesenchymal stem cells (MSCs) as a new approach to enhance the efficacy of sorafenib. Material and methods A subcutaneous xenograft model of HCC, established by human HepG2 cell lines, was implanted into the flank of nude mice and was used to evaluate tumor growth after treatment with sorafenib alone or in combination with MSCs. The aspartate aminotransferase, alanine aminotransferase, blood urea nitrogen, and creatinine levels were measured for safety assessment. Histopathological studies were performed using hematoxylin and eosin staining, and immunohistochemistry tests were performed to evaluate proliferation (Ki67) and angiogenesis (CD34). The TUNEL assay was used to detect apoptosis and measure the expression of major inflammatory cytokines (IL-1a, IL-10, and TNF-alpha) with real-time polymerase chain reaction. Result Sorafenib, in combination with MSCs, strongly inhibited tumor growth in the xenograft model. Furthermore, the combination therapy significantly inhibited HCC cell proliferation, decreased tumor angiogenesis, and induced apoptosis and maintained antitumor-associated anti-inflammatory effects of MSCs. Conclusion This combination therapy strategy could be used as a new therapeutic approach to the treatment of HCC that significantly improves upon the results achieved using sorafenib as monotherapy.

Summary: This study aimed to develop a controlled-shape hydrogel and apply conditioned-media of mesenchymal stromal cells (CM-MSCs) to improve primary hepatocyte functions. The patterned alginate hydrogel was produced using an electrodeposition method. The presence of CM-MSCs significantly increased urea synthesis in the patterned hydrogel alginate compared to 2D culture after 3 days.

The novel treatments based on tissue engineering, cell therapy and nanotechnology for cutaneous leishmaniasis

Summary: Cutaneous leishmaniasis (CL) is a global health issue, and conventional treatments have drawbacks. Topical treatment is a cost-effective approach for CL, and electrospun fibers and hydrogels are promising drug release systems for treating skin lesions. Polymeric scaffolds, microneedle patches, and nanocarriers enhance drug delivery to the skin. Additionally, cell therapy shows potential in overcoming leishmaniasis. This study investigates various treatment options, such as tissue engineering scaffolds, nanotechnology, and cell therapy, for reducing CL complications.

The Efficiency of Stem Cells (SCs) Differentiation into Functional Hepatocytes for Treating Liver Disorders: A Systematic Review

Summary: Stem cells offer a new avenue for treating liver disorders. They have the potential to differentiate into various specialized cell types, and efforts have been made to enhance their differentiation into hepatocytes. However, the effectiveness of this differentiation process depends on factors such as cell-to-cell adhesion and biological changes. A comprehensive review of studies on stem cell differentiation revealed that most research focused on mesenchymal stem cells, with acute liver failure being the most investigated liver disorder. Tissue engineering methods were commonly used for differentiation, and functional parameters were frequently evaluated.

Evaluation of the osteoconductive properties of scaffold containing platete-enriched-fibrin (PRF) with three calcium phosphate (TCP) in the alveolar socket repair after tooth extraction: An animal study

Summary: This study investigated the properties of the PRF scaffold with tricalcium phosphate nanoparticles in socket preservation and found that it promotes bone regeneration. The experimental results showed that PRF/TCP scaffold accelerates bone formation, resulting in a higher amount of bone tissue and collagen fibers. The mechanical and chemical tests also confirmed that the addition of TCP to the PRF scaffold reduces swelling and prolongs degradation time. In conclusion, the injectable PRF\TCP hydrogel is a promising candidate for bone repair and regeneration.

Advantages of Material Biofunctionalization Using Nucleic Acid Aptamers in Tissue Engineering and Regenerative Medicine

Synthetic ligands like nucleic acid aptamers have advantages in terms of synthesis ease, minimal immunogenicity, specificity, and stability. This study reviews the advantages of nucleic acid aptamers in tissue engineering, such as attracting endogenous stem cells for tissue regeneration and improving controlled drug delivery.

Cytotoxicity of WT1-reactive T cells against Wilms tumor: An implication for antigen-specific adoptive immunotherapy

Summary: This study aimed to investigate the feasibility of isolating WT1-reactive T cells from PBMCs and assess their cytotoxicity against WiTu cells. The results showed higher proportions of WT1-reactive T cells were isolated from Wilms tumor patients compared to healthy donors. WT1-reactive T cells exhibited anti-tumoral activity in a dose-dependent manner and mediated significantly greater cytotoxicity than non-WT1-reactive PBMCs on WT1+ WiTu cells. The cytotoxicity of standard chemotherapy was significantly lower than that of WT1-reactive T cells.
