Summary: In this study, the inhibitory signal of natural killer (NK) cells in hepatocytes and the toxic effect of triptolide (TP) on this signal were investigated. It was found that TP impairs the inhibitory signal of NK cells and activates them, leading to hepatocellular toxicity. Furthermore, TP significantly inhibits MHC class I molecules in murine hepatocytes, but this effect can be attenuated by IFN-alpha, which protects the liver against TP and LPS-induced injury.

Summary: Levels of soluble HLA molecules (sHLA) are significantly increased in COVID-19 patients and convalescent individuals, positively correlating with SARS-CoV-2-directed cellular immunity. Severe COVID-19 cases show reduced sHLA levels. The soluble immunopeptidome in COVID-19 patients exhibits increased diversity of HLA-presented peptides, and a SARS-CoV-2-derived peptide from the viral nucleoprotein is identified. Levels of sHLA directly correlate with the diversity of the soluble immunopeptidome.

Summary: This study investigates the role of pre-cDC1s in alloreactivity and antitumor responses. It reveals that pre-cDC1s have different expression levels of PD-L1 and PIR-B compared to CD8 alpha+ cDC1s, and they play a distinct role in T-cell proliferation and PD-1 expression. The study also suggests the potential clinical relevance of pre-cDC1s in suppressing graft-versus-host disease (GvHD) while promoting graft-versus-leukemia (GvL) responses.

Summary: RhCMV-based vaccination against SIV in rhesus macaques can effectively control SIV infection in approximately 55% of vaccinated monkeys by eliciting MHC-E-restricted CD8+ T cells. Analysis of the Mamu-E genomic sequences in rhesus macaques reveals gene duplication and different allele groups that are associated with the lack of vaccine protection.

Summary: Using two-photon imaging, we demonstrate that pneumococci and microspheres can translocate from the nasal cavity to the meninges of mice within minutes of inhaled delivery. This suggests the existence of an inward flow of fluid connecting the nasopharynx to the meninges. Our findings provide new insights into the mechanisms of pathogens invading the central nervous system and have implications for drug delivery and air pollution-related brain damage.
