Focal adhesion kinase (FAK) is a cytoplasmic tyrosine kinase that regulates a plethora of downstream signaling pathways essential for cell migration, proliferation and death, processes that are exploited by cancer cells during malignant progression. These well-established tumorigenic activities, together with its high expression and activity in different cancer types, highlight FAK as an attractive target for cancer therapy. We have assessed and characterized the therapeutic potential and the biological effects of BI 853520, a novel small chemical inhibitor of FAK, in several preclinical mouse models of breast cancer. Treatment with BI 853520 elicits a significant reduction in primary tumor growth caused by an anti-proliferative activity by BI 853520. In contrast, BI 853520 exerts effects with varying degrees of robustness on the different stages of the metastatic cascade. Together, the data demonstrate that the repression of FAK activity by the specific FAK inhibitor BI 853520 offers a promising anti-proliferative approach for cancer therapy.

Summary: NUT midline carcinoma is an aggressive neoplasm with chromosomal rearrangements of the NUT gene. The article presents a strategy to detect this rare tumor through DNA methylation array analysis, using molecular data and computational strategies. This combined approach can detect the unusual cell lineage of NUT midline carcinomas for diagnostic use.

Summary: Myoepithelial neoplasms of soft tissue are rare tumors with clinical, morphological, immunohistochemical, and genetic heterogeneity. The diagnosis often requires immunostaining to confirm myoepithelial differentiation. In some cases, tumors may have typical morphology but with immunophenotype that fails to confirm myoepithelial differentiation, termed as myoepithelioma-like tumors.

Brown Tumors Belong to the Spectrum of KRAS-driven Neoplasms

Summary: Brown tumors are rare bone lesions associated with hyperparathyroidism. This study found that brown tumors may be true neoplasms driven by the activation of the RAS-MAPK signaling pathway. Regression of brown tumors after normalization of hyperparathyroidism suggests a second hit mediated by endocrine stimulation is required for tumor development. Brown tumors have a pathogenic relation to nonossifying fibroma and giant cell granuloma of the jaws.

Summary: This study analyzed the genetic and epigenetic alterations of hepatocellular adenomas (HCA) with atypical morphological features, and found differences with hepatocellular carcinoma (HCC) or HCA without atypical morphology. Therefore, targeted sequencing and/or combined methylation/copy number profiling should be performed to differentiate between HCC and HCA when diagnosis is difficult.

Cemento-osseous dysplasia is caused by RAS-MAPK activation

Summary: Cemento-osseous dysplasia (COD) is a benign fibro-osseous lesion that occurs exclusively in the tooth-bearing areas of the jaws. It can be classified into periapical, focal, and florid subtypes based on the site and extent of involvement, which share the same histomorphology. Most cases are asymptomatic and self-limited, requiring no specific treatment. The molecular pathogenesis of COD, specifically the mechanism underlying the spontaneous growth arrest in most lesions, is still unknown.

Superficial mesenchymal tumours expressing epithelial markers on immunohistochemistry: Diagnostic clues and pitfalls

Summary: The diagnosis of mesenchymal neoplasms in superficial soft tissue is challenging due to their rarity and overlapping features. The classification of these tumors has expanded to include potential new entities that were described after the publication of the 5th edition of WHO classification in 2020. Although non-mesenchymal tumors are more common in the skin and superficial soft tissue, specific entities in the mesenchymal category can express epithelial markers, highlighting the importance of being aware of diagnostic pitfalls. This article provides an overview on the differential diagnosis of these mesenchymal tumors in the skin.

Summary: This study presents a case of a 60-year-old Italian patient with Paget disease of bone and an osteoclast-rich tumor. Through analysis of clinical, morphological, and genetic data, the authors suggest that osteoclast-rich lesions in Paget disease of bone are genetically distinct from classical giant cell tumor of bone. The importance of differentiating these lesions is discussed.
