Summary: An extracellular cyclodextrin glucanotransferase (CGTase) was characterized from Caldalkalibacillus mannanilyticus IB-OR17-B1 for the first time. The enzyme displayed maximum activity at pH 8 and 60°C, and showed stability in the pH range of 7-10 and temperatures of 70°C or below. The thermal stability of the enzyme improved in the presence of calcium and magnesium salts, while it was inhibited by high concentrations of Ag+, Cu2+, Zn2+, Fe2+, and Fe3+ cations. The enzyme converted potato starch into a mixture of alpha-, beta-, and gamma-cyclodextrins.

Summary: Nonpathogenic Bacillus subtilis was used as a host to express cyclodextrin glucanotransferase (CGTase), and the optimal signal peptide and promoter were identified. The highest yield of a-monoglucosyl hesperidin reached 2.70 g L-1, which is the highest level achieved using recombinant CGTase to date.

Summary: In this study, the transglucosylation of baicalin was achieved using CGTases from Thermoanaerobacter sp. and Bacillus macerans with a-cyclodextrin as a glucosyl donor. Optimized conditions resulted in 17 enzymatically modified baicalin derivatives, with the main glucosides identified as BG(1) and BG(2). The water solubility of BG(1) and BG(2) was significantly increased compared to that of baicalin, and both glucosides exhibited antioxidant and anti-glycation properties.

Summary: Antibiotic resistance is a growing problem, and finding new treatment alternatives is crucial. Bacteriocins, small antimicrobial peptides, have shown effectiveness against multidrug-resistant microbes and possess low cytotoxicity to human cells, tolerance to enzyme degradation, and stability to temperature and pH variations. Despite their diverse structures and functions, the mechanisms of action of bacteriocins are not well understood. This review focuses on bacteriocins produced by Bacillus species, describing their mechanisms of action, culture conditions for improved production, and state-of-the-art methodologies for identification. The utilization of bacteriocins as food preservatives and novel cancer treatments is also discussed.

Summary: This study isolated alkaline-tolerant and moderate-temperature pectate lyase-producing bacteria and investigated the characteristics and influencing factors of the enzyme under different conditions. The results showed that the pectate lyase from this strain has potential for industrial applications, particularly in the clarification of fruit juices.

Summary: Alkali lignin-degrading Bacillus were isolated from forest soils in China and identified as Bacillus subtilis TR-03 and Bacillus cereus TR-25. The strain TR-03 showed efficient alkali lignin degradation and decolorization capabilities, mainly through the production of lignin peroxidase and laccase. Additionally, physical and chemical changes of lignin were observed via SEM and FTIR analysis to confirm the lignin degradation by Bacillus subtilis TR-03.

Summary: This study discussed the production, partial purification, and characterization of ligninolytic enzymes from various fungi in Potato Dextrose (PD) broth. The enzymes laccase, lignin peroxidase, and manganese peroxidase were largely isolated using ammonium sulphate precipitation followed by dialysis, resulting in purified enzymes with enhanced specific activities. The highest specific activity was found in Hypsizygus ulmarius laccase, followed by lignin peroxidase and manganese peroxidase.

Summary: Novel Janus nanoparticles based on polyamidoamine dendrons modified Au colloids were prepared. The nanoparticles were functionalized with horseradish peroxidase on one side and with a ssDNA aptamer for CRP on the other side. These nanoparticles were used as biorecognition/signaling elements to construct an amperometric aptasensor for specific detection of the cardiac biomarker.

Summary: The design of a novel particle-to-particle intercommunicated nanosystem for dual delivery is described in this study. The nanosystem consists of an Au-mesoporous silica Janus nanoparticle loaded with paracetamol and mechanized with light-sensitive supramolecular gates, as well as a mesoporous silica nanoparticle loaded with rhodamine B and gated with thiol-sensitive ensembles. Upon irradiation with a near-UV light laser, the analgesic drug is released from the Janus nanomachine, while the addition of N-acetylthiocholine leads to the enzymatic production of thiocholine, causing the disruption of the gating mechanism at the second mesoporous silica nanoparticle and subsequent dye release.
