Pyroptosis, a type of inflammatory process caused by Gasdermins (GSDMs) proteins, has been found to be enhanced in NDs, along with the aggregation of pathological proteins. This review aims to provide an overview of the molecular mechanisms driving pyroptosis, the links between pyroptosis and NDs, and emerging therapeutic strategies in Traditional Chinese Medicine (TCM) for inhibiting pyroptosis in the treatment of NDs.

This article introduces several well-studied analgesic alkaloids derived from TCM and their mechanisms and clinical applications, using the concept of drug-cloud (dCloud) theory. Drug combinations and the combination of herbal drugs with specific properties in TCM are explored for better therapeutic effects.
