Technological and probiotic potential of BGRA43 a natural isolate of Lactobacillus helveticus

Lactobacillus helveticus BGRA43 is a human intestinal isolate showing antimicrobial activity, amongst others, against Yersinia enterocolitica, Shigella sonnei, Shigella flexneri, and Streptococcus pneumoniae. BGRA43 produces PrtH proteinase with proteolytic activity on both casein and beta-lactoglobulin (BLG). BGRA43 is able to reduce the allergenicity of BLG. Bioactive peptides released in BGRA43 fermented milk are potent modulators of innate immunity by modulating the production of proinflammatory cytokines IL-6 and TNF-alpha. BGRA43 is able to survive in simulated gastric and intestinal conditions. The growth of BGRA43 in milk results in a fast acidification lowering the milk pH to 4.53 generating mild, homogeneous, and viscous yogurt-like product. The strain BGRA43 grows suitably in pure cow or goat's milk as well as in milk containing inulin or nutrim even when they are used as the sole carbon source. It is suggested that strain BGRA43 could be used as a single-strain culture for the preparation of yogurt-like products from bovine or caprine milk. Overall, L. helveticus BGRA43 could be considered as a potential probiotic candidate with appropriate technological properties attractive for the dairy industry.

Novel RclSAR three-component system regulates expression of the intI1 gene in the stationary growth phase

Summary: The rapid and appropriate response of Pseudomonas spp. to environmental fluctuations is facilitated by various signal transduction regulatory systems. In this study, a novel activator of the intI1 transcription, sensor kinase RclS, was discovered in Pseudomonas putida WCS358. The regulatory mechanism is limited to the stationary growth phase and appears to be indirect, possibly through the regulation of rpoS expression. The RclSAR regulatory system seems to be part of a complex intI1 regulatory network that includes major stress response regulons.

Redesigned pMAL expression vector for easy and fast purification of active native antimicrobial peptides

By introducing a His(6) tag and an enterokinase cleavage site, the purification strategy was optimized, and successfully applied in the purification of active native small peptides.

In vitro colistin susceptibility of pandrug-resistant Ac. baumannii is restored in the presence of selenium nanoparticles

Summary: This study aimed to investigate the synergistic activity of colistin and selenium nanoparticles against pandrug-resistant Ac. baumannii. The results showed an exceptionally strong synergistic activity between colistin and SeNPs. This combination could be a new option to fight pandrug-resistant Ac. baumannii.

Comparative genomics of trimethoprim-sulfamethoxazole-resistant Achromobacter xylosoxidans clinical isolates from Serbia reveals shortened variant of class 1 integron integrase gene

Summary: This study found that 33 out of 98 Achromobacter spp. isolates from pediatric patients were resistant to SXT, and among these, 10 resistant isolates were found to carry the intI1 gene. Genome sequencing revealed a deletion of 86 amino acids in the IntI1S protein compared to canonical IntI1 protein. The results suggest that IntI1S isolates are derived from clinical settings and that cystic fibrosis patients may serve as potential reservoirs for healthcare-associated infections in non-cystic fibrosis patients.

Influence of growth conditions on an antioxidative system in two bell pepper genotypes differing in susceptibility to phytopathogen bacteria Xanthomonas euvesicatoria

Summary: With changing environmental conditions, food production is becoming increasingly challenging. One solution to this problem is the production of cultivars resistant to various abiotic and/or biotic stresses. In this study, the response of antioxidant system in two different bell pepper genotypes to Xanthomonas euvesicatoria was analyzed. The susceptible genotype showed a higher activity of antioxidant enzymes and concentration of ascorbic acid, while the tolerant genotype adapted through regulation of chlorophyll and carotenoid concentration. Obtaining a tolerant genotype through crossbreeding can provide tolerance to both biotic and abiotic stressors.

Summary: This study conducted a comprehensive analysis of microbial diversity in the Vranjska Banja hot spring using culture-independent and culture-dependent methods. Novel taxa were discovered, and their genomic characteristics were analyzed. These microbes have the potential to produce thermostable enzymes and antimicrobial molecules, which can be of great importance for various industrial, agricultural, and biotechnological applications.

Isolation, Characterization, Genome Analysis and Host Resistance Development of Two Novel Lastavirus Phages Active against Pandrug-Resistant Klebsiella pneumoniae

Summary: Klebsiella pneumoniae is a global health threat and phages LASTA and SJM3 show potential in combating pandrug-resistant K. pneumoniae infections. The two phages have narrow host ranges and long latent periods, but are not lysogenic. Genome analysis reveals they belong to the new genus Lastavirus. In tests, both phages demonstrate significant bacterial reduction, with SJM3 performing slightly better.

Virulence potential of multidrug-resistant Acinetobacter baumannii isolates from COVID-19 patients on mechanical ventilation: The first report from Serbia

Summary: Since the declaration of the COVID-19 pandemic, the disease has overwhelmed the health system, leading to an increase in ICU admissions. This study examines the antibiotic resistance and virulence potential of A. baumannii isolates recovered from COVID-19 patients requiring mechanical ventilation. The results reveal the presence of multidrug-resistant A. baumannii strains with varying levels of antibiotic resistance and virulence potential.

Culture-Dependent and Metabarcoding Characterization of the Sugar Beet (Beta vulgaris L.) Microbiome for High-Yield Isolation of Bacteria with Plant Growth-Promoting Traits

Summary: This study aimed to investigate whether 16S rRNA gene metabarcoding has predictive power in identifying the majority of known bacteria with plant-beneficial traits in the sugar beet microbiome. Through isolation and testing of bacterial isolates, it was found that culture-dependent analysis is necessary for identifying bacteria with beneficial traits, and plant-based media is effective for high-yield isolation of such bacteria.

Highly stable and versatile a-amylase from Anoxybacillus vranjensis ST4 suitable for various applications

Summary: Amylase from thermophilic bacterial strain Anoxybacillus vranjensis ST4 was cloned, expressed, and purified successfully. The enzyme showed maximum activity at 60-80°C and was active and stable over a wide pH range (4.0-9.0). It efficiently hydrolyzed starch granules, releasing maltose, maltotriose, and maltopentaose as the main products. These findings indicate that this highly stable and versatile enzyme has great potential for industrial applications.

Improvement of nutritional and bioactive properties of barley b-glucan-based food products using Bacillus subtilis 168 endo-beta-1,3-1,4-glucanase

Summary: The study focused on the impact of the enzyme endo-beta-1,3-1,4-glucanase from Bacillus subtilis 168 on improving the nutritional and bioactive properties of barley beta-glucan. A new procedure for isolating pure beta-glucan from barley flour was developed, enabling it to be free from starch contamination. The cloned enzyme showed high efficiency in degrading beta-glucan and increasing its antioxidant capacities, suggesting potential applications in functional foods.

Short-term effects of Brevibacillus laterosporus supplemented diet on worker honey bee microbiome: a pilot study

Summary: In this study, honey bees were fed with a diet supplemented with spores of Brevibacillus laterosporus BGSP11. The short-term effects on their micro- and mycobiota were analyzed using Illumina MiSeq sequencing. The results show that this treatment does not harm the bacterial microbiome of the bees, but slightly affects the composition of core microbiota. The treatment leads to an increase in beneficial bacteria species and a reduction in potential pathogens. However, it has a more profound effect on the mycobiota, reducing phytopathogenic fungi that can affect the quality of honey.

Rhizosphere microbiomes of resurrection plants Ramonda serbica and R. nathaliae: comparative analysis and search for bacteria mitigating drought stress in wheat (Triticum aestivum L.)

Summary: Rhizosphere microbial communities play a key role in plant health and productivity. The rhizobacteria Pseudomonas putida P2 and Bacillus cereus P5 were effective in promoting plant growth and alleviating oxidative stress in the Belija wheat cultivar under drought conditions. Next Generation Sequencing revealed extremophilic bacteria such as Vicinamibacter silvestris, Chthoniobacter flavus, and Gaiella occulta in the rhizosphere of resurrection plants R. serbica and R. nathaliae. Penicillium was the most abundant fungi, while Fusarium and Mucor were specific to R. serbica and Metarhizium and Tolypocladiumu were specific to R. nathaliae. These findings highlight the importance of in vivo experiments to confirm the properties of plant growth-promoting bacteria and the potential of resurrection plant rhizosphere as a valuable source for improving crop drought stress tolerance.

Exploring the antibacterial potential of Lactococcus lactis subsp. lactis bv. diacetylactis BGBU1-4 by genome mining, bacteriocin gene overexpression, and chemical protein synthesis of lactolisterin BU variants

Summary: The lactic acid bacterium Lactococcus lactis BGBU1-4 produces a bacteriocin called lactolisterin BU (LBU) which has potent antibacterial activity against Gram-positive pathogens. Additionally, it produces an unknown antibacterial product. It was discovered that LBU breaks down to form a truncated version consisting of 31 amino acids, called LBU1-31, which has 12.5% the antibacterial activity of the full-length LBU. The study also revealed that the N-terminal 24 amino acids of LBU are responsible for antibacterial activity.

Genotype-Dependent Antioxidative Response of Four Sweet Pepper Cultivars to Water Deficiency as Affected by Drought-Tolerant Bacillus safensis SS-2.7 and Bacillus thuringiensis SS-29.2 Strains

Summary: In this study, the effects of two drought-tolerant Bacillus strains, SS-2.7 and SS-29.2, on four sweet pepper genotypes (133, 274, California Wonder-CalW, and Matica) under water deficiency conditions were examined. The results showed that antioxidant enzyme activity under drought conditions was dependent on genotype and bacterial treatment. It was also observed that different genotypes exhibited varying responses to bacterial treatment under water deficiency. The study highlights the growth-promoting effect of drought-tolerant Bacillus strains on selected pepper genotypes and the genotype-dependent response to biostimulant application, especially under stress conditions.
