Summary: Lactic acid plays a crucial role in the transformation and oncogenic function of stromal cells in the tumor microenvironment. It not only provides energy for stromal cells but also acts as a signaling molecule to regulate their activity. Activated stromal cells, in turn, support tumor development.

Summary: Components of the tumor microenvironment, such as tumor-associated macrophages (TAMs), have a dual impact on tumor development and treatment. A novel bladder tumor model is developed to study the impact of bacterial distribution on immunomodulation within the tumor microenvironment. It is shown that biofilms induce inflammatory conditions that promote macrophage polarization towards a pro-tumor state. Additionally, extratumoral biofilms can shield tumors from immune attack by TAMs. These findings provide insights into the role of biofilm-mediated inflammation in tumor progression and suggest new therapeutic strategies.

Summary: Innovative scaffold designs that modulate the local inflammatory microenvironment through favorable macrophage polarization and suppressing oxidative stress are needed for successful clinical translation of regenerative cell therapies and graft integration. We herein report the derivation of a hydrazone-crosslinked gallol-functionalized hyaluronic acid-based hydrogel that displayed outstanding viscoelastic properties and immunomodulatory characteristics.

Summary: Lactic acid metabolism-related genes (LRGs) play a significant role in lung adenocarcinoma (LUAD), with the identification of distinct molecular subtypes and the development of a prognostic signature. Patients with low LRG_score demonstrate better prognosis, higher immune score, lower somatic mutation rate, and increased responsiveness to anti-PD-1/PD-L1 medications. The stability and reliability of LRG_score were validated in cell lines, clinical tissue samples, and HPA databases, indicating its potential to improve prognostic information and guide drug treatment strategies for LUAD patients.

Summary: Poly(lactic acid) (PLA) plastics are of great significance to the sustainable development of the world. The production of PLA involves the synthesis of lactide (LT) from lactic acid (LA) and the subsequent ring-opening polymerization. The quality and capacity of LA and LT directly affect the production and properties of PLA. This review summarizes the recent research progress on LA, LT, and PLA, and highlights the latest methods with significant advantages in energy consumption, reaction integration, efficiency, and yield in the production process.

Summary: This study demonstrates that creatine supplementation enhances anti-tumor immunity by upregulating the function of macrophages through increased ATP production. Creatine supplementation significantly inhibits tumor growth and increases tumor antigen-specific CD8+ T cells. The enhancement of macrophage function by creatine is achieved through increased ATP levels, ultimately contributing to enhanced anti-tumor immunity orchestrated by CD8+ T cells.

Summary: The study developed a tumor-in-a-dish experimental system to investigate early events promoting tumor growth and immune suppression in metastatic melanoma. Through co-culturing murine melanoma tumor cells, fibroblasts, and macrophages in a 3D collagen matrix, the researchers found that macrophages acquired features consistent with those in in vivo melanoma tumors over 7 days. This system provides a platform for studying macrophage-stromal interactions in the melanoma microenvironment and testing potential therapies to stimulate immune activity in macrophages.

Summary: An activatable immunomodulatory nanoadjuvant, CSC@Syro, is constructed to relieve the immunosuppressive tumor microenvironment and boost tumor immune response. The nanoadjuvant induces tumor cell ferroptosis and immunogenic cell death, while blocking lactate metabolism and neutralizing acidic tumor microenvironment. This synergistic therapy integrates tumor metabolic regulation and ferroptosis with immunotherapy, effectively suppressing tumor proliferation and metastasis.
