Summary: Glioblastoma multiforme (GBM) is a highly aggressive form of brain cancer that is difficult to treat due to its resistance to traditional cancer treatments. The unique biology of GBM cells and the blood-brain barrier contribute to this resistance. However, there are several treatment options available, including surgery, chemoradiotherapy, adjuvant chemotherapy, immunotherapy, targeted therapies, and tumor-treating fields. Ongoing clinical trials aim to improve outcomes for GBM patients.

Summary: This study investigates the role of the gene STAC1 in glioblastoma cell invasion and survival. It is found that STAC1 overexpression promotes invasion and inhibits apoptosis, while knockdown of STAC1 has the opposite effect. The study also reveals that STAC1 regulates AKT and calcium channel signaling in glioblastoma cells. These findings provide valuable insights into the pathogenic roles of STAC1 in high-grade glioblastoma and highlight its potential as a promising therapeutic target.

Summary: By studying the GBM BVZ subtypes, we discovered the differential expression of related miRNAs and mRNAs, clinical characteristics, and functional pathways. Based on the expression of miR-21 and miR-10b, approximately 30% of GBM patients were classified as having the GBM BVZ-responsive subtype, which was associated with significantly shorter survival time. SVM classifiers and miRNA panels were used to classify and detect GBM BVZ subtypes.

Summary: In this study, two deep learning models were used to predict the transcriptional subtypes and prognosis of glioblastoma (GBM) cells from histology images. The results showed consistent associations between spatial cellular organization and patient prognosis. The study also confirmed that transcriptional heterogeneity and cell-state plasticity are key factors in the development of therapeutic resistance in GBM.

Summary: The study revealed differential metabolic responses to TMZ in resistant neurospheroidal and susceptible glioblastoma cell lines, with identified mutations in signaling pathway regulators of growth and energy metabolism. Using an integrated approach, rewiring in glycolysis, TCA cycle, malate aspartate shunt, and oxidative phosphorylation pathways were identified as key factors in shaping TMZ resistance. Differential killing of TMZ resistant NSP by Rotenone at low concentrations, with an IC50 value three orders of magnitude lower than in U87MG, highlights the effectiveness of the integrated systems-based approach.

Summary: In this study, convection-enhanced delivery (CED) of 23 +/- 3 nm gold nanoparticles (AuNPs) labeled with the beta-particle-emitting radionuclide 177Lu (177Lu-AuNPs) was found to be highly effective for treating orthotopic U251-Luc human glioblastoma multiforme (GBM) tumors in NRG mice. The cytotoxicity of 177Lu-AuNPs on U251-Luc cells was confirmed through in vitro experiments, while in vivo studies showed that the tumors treated with 177Lu-AuNPs exhibited growth arrest and improved long-term survival compared to control mice. Overall, this study highlights the potential of CED using 177Lu-AuNPs as a promising treatment for GBM patients. Rating: 8/10
