Efficient in vivo antitumor effect of an immunotoxin based on ribotoxin α-sarcin in nude mice bearing human colorectal cancer xenografts

Tagging of RNases, such as the ribotoxin alpha-sarcin, with the variable domains of antibodies directed to surface antigens that are selectively expressed on tumor cells endows cellular specificity to their cytotoxic action. A recombinant single-chain immunotoxin based on the ribotoxin alpha-sarcin (IMTXA33 alpha S), produced in the generally regarded as safe (GRAS) yeast Pichia pastoris, has been recently described as a promising candidate for the treatment of colorectal cancer cells expressing the glycoprotein A33 (GPA33) antigen, due to its high specific and effective cytotoxic effect on in vitro assays against targeted cells. Here we report the in vivo antitumor effectiveness of this immunotoxin on nude mice bearing GPA33-positive human colon cancer xenografts. Two sets of independent assays were performed, including three experimental groups: control (PBS) and treatment with two different doses of immunotoxin (50 or 100 mu g/injection) (n = 8). Intraperitoneal administration of IMTXA33 alpha S resulted in significant dose-dependent tumor growth inhibition. In addition, the remaining tumors excised from immunotoxin-treated mice showed absence of the GPA33 antigen and a clear inhibition of angiogenesis and proliferative capacity. No signs of immunotoxin-induced pathological changes were observed from specimens tissues. Overall these results show efficient and selective cytotoxic action on tumor xenografts, combined with the lack of severe side effects, suggesting that IMTXA33 alpha S is a potential therapeutic agent against colorectal cancer.

Iatrogenic Neonatal Esophageal Perforation: A European Multicentre Review on Management and Outcomes

Summary: The aim of this multicenter retrospective study and literature review was to review management and outcomes of neonatal esophageal perforation (NEP). The study found that NEP during NGT insertion is rare even in premature infants. In this small cohort, conservative management seems to be safe. A larger sample size will be necessary to answer questions on efficacy of antibiotics, antacids and NGT re-insertion time frame in NEP.

Identification of NRF2 Activation as a Prognostic Biomarker in T-Cell Acute Lymphoblastic Leukaemia

Summary: The standard-of-care treatment for T-cell acute lymphoblastic leukaemia (T-ALL) achieves reasonable response rates, but relapsed or treatment-resistant patients have poor outcomes. This study explores the potential of NRF2 activation as a prognostic biomarker in T-ALL. Through analysis of transcriptomic, genomic, and clinical data, high NFE2L2 levels were found to be associated with shorter overall survival in T-ALL patients. The PI3K-AKT-mTOR pathway plays a role in the oncogenic signaling induced by NRF2 in T-ALL. High NFE2L2 levels are also associated with drug resistance in T-ALL, possibly due to NRF2-induced glutathione biosynthesis. These findings suggest that NFE2L2 levels could be a predictive biomarker for poor treatment response in T-ALL patients, offering insights for improved patient stratification and targeted therapies.

Summary: The aim of this international study was to assess the current status of transition of care (TOC) and adult care (AC) programs for patients with anorectal malformations (ARM) and Hirschsprung disease (HD). The results showed that less than 50% of the participating centers had installed these programs, and there was considerable heterogeneity in the content and implementation of the programs. Sharing best practice examples and considering local and national healthcare programs may improve the continuity of care in the future.

A New Optimized Version of a Colorectal Cancer-Targeted Immunotoxin Based on a Non-Immunogenic Variant of the Ribotoxin alpha-Sarcin

Summary: Due to its high incidence and mortality, cancer is still a major risk to human health and lifespan. Immunotherapy and immunotoxins have emerged as promising approaches to overcome this global disease. This study designed two non-immunogenic variants of immunotoxins and demonstrated their enhanced antitumor activity and lower immunogenicity compared to the wild-type immunotoxin. These findings provide a step forward in optimizing immunotoxins for improved outcomes in cancer treatment.

Effect of Postnatal Epigallocatechin-Gallate Treatment on Cardiac Function in Mice Prenatally Exposed to Alcohol

Summary: Prenatal alcohol exposure can cause cardiovascular health issues in offspring. This study investigated the effects of the compound EGCG on cardiac dysfunction in mice prenatally exposed to alcohol. The results showed that prenatal alcohol exposure led to cardiac alterations and dysfunction, but postnatal treatment with EGCG restored physiological levels of heart biomarkers and improved cardiac function. These findings suggest that postnatal EGCG treatment can attenuate the damage caused by prenatal alcohol exposure.

Summary: This study aimed to explore the role of neutrophil-to-lymphocyte ratio (NLR) as a predictor of surgical site infection (SSI) after appendectomy in children. The results showed that NLR value at admission was the most promising predictive factor for SSI in children undergoing appendectomy. It is an easy, simple, inexpensive, and rapid method to detect patients at high risk for SSI.
