GABA promotes -cell proliferation, but does not overcome impaired glucose homeostasis associated with diet-induced obesity

-Aminobutyric acid (GABA) administration has been shown to increase -cell mass, leading to a reversal of type 1 diabetes in mice. Whether GABA has any effect on cells of healthy and prediabetic/glucose-intolerant obese mice remains unknown. In the present study, we show that oral GABA administration (ad libitum) to mice indeed increased pancreatic -cell mass, which led to a modest enhancement in insulin secretion and glucose tolerance. However, GABA treatment did not further increase insulin-positive islet area in high fat diet-fed mice and was unable to prevent or reverse glucose intolerance and insulin resistance. Mechanistically, whether in vivo or in vitro, GABA treatment increased -cell proliferation. In vitro, the effect was shown to be mediated via the GABA(A) receptor. Single-cell RNA sequencing analysis revealed that GABA preferentially up-regulated pathways linked to -cell proliferation and simultaneously down-regulated those networks required for other processes, including insulin biosynthesis and metabolism. Interestingly, single-cell differential expression analysis revealed GABA treatment gave rise to a distinct subpopulation of cells with a unique transcriptional signature, including urocortin 3 (ucn3), wnt4, and hepacam2. Summary: The upregulation of CBS in cancer cells has been identified as a tumor-promoting factor, leading to the synthesis of a novel CBS-targeting prodrug YD0251 with higher efficacy in inhibiting tumor growth compared to the prototypical CBS inhibitor AOAA. The prodrug was successful in suppressing tumor growth in vivo with low toxicity, and a potential plasma marker for CBS-expressing tumors was identified for potential future theranostic applications to match CBS inhibitor therapy with high-CBS expressors.

Summary: This article introduces a versatile microfluidic device for 3D fluorescence imaging of mouse or human pancreas PCTs, positioning them within a chamber with tall channels for media exchange. The device localizes shear stress to small regions on tissue surfaces and allows for controlled media exchange at lower flowrates than conventional chambers. It enables high-resolution imaging of immunofluorescently labeled PCTs on a confocal and with a large field of view on a SPIM without compromising image quality.
