Hepatocellular carcinoma (HCC) is a prevalent liver malignancy that can be developed from nonalcoholic fatty liver disease (NAFLD). Numerous pathophysiological alterations, including insulin resistance, specific cytokine release, oxidative stress, and mitochondrial damage, are involved in the transition of NAFLD to cirrhosis and HCC. MicroRNAs, as post-transcriptional modulators, play a critical role in the pathogenesis of NAFLD-related HCC by regulating lipid metabolism, glucose homeostasis, cell proliferation, apoptosis, migration, and differentiation. This review summarizes the current progress of microRNAs in the risk and prognosis of NAFLD-related HCC. Anat Rec, 302:193-200, 2019. (c) 2018 Wiley Periodicals, Inc.

Disulfidptosis: a new target for metabolic cancer therapy

Peijie Zheng, Chuntao Zhou, Yuemin Ding, Shiwei Duan

Summary: Altered metabolism in cancer can be targeted for treatment by exploiting regulated cell death (RCD) mechanisms. A recent study discovered a new metabolic-related RCD called disulfidptosis, which can be triggered by glucose transporter (GLUT) inhibitors, leading to inhibition of cancer growth. This review summarizes the underlying mechanisms of disulfidptosis and discusses potential future research directions, as well as the challenges in translating disulfidptosis research into clinical applications.

Targeting the p53 signaling pathway in cancers: Molecular mechanisms and clinical studies

Jinze Shen, Qurui Wang, Yunan Mao, Wei Gao, Shiwei Duan

Summary: Tumor suppressor p53 activates downstream genes in response to stress and regulates various biological responses. Activation and inactivation of p53 are finely regulated through post-translational modifications. Abnormal activation of p53 is closely related to cancer, and targeted therapy of the p53 signaling pathway is expected to be widely used in the future.

Nanopore sequencing technology and its applications

Peijie Zheng, Chuntao Zhou, Yuemin Ding, Bin Liu, Liuyi Lu, Feng Zhu, Shiwei Duan

Summary: Since its development in 1977, Sanger sequencing has played a crucial role in molecular biology research by decoding biological genetic codes. Nanopore sequencing, a leading third-generation sequencing technology known for its long reads, portability, and affordability, is widely used in various scientific domains, including epidemic prevention and control, disease diagnosis, and animal and plant breeding. Despite initial concerns regarding its high error rates, constant advancements in sequencing platforms and algorithm analysis have effectively addressed the issue of accuracy. During the COVID-19 pandemic, nanopore sequencing has been instrumental in detecting the SARS-CoV-2 virus genome and containing the outbreak. However, limited understanding of this technology hinders its widespread adoption and application. Nanopore sequencing has the potential to become the primary choice for COVID-19 prevention and control, as well as future epidemics, while also creating value in fields such as oncology and botany. This article aims to introduce the contributions of nanopore sequencing during the COVID-19 pandemic, promote public understanding, and facilitate its utilization in emerging outbreaks worldwide. The applications of nanopore sequencing in microbial detection, cancer genomes, and plant genomes are discussed, along with strategies to enhance its accuracy.
