The effect of orexin B on steroidogenic acute regulatory protein, P450 side-chain cleavage enzyme, and 3β-hydroxysteroid dehydrogenase gene expression, and progesterone and androstenedione secretion by the porcine uterus during early pregnancy and the estrous cycle

The aim of this study was to investigate the effect of orexin B (OXB) on progesterone (P-4) and androstenedione (A(4)) secretion by porcine endometrial and myometrial tissue explants and on the expression of key steroidogenic proteins and enzymes involved in steroid production. The hormones secretion and the expression of steroidogenic acute regulatory protein (StAR), P450 side-chain cleavage enzyme (CYP11A1), and 3 beta-hydroxysteroid dehydrogenase (HSD3B1) were analyzed on days 10 to 11, 12 to 13, 15 to 16, and 27 to 28 of pregnancy and during the luteal phase of the estrous cycle (days 10 to 11). Endometrial and myometrial explants were cultured in vitro in the presence of OXB (1, 10, or 100 nM) and OXB (1, 10, or 100 nM) with 1 mu M of JNJ (OX2R antagonist). Gene expression was examined by real-time PCR, and steroid secretion was determined by radioimmunoassay. Orexin B modulated StAR, CYP11A1, HSD3B1 mRNA content depending on the type of uterine tissue, the applied OXB dose, and the stage of pregnancy or the estrous cycle (P < 0.05). Orexin B increased P-4 secretion in all stages of early gestation (P < 0.05). Orexin B enhanced the release of A(4) on days 12 to 13, 15 to 16, and 27 to 28 of gestation, whereas on days 10 to 11 of early pregnancy, A(4) secretion decreased in the endometrium and increased in the myometrium (P < 0.05). These results indicate that OXB affects the expression of key steroidogenic regulators and the secretion of steroid hormones in the porcine uterus during early pregnancy.

Summary: This study aimed to evaluate the effects of orexin A (OXA) on mRNA abundance of enzymes involved in prostaglandin production and the concentrations of prostaglandin E2 (PGE2) and F-2 alpha (PGF(2 alpha)) in endometrial and myometrial explants. The results suggest that OXA may affect the synthesis and secretion of PGE2 and PGF(2 alpha) in the uterus by regulating the mRNA levels of enzymes involved in prostaglandin production.

Chemerin Affects P4 and E2 Synthesis in the Porcine Endometrium during Early Pregnancy

Chemerin achieves this impact by regulating the expression levels of specific steroidogenic enzyme proteins at different stages of pregnancy and the estrous cycle. Additionally, Chemerin also influences hormone synthesis through the Erk and Akt signaling pathways.

This study investigates the influence of Chemerin on alternative mRNA transcription in porcine luteal cells, including differential expression of long non-coding RNAs (DELs) and their interactions with differentially expressed genes (DEGs), differences in alternative splicing of transcripts (DASs), and allele-specific expression (ASEs). The results provide new insights into the effects of Chemerin on the functions of the corpus luteum.

Summary: This study examined the effects of CHEM on differentially regulated proteins in porcine endometrial tissue during implantation. It was found that CHEM modulates protein expressions involved in endometrial cell growth, migration, immune response, sensitivity to P4, and formation of new blood vessels. CHEM also appears to play a role in endothelial cell dysfunction during preeclampsia.

Summary: This study aimed to investigate the influence of steroid hormones on the expression of the chemerin system in the uterus of domestic pigs. The results showed that both estradiol and progesterone can modulate the expression of chemerin receptor genes and proteins, as well as the abundance of RARRES2 mRNA transcripts. Additionally, estradiol increased chemerin protein secretion during the maternal recognition of pregnancy and decreased its release during embryo implantation.

Galanin Receptors (GALR1, GALR2, and GALR3) Immunoexpression in Enteric Plexuses of Colorectal Cancer Patients: Correlation with the Clinico-Pathological Parameters

Summary: This study found that the expression levels of GALR1 and GALR3 proteins were elevated in colorectal cancer tissue, and higher GALR3 expression was associated with better prognosis. The expression of GALR1 and GALR3 slightly decreased in myenteric plexuses close to the cancer, but showed no correlation with tumor progression. The expression levels of GALRs in submucosal plexuses did not significantly change between distal and close to the tumor. However, elevated GALR1 expression in submucosal plexuses close to the tumor correlated with poor prognosis, higher tumor grading, and shorter overall survival.

Adaptation of the Porcine Pituitary Transcriptome, Spliceosome and Editome during Early Pregnancy

Summary: The aim of this study was to investigate the transcriptomic changes and mechanisms accompanying transcription and translation processes in the pituitary of the domestic pig. Through high-throughput sequencing of RNA, the study obtained information on gene expression changes, alternative splicing events, allele-specific expression sites, and RNA editing events. The study also confirmed the expression profiles of select phenomena and identified intracellular pathways that induce changes in transcription and translation regulation.
