High cytoplasm HABP1 expression as a predictor of poor survival and late tumor stage in pancreatic ductal adenocarcinoma patients

Background: Hyaluronan-binding protein 1 (HABP1) overexpression has been confirmed in different malignancies and found to be strongly associated with tumor development and progression. The aim of the present study was to explore the impact of HABP1 in pancreatic ductal adenocarcinoma (PDAC) patients. Method: HABP1 expression was evaluated in 89 PDAC specimens. Results: The expression of HABP1 was significantly higher in tumor tissues than that in adjacent normal tissues. High nucleus HABP1 expression and high cytoplasm HABP1 expression were both detected in PDAC tissues. Overall survival analysis by optical density showed that the mean survival was similar between patients with low and high optical density values of HABP1 expression (P = 0.312). The similar result was also found between patients with low-moderate or high nucleus HABP1 expression (P = 0.275). However, the mean survival was significantly poorer in patients with cytoplasm HABP1 overexpression (P < 0.001). High cytoplasm HABP1 expression was strongly correlated with late tumor stages, arterial involvement, lymph node metastasis and carbohydrate antigen 19-9 levels. Conclusion: High cytoplasm HABP1 expression may prove to be a predictor of poor survival and late tumor stage in PDAC patients. HABP1 could serve as a promising biomarker to identify subsets of PDAC patients with high malignant clinical behavior. Mechanical stretch promotes hypertrophic scar formation through mechanically activated cation channel Piezo1

Summary: The study identified that Piezo1 channel is overexpressed in myofibroblasts of hypertrophic scar (HS) tissues and plays a regulatory role in fibroblasts, thus participating in HS formation. Piezo1 activity enhances various cellular biological behaviors in response to mechanical stretch in dermal fibroblasts, including proliferation, motility, and differentiation. Additionally, blocking Piezo1 with GsMTx4 peptide injection protected rats from stretch-induced HS formation, suggesting Piezo1 as a potential therapeutic target for HS.

Summary: This study reveals the overexpression of HDAC5 in hypertrophic scars and its depletion can inhibit scar formation. Knockdown of HDAC5 downregulates Smad2/3 phosphorylation in the TGF-8 signaling pathway and increases Smad7 expression. Moreover, HDAC5 interacts with MEF2A to suppress its binding to the Smad7 promoter region, resulting in repression of Smad7 promoter activity. Luciferase reporter assays and ChIP-qPCR assays confirmed this mechanism.

Patients who could undergo curative resection had better survival rates. A more accurate diagnostic workup and staging had a favorable impact on survival.

Neoadjuvant chemotherapy followed by surgery for HPV-associated tonsillar cancer: Does imaging reflect the pathological response?

Summary: This study evaluated the outcome of tonsillar cancer managed with neoadjuvant chemotherapy followed by surgery. The results showed that neoadjuvant chemotherapy reduced tumor volume and pathological adverse features, significantly decreasing the need for adjuvant therapy. A greater reduction in tumor volume predicted a complete pathologic response. There was no significant difference in survival rates between the groups.
