Treatment of Radiation-Induced Oral Mucositis Using a Novel Accepted Taste of Prolonged Release Mucoadhesive Bi-medicated Double-Layer Buccal Films

The aim of this study was to develop a novel double-layer bi-medicated prolonged release mucoadhesive buccal film (MBF) containing lidocaine hydrochloride (LC) and diclofenac potassium (DK). The ultimate goal of the prepared system is the treatment of radiation-induced oral mucositis pain with improved patient acceptance. MBFs were prepared using 3 x 2(2) randomized full factorial design for film optimization. Nanoemulsion system (NES) was used to mask DK bitter taste. The prepared films were characterized, viz thickness, mass uniformity, surface pH, folding endurance, swelling studies, ex vivo bioadhesive strength, in vitro drug release, and ex vivo permeation. The in vivo evaluation was carried out by testing the anti-inflammatory and analgesic activities on rats followed by a clinical study on patients to prove their acceptance. The optimized MBF composed of 10% w/w HPMC-4KM, 50 mg LC, and 50 mg DK-NES was selected due to prolonged in vitro drug release pattern and ex vivo permeability (95.24 +/- 2.14 and 93.48 +/- 3.24% in 6 h, respectively). MBF exposed a strong anti-inflammatory effect from 61 to 87% inhibition with a strong analgesic effect when compared to DK (R) and LC (R), respectively. The clinical study revealed that films were accepted by the patients, and the presence of LC on the outer side helped in pain feeling reduction while DK-NES in the inner side facilitated in rapidly relieving the inflammation effect.

Results showed that the transethosomal formulation increased telmisartan's transdermal absorption and bioavailability. Formulation and characterization of lamotrigine nasal insert targeted brain for enhanced epilepsy treatment

Summary: This study aimed to develop a nano-vesicular carrier system to enhance the therapeutic effect of lamotrigine for epilepsy treatment. The results showed that the optimized formulation exhibited high drug entrapment efficiency and sustained release. In vivo studies demonstrated improved brain targeting and absorption, indicating a promising approach for maximizing the antiepileptic effect of lamotrigine.

One-pot microwave synthesis of chitosan-stabilized silver nanoparticles entrapped polyethylene oxide nanofibers, with their intrinsic antibacterial and antioxidant potency for wound healing

Summary: Different physical and chemical techniques were used to prepare CHS/AgNPs nanocomposite, and microwave heating reactor was adopted due to its advantages of low energy consumption and shorter processing time. UV-Vis, FTIR, and XRD analysis confirmed the creation of AgNPs, and TEM images showed spherical particles with a size of 20 nm. CHS/AgNPs were embedded in PEO nanofiber through electrospinning, and their biological properties, cytotoxicity, antioxidant, and antibacterial activity were investigated. The nanofibers exhibited excellent antibacterial activity against E. coli and S. aureus, with ZOI of 51.2 +/- 3.2 and 47.2 +/- 2.1, respectively. Moreover, they showed non-toxicity against human skin fibroblast and keratinocytes cell lines, indicating their potential for wound infection prevention with minimal adverse effects.

Lyophilized Nasal Inserts of Atomoxetine HCl Solid Lipid Nanoparticles for Brain Targeting as a Treatment of Attention-Deficit/Hyperactivity Disorder (ADHD): A Pharmacokinetics Study on Rats

Summary: The study investigated the use of lyophilized nasal inserts of nanosized atomoxetine HCl solid lipid nanoparticles (ATM-SLNs) to directly deliver atomoxetine to the brain. A full factorial design was used to optimize the formulation of the ATM-SLNs, and Design Expert software was used to select the optimal formulations. The two optimum formulations were then evaluated for their physicochemical properties and compared to atomoxetine oral solution and intraperitoneal injection.

Enhance the oral insulin delivery route using a modified chitosan-based formulation fabricated by microwave

Summary: Chitosan was modified and functionalized to improve oral insulin delivery. The Cs-DNS NPs had a particle size of 33.29 ± 5.08 nm and positive zeta potential. In vitro experiments showed high insulin entrapment efficiency and gradual release at different pH levels. In vivo experiments demonstrated that insulin-incorporated Cs-DNS NPs effectively lowered serum glucose levels, with a bioavailability of 17.5 ± 0.31% and pharmacological availability of 8.6 ± 0.8%.

Summary: This study aimed to enhance the bioavailability of RXB using hybrid nanoparticles. It was found that using GMO-SAIB as co-formers effectively improved the bioavailability of RXB. The optimized formula was characterized and tested for its physical and chemical properties as well as pharmacokinetic properties. The study provides guidance for further improvement in drug delivery.
