False-positive elevations are commonly seen, while some patients with proven relapse do not show elevated levels. Summary: Gastrointestinal cancer is a common and deadly form of cancer, with surgical excision and chemotherapy as the main treatment methods. However, these methods often come with serious adverse reactions and drug toxicity. Ganoderma lucidum has been used medicinally and as a food source in China for a long time, and its bioactive compounds have shown potential anti-tumor activities. This review aims to compile the characteristics, anti-GI cancer mechanisms, and clinical applications of G. lucidum, providing a reference for further research on its role in the prevention and treatment of GI cancer from both traditional Chinese and western medicine perspectives.

Summary: This study aimed to characterize the genomic features of microsatellite stable (MSS) tumors with high tumor mutational burden (TMB-H) that are independent of mismatch repair deficiency (dMMR) or microsatellite instability-high (MSI-H) status. The results showed that mutations in certain genes might impair the antitumor immune response despite TMB-H, while mutations in other genes were related to TMB-H with enhanced antitumor immune response. These findings are important for tailoring treatment with immune checkpoint inhibitors.

Summary: This study revealed the molecular profiles of Chinese patients with advanced breast cancer (ABC) and highlighted the clinical importance of ctDNA-derived markers for predicting treatment response, prognosis, and disease progression. Common gene mutations included TP53 and PIK3CA, while frequent CNVs included amplifications of ERBB2 and FGFR1. Furthermore, TP53 and PIK3CA mutations were associated with survival outcomes in triple-negative breast cancer (TNBC) patients, and high ctDNA fraction and blood-based tumor mutation burden (bTMB) were associated with progression-free survival (PFS) in TNBC and HER2+ patients.

Summary: Gastrointestinal (GI) cancers are responsible for 35% of cancer-related deaths due to their ability to spread and form drug-resistant metastases, particularly in the liver and lungs. Circulating tumour cells (CTCs) are crucial in the formation of new metastases and their presence in the bloodstream is associated with a poor prognosis. Targeting CTCs could be a novel strategy in combating metastasis by understanding their biology and weaknesses. This article outlines literature on the biological features of CTCs in GI cancers and their impact on metastasis formation, along with relevant clinical studies.

Summary: Histopathological diagnosis of pulmonary tumors is crucial for making treatment decisions. This study compared the diagnostic value of multiple immunohistochemical markers in distinguishing primary lung adenocarcinoma from pulmonary metastasis from the gastrointestinal tract. The results suggest that CDH17, GPA33, and SATB2 may be used as alternative markers to CDX2 and CK20. However, no single or combination of markers can definitively distinguish between primary lung cancers and metastatic GI tract cancers.

Summary: This study investigates the effects of gastrointestinal microbiota dysbiosis on systemic inflammation during early ovarian cancer development using an animal model. The results show that CRP serum levels were significantly lower and TNF-alpha levels were significantly higher in the microbiota dysbiosis group compared to the control group. The abundance of specific microbial communities correlated with CRP levels. These findings suggest the potential importance of GI microbial profiles in identifying individuals at risk of ovarian cancer.
