Summary: We developed a simple, portable, low-cost, colorimetric, and remotely controllable platform for reliable, high-throughput, and rapid diagnosis using loop-mediated isothermal amplification (LAMP) assays. This platform is capable of performing nucleic acid isolation and detection from sampling, and we confirmed its specificity and sensitivity through experiments.

Summary: This study investigates the synthesis of 2,3-Butanediol (2,3-BD) using Bacillus licheniformis 24 and chicory inulin as a substrate. The pH value has a significant impact on the process, with pH 6.25 resulting in the highest concentration of 2,3-BD. Genomic analysis reveals the involvement of several glycoside hydrolase enzymes in the hydrolysis of inulin. The overexpression of certain genes, such as sacC and sacA/fruA, is crucial for inulin hydrolysis.

Summary: Bacillus licheniformis is a Gram positive spore-forming bacterial species with high biotechnological potential, used for producing bioactive compounds and bioproducts in various fields. Its applications include probiotics, bioflocculation, biofuel production, and bioremediation among others, but safety concerns exist due to potential transfer of antibiotic resistance genes. Developing a standardized protocol to describe novel strains is crucial for evaluating the biotechnological potential of this species.

Summary: The study aimed to enhance the natural cellulase activity of Bacillus licheniformis and B. velezensis by cloning and heterologous expression of cel8A and cel48S genes from Acetivibrio thermocellus. The results demonstrate the potential for genetic complementation to significantly increase cellulase activity in bacilli.

Summary: Depression and anxiety, common mental disorders, can be treated by regulating the gut microbiota composition using probiotics like Bacillus licheniformis. This study found that B. licheniformis reduced depressive-like and anxiety-like behaviors in rats by changing gut microbiota composition, increasing short chain fatty acids (SCFAs) in the colon, and altering neurotransmitter levels in the brain. The gut microbiome, specifically Parabacteroides, Anaerostipes, Ruminococcus-2, and Blautia, showed significant correlation with neurotransmitters and SCFAs, suggesting their important role in reducing depressive-like behaviors.

Summary: A genetically encoded biosensor responding to malate was constructed in Bacillus licheniformis, enabling direct and precise detection of malate in microbial metabolism. The biosensor can respond to malate concentrations ranging from 5 to 15 g/L, showing a linear correlation between eGFP fluorescence and malate concentration. This engineering biosensor enriches the toolbox of synthetic biology for pathway engineering in malate production in microorganisms.

Summary: This study aims to identify the targets that affect bacitracin production in Bacillus licheniformis through RNA sequencing analysis. The research reveals that TEIIbac gene expression and oxygen supply play critical roles in bacitracin production. Overexpression of TEIIbac and increased oxygen availability through VHb expression significantly enhanced bacitracin production. The findings provide valuable insights for the industrial production of secondary metabolites under aerobic conditions.

Summary: The study found that the marine microorganism B. licheniformis can inhibit the transformation of C. irritans and has an anti-C. irritans effect. Further research showed that B. licheniformis mainly exerts its anti-pathogen mechanism through energy metabolism, transcription and translation, lysosome-based degradation, and PI3K-Akt pathways.

Summary: In this study, a flow-focusing microfluidic device was used to assemble 120 picolitre core-shell beads into a monolayer for digital PCR analysis. The DNA concentration of the sample was quantified through polymerase chain reaction (PCR) amplification and fluorescence detection. Wide-field fluorescence images were captured using a low-cost digital camera and macro lens, and analyzed using a custom Python script. The results of the digital PCR analysis were compared with those obtained through RT-qPCR.
