Summary: IL-3 inhibits the differentiation of Th17 cells and promotes the development of Treg cells, regulating the pathogenesis of rheumatoid arthritis. IL-3 treatment significantly decreases the pathogenic Th17 cell responses and arthritic scores.

Summary: The study found that in early rheumatoid arthritis patients, intermediate monocytes did not correlate with bone characteristics, but showed a positive correlation with circulating CXCR3(+) Th17 cells. Future longitudinal studies in patients with longer disease duration are needed to further explore the potential role of intermediate monocytes in driving bone loss in RA.

Summary: The current study evaluated the anti-arthritic activity of n-hexane and aqueous extracts of Cassia absus seeds. The extracts showed significant inhibition of paw edema and reduction in paw size and joint diameter. The extracts also improved the levels of inflammatory and oxidative biomarkers. Polymerase chain reaction investigation revealed the modulation of various inflammatory factors by the extracts. In conclusion, Cassia absus can effectively alleviate arthritis symptoms by modulating oxidative and inflammatory biomarkers.

Summary: IL-6-triggered Th17 cell expansion plays a crucial role in immune diseases like RA. The study reveals that the differentiation of Th17 cells induced by IL-6 involves not only JAKSTAT3-RORγt signaling but also GRK2-A(3)AR-cAMP-PKA-CREB/ICER-RORγt pathway, highlighting the significance of GRK2-controlled cAMP signaling in Th17 cell differentiation and its potential therapeutic application in Th17-driven immune diseases.

Summary: The study found that under cytokine conditions mimicking autoimmune inflammation in rheumatoid arthritis, the polarization of peripheral naive and non-naive T cells into Th9 or Th17 cells was similar. This suggests that the differentiation of Th9 cells may also be induced by Th17-driving conditions.

Summary: The study introduces a novel IL-6 trans-signaling trap called cs-130Fc, which exhibits improved inhibition of and increased selectivity for IL-6 trans-signaling compared to the conventional sgp130Fc fusion protein. These variants may provide support for the development of more precisely targeted therapeutics for inflammatory disorders associated with IL-6 trans-signaling.

Summary: The host defence peptide cathelicidin is a potent Th17 potentiator, enhancing aryl hydrocarbon receptor and ROR gamma t expression in a TGF-beta 1-dependent manner, directing T cells towards Th17 phenotype while protecting them from apoptosis. Neutrophils secrete cathelicidin, which is required for maximum Th17 differentiation and plays a role in early neutrophilia directing subsequent adaptive immune responses.

Summary: The study revealed that patients with PsA have an increased Th17:Treg ratio, which can be reversed by anti-TNF therapy. Treg from patients treated with different therapies showed contrasting effects on regulating effector T-cell migration, with TNF blockade boosting Treg suppression of migration.

Summary: Lameness in dairy cattle can be caused by infectious or noninfectious agents, with joint lesions being the second most common cause. Fibroblast-like synoviocytes (FLS) play a key role in joint diseases and the expression of proinflammatory mediators. Tumor necrosis factor-alpha (TNF-alpha) is a potent proinflammatory cytokine that increases the expression of proinflammatory cytokines in bovine FLS (bFLS). This study found that TNF-alpha induces metabolic reprogramming in bFLS, favoring glycolysis, and increasing the expression of IL-6, IL-8, IL-1 beta, and COX-2/PGE2.
