Thermal Stability of a Mercuric Reductase from the Red Sea Atlantis II Hot Brine Environment as Analyzed by Site-Directed Mutagenesis

The lower convective layer (LCL) of the Atlantis II brine pool of the Red Sea is a unique environment in terms of high salinity, temperature, and high concentrations of heavy metals. Mercuric reductase enzymes functional in such extreme conditions could be considered a potential tool in the environmental detoxification of mercurial poisoning and might alleviate ecological hazards in the mining industry. Here, we constructed a mercuric reductase library from Atlantis II, from which we identified genes encoding two thermostable mercuric reductase (MerA) isoforms: one is halophilic (designated ATII-LCL) while the other is not (designated ATII-LCL-NH). The ATII-LCL MerA has a short motif composed of four aspartic acids (4D414-417) and two characteristic signature boxes that played a crucial role in its thermal stability. To further understand the mechanism behind the thermostability of the two studied enzymes, we mutated the isoform ATII-LCL-NH and found that the substitution of 2 aspartic acids (2D) at positions 415 and 416 enhanced the thermal stability, while other mutations had the opposite effect. The 2D mutant showed superior thermal tolerance, as it retained 81% of its activity after 10 min of incubation at 70 degrees C. A three-dimensional structure prediction revealed newly formed salt bridges and H bonds in the 2D mutant compared to the parent molecule. To the best of our knowledge, this study is the first to rationally design a mercuric reductase with enhanced thermal stability, which we propose to have a strong potential in the bioremediation of mercurial poisoning. IMPORTANCE The Red Sea is an attractive environment for bioprospecting. There are 25 brine-filled deeps in the Red Sea. The Atlantis II brine pool is the biggest and hottest of such hydrothermal ecosystems. We generated an environmental mercuric reductase library from the lowermost layer of the Atlantis II brine pool, in which we identified two variants of the mercuric reductase enzyme (MerA). One is the previously described halophilic and thermostable ATII-LCL MerA and the other is a nonhalophilic relatively less thermostable enzyme, designated ATII-LCL-NH MerA. We used the ATII-LCL-NH enzyme as a parent molecule to locate the amino acid residues involved in the noticeably higher thermotolerance of the homolog ATII-LCL MerA. Moreover, we designed a novel enzyme with superior thermal stability. This enzyme might have strong potential in the bioremediation of mercuric toxicity.

Comparative 16S Metabarcoding of Nile Tilapia Gut Microbiota from the Northern Lakes of Egypt

Bioprospecting the microbiome of Red Sea Atlantis II brine pool for peptidases and biosynthetic genes with promising antibacterial activity

Summary: In this study, two clones with antibacterial and anticancer effects were identified through screening of DNA from marine environments. These clones showed promising antibacterial activity against challenging bacterial strains and also exhibited inhibitory effects on cancer cells. Additionally, potential proteases and biosynthetic genes were detected. Therefore, searching for new molecules in extreme marine environments is of great importance.

Gene Networks and Pathways Involved in Escherichia coli Response to Multiple Stressors

Summary: This study identified key genes and pathways involved in the tolerance of Escherichia coli to multiple stressors using a systems biology approach. The study found that energy-requiring metabolic pathways, transport, and motility are generally downregulated in response to stress, while genes related to survival, stress response, biofilm formation, and DNA repair are mainly upregulated. Additionally, 15 genes with unknown functions were found to potentially play important roles in the stress response.

Summary: This article summarizes the activities of the Bacterial Viruses Subcommittee of the International Committee on Taxonomy of Viruses from March 2021 to March 2022. It provides an overview of the new classifications proposed in 2021, approved by the Executive Committee, and ratified by vote in 2022. The taxonomy of bacterial viruses underwent significant changes, including the abolition of the paraphyletic morphological families Podoviridae, Siphoviridae, and Myoviridae, as well as the order Caudovirales, and the establishment of a binomial nomenclature system for species. Characterization and comprehensive genome analysis of novel bacteriophage, vB_Kpn_ZCKp20p, with lytic and anti-biofilm potential against clinical multidrug-resistant Klebsiella pneumoniae

Summary: The study isolated six bacteriophages from urban and medical sewage, with phage vB_Kpn_ZCKp20p having the broadest host range and showing high lytic efficiency. The phage also exhibited stability at high temperatures and wide pH range, and possessed antibiofilm activity without cytotoxicity to human skin fibroblasts. Genomic analysis suggests that vB_Kpn_ZCKp20p represents a new species but belongs to the same genus as Klebsiella phages ZCKP8 and 6691. Overall, vB_Kpn_ZCKp20p shows potential as a novel phage for treating biofilm-forming K. pneumoniae and could be a promising source for antibacterial and antibiofilm products.

Computational comparative analysis identifies potential stemness-related markers for mesenchymal stromal/stem cells

Summary: This study investigated the protective effects of pomegranate juice on BaP-induced pathologies using histochemical, microbiomic, and metabolomic approaches. Pomegranate juice partially restored tissue damage in the colon caused by BaP, while the fecal microbiome and metabolome remained resilient, except for specific changes related to pomegranate and BaP biotransformation. The analysis also revealed associations between fecal microbiome and metabolome clusters, which could be utilized for metabotype prediction and gut health biomarker discovery.
