ADAM8 in invasive cancers: links to tumor progression, metastasis, and chemoresistance

Ectodomain shedding of extracellular and membrane proteins is of fundamental importance for cell-cell communication in neoplasias. A Disintegrin And Metalloproteinase (ADAM) proteases constitute a family of multifunctional, membrane-bound proteins with traditional sheddase functions. Their protumorigenic potential has been attributed to both, essential (ADAM10 and ADAM17) and 'dispensable' ADAM proteases (ADAM8, 9, 12, 15, and 19). Of specific interest in this review is the ADAM proteinase ADAM8 that has been identified as a significant player in aggressive malignancies including breast, pancreatic, and brain cancer. High expression levels of ADAM8 are associated with invasiveness and predict a poor patient outcome, indicating a prognostic and diagnostic potential of ADAM8. Current knowledge of substrates and interaction partners gave rise to the hypothesis that ADAM8 dysregulation affects diverse processes in tumor biology, attributable to different functional cores of the multidomain enzyme. Proteolytic degradation of extracellularmatrix (ECM) components, cleavage of cell surface proteins, and subsequent release of soluble ectodomains promote cancer progression via induction of angiogenesis and metastasis. Moreover, there is increasing evidence for significance of a non-proteolytic function of ADAM8. With the disintegrin (DIS) domain ADAM8 binds integrins such as beta 1 integrin, thereby activating integrin signaling pathways. The cytoplasmic domain is critical for that activation and involves focal adhesion kinase (FAK), extracellular regulated kinase (ERK1/2), and protein kinase B (AKT/PKB) signaling, further contributing to cancer progression and mediating chemoresistance against first-line therapies. This review highlights the remarkable effects of ADAM8 in tumor biology, concluding that pharmacological inhibition of ADAM8 represents a promising therapeutic approach not only for monotherapy, but also for combinatorial therapies.

Summary: Fn14 is cleaved by the protease gamma-secretase, leading to secretion of sFn14. Inhibition of gamma-secretase increases full-length Fn14 at the cell surface and enhances TWEAK ligand-stimulated Fn14 signaling. The release of sFn14 can be detected in tumor cells, plasma, and blood from cancer patients, suggesting it as a biomarker for monitoring gamma-secretase activity.

Summary: Different forms of BCAM protein are associated with metastasis and poor prognosis in ovarian cancer. The soluble form of BCAM inhibits tumor cell adhesion and migration, but promotes the dispersion and invasion of tumor cell spheroids. These findings provide new insights for the treatment of ovarian cancer.

Summary: This study found that the expression of calcium-dependent protease calpain-2 is increased in glioblastoma. Treatment with a calpain inhibitor increased the sensitivity of primary glioblastoma cells to TMZ. The knockdown of calpain-2 resulted in DNA damage and disrupted TP53 signaling. Therefore, calpain-2 may serve as a prognostic factor for GBM outcome.

Summary: This study suggests that patients with 'postural instability and gait difficulty' (PIGD) may experience more beneficial non-motor effects compared to patients with 'tremor-dominant' symptoms undergoing deep brain stimulation (DBS) for Parkinson's disease (PD). These differences in clinical efficacy on non-motor aspects should be taken into consideration when advising and monitoring PD patients undergoing DBS.

Identification of Dysregulated microRNAs in Glioblastoma Stem-like Cells

Summary: This study explored changes in miRNA expression profiles in patient-derived glioblastoma stem-like cells (GSCs) upon differentiation into glial fiber acid protein (GFAP)-expressing astrocytic tumor cells. The results showed that miR-17-5p and miR-425-5p were downregulated, while miR-223-3p and let-7-5p were upregulated in differentiated cells. Among them, miR-425-5p consistently showed the highest downregulation in all GSCs. Transfection of a 425-5p miRNA mimic demonstrated downregulation of GFAP protein in differentiated patient-derived GBM cells, suggesting direct regulation of miRNAs in the GSC/GBM cell transition.
