Summary: This study constructed a single-cell transcriptome atlas of non-haematopoietic cells (NHCs), including mesenchymal stromal cells and endothelial cells, collected from human samples. The atlas revealed 30 distinct subclusters of NHCs, including previously unrecognized ones. Comparative analyses with lymphoma NHCs showed subcluster-specific changes in gene expression and interaction with malignant cells in follicular lymphoma NHCs, providing insights into stromal remodelling in lymphoma and potential clinical biomarkers. The study updates NHC taxonomy in human lymph nodes and provides a rich resource for advancing lymphoma management and therapy.

Summary: This study investigates the mechanism by which TET2-mutant immune cells enable the development of Angioimmunoblastic T-cell lymphoma (AITL). The results reveal that Tet2-deficient immune cells play a crucial role in AITL development, and aberrant GCB cells in AITL undergo clonal evolution with recurrent mutations in genes related to core histones. Cd40-Cd40lg is identified as a possible mediator of interactions between GCB and tumor cell clusters.

Summary: We report a case of AITL patient who experienced deterioration of the condition due to massive pericardial effusion one month after autologous transplantation. The patient was diagnosed with cardiac tamponade caused by AITL recurrence, identified by the presence of p.Gly17Val RHOA mutation and T-lineage cells with abnormal immunephenotype in the pericardial effusion. This case suggests that precision medicine approach by detecting the p.Gly17Val RHOA mutation is useful for AITL management.

Summary: Vanishing bile duct syndrome (VBDS) is a rare hepatic disorder with no specific treatment. We treated a patient with classic Hodgkin lymphoma (HL) and VBDS using brentuximab vedotin (BV), achieving a partial metabolic response. Our experience suggests BV could be a treatment option for classic HL with VBDS.
