Recent studies have demonstrated that DNA demethylation agents can mimic a viral infection by induction of dsRNAs. This viral mimicry leads to an antiviral response mediated by the cytosolic pattern recognition receptor MDA5, followed by MAVS (IPS1) activation, IRF7 nuclear translocation and upregulation of type III Interferon and interferon-stimulated genes.

Summary: The study demonstrates that DNA hypomethylating agents directly modulate the anti-tumor response and effector function of CD8(+) T cells, promoting tumor infiltration and suppressing growth. This effect is achieved through the regulation of T cell activation-related transcriptional networks, ultimately enhancing the immune response against tumors.

Extracellular vesicles shed by follicular lymphoma B cells promote polarization of the bone marrow stromal cell niche

Summary: Follicular lymphoma-derived extracellular vesicles (EVs) can enhance the support of FL B-cell growth by BM-MSCs, alter gene expression, activate different signaling pathways, and potentially contribute to the formation of a permissive stromal niche in the bone marrow for FL.

Summary: The study identified three distinct human lymphoid stromal cell (LSC) subsets with different functions in healthy tissue. In lymphoma, LSCs can be reprogrammed in response to cytokines like tumor necrosis factor and transforming growth factor beta, leading to increased expression of chemokines. This bidirectional crosstalk between LSCs and malignant B cells may be a targetable axis in lymphoma.

Measuring the effect of drug treatments on primary human CD8+ T cell activation and cytolytic potential

Helen Loo Yau, Daniel D. De Carvalho

Summary: CD8+ T cells are key effector cells in the adaptive immune response, and drug treatments may impact their function. This protocol outlines robust methods for measuring the effects of drugs on the activation and cytolytic potential of primary human CD8+ T cells through ex vivo polyclonal activation and cell culture conditions.
